Surface functionalized AuNCs optical biosensor as an emerging food safety indicator: Fundamental mechanism to future prospects

Comparative Transcriptome Analysis of Head Kidney of Aeromonas hydrophila-infected Hypoxia-tolerant and Normal Large Yellow Croaker

In this work, we constructed a machine learning-assisted dual-channel visual sensor array for identifying the origin of Lilium bulbs (BH). Nanogold clusters (AuNCs) and quantum dots (QDs) were selected and combined into sensor arrays. The amino acids existed in lilium bulbs could induce aggregation-induced fluorescence enhancement effect (AIEE) of AuNCs through hydrogen bonding. The hydrogen protons released from the amino acids and phenolic acids could interact with the COO- group of QDs, resulting in aggregation-caused quenching (ACQ) of QDs. Due to the different contents of amino acids and phenolic acids in BH from different origins, the sensor array can produce distinct and different fluorescent colors, such as red, blue, green and purple. In conjunction with pattern recognition by the RF model, the sensor array clearly identifies the origin of BH with 94.4% prediction accuracy. The visual sensor array constructed in this work exhibited the advantages of simplicity, speed, accuracy and portability, showing potential application prospects in identifying the origin of food and traditional Chinese medicine. 

Machine learning-assisted visual sensor array for identifying the origin of Lilium bulbs

In this work, a microfluidic immunosensor chip was developed by incorporating microfluidic technology with electrochemiluminescence (ECL) for sensitive detection of human epidermal growth factor receptor-2 (HER2). The immunosensor chip can achieve robust reproducibility in mass production by integrating multiple detection units in a series. Notably, nanoscale materials can be better adapted to microfluidic systems, greatly enhancing the accuracy of the immunosensor chip. Ag@Au NCs closed by glutathione (GSH) were introduced in the ECL microfluidic immunosensor system with excellent and stable ECL performance. The synthesized CeO2-Au was applied as a coreaction promoter in the ECL signal amplification system, which made the result of HER2 detection more reliable. In addition, the designed microfluidic immunosensor chip integrated the biosensing system into a microchip, realizing rapid and accurate detection of HER2 by its high throughput and low usage. The developed short peptide ligand NARKFKG (NRK) achieved an effective connection between the antibody and nanocarrier for improving the detection efficiency of the sensor. The immunosensor chip had better storage stability and sensitivity than traditional detection methods, with a wide detection range from 10 fg·mL-1 to 100 ng·mL-1 and a low detection limit (LOD) of 3.29 fg·mL-1. In general, a microfluidic immunosensor platform was successfully constructed, providing a new idea for breast cancer (BC) clinical detection.

Microfluidic Immunosensor Platform for Sensitive Detection of Human Epidermal Growth Factor Receptor-2 Based on Enhanced Cathode Electrochemiluminescence of Bimetallic Nanoclusters.

Introduction This research aimed to assess the impact of Fermented Chinese herbs (FCHM) on the intestinal barrier, immunity, and protection against Aeromonas hydrophila (A. hydrophila) infection in largemouth bass (Micropterus salmoides). Methods Four experimental diets were formulated, including H0 (basal diet), H1, H2, and H3, which contained 1%, 3%, and 5% FCHM added to the basal diet, respectively. The fish were randomly allocated to four treatment groups, each with 3 parallel per treatment, consisting of 20 fish per replicate and were raised for 56 days. Results and discussion The experiment revealed that: compared with the control group, adding 1% FCHM significantly improved the weight gain rate (WGR) and specific growth rate (SGR) of the juvenile largemouth bass (P< 0.05). The ingestion of FCHM substantially elevated the activities of CAT, SOD, GSH-PX, APK, ACP, and LZM, and T-AOC level in the gut region of largemouth bass, while decreasing the MDA content in intestine (P< 0.05). Supplementation with FCHM enhanced the intestinal villus height and relative mRNA expression of intestinal barrier genes ZO-1, Claudin, and Occludin in juvenile largemouth bass. After injecting A.hydrophila, all groups of largemouth bass experienced mortality, but the consumption of FCHM resulted in a decrease in cumulative mortality. After infected with A.hydrophila, the antioxidant enzymes and immune enzymes activities of all test groups were enhanced compared to those before infection, and the antioxidant enzymes and immune enzymes activities of all groups were considerably higher than the control after feeding FCHM (P< 0.05). After infected with A.hydrophila, the intestinal MDA content of largemouth bass was higher compared with that before infection in all cases (P< 0.05), but after feeding FCHM, the MDA content was lower than the control (P< 0.05). Upon consuming FCHM, the mRNA relative expressions of pro-inflammatory biomarkers IL-1β, TNF-α, IL-15 and IL-8 in largemouth bass infected with A.hydrophila were decreased in comparison to the control group. In contrast, the mRNA expression of the anti-inflammatory biomarkers TGF-β and IL-10 were significantly elevated (P< 0.05). In summary, FCHM could improve the intestinal morphology, immunity, and antioxidant capacity of juvenile largemouth bass, and enhance it against A.hydrophila, with a better effect at 1% addition.

Amphioxus, one of the most closely related invertebrates to vertebrates, is an important animal model for studying the origin and evolution of vertebrate immunity, especially the transition from innate immunity to adaptive immunity. The current research progresses of amphioxus in the field of immune organs, immune cells, complement system, cytokines, nuclear factor kappa B, immune-related lectins and enzymes are summarized, and some issues that remain to be understood or are in need of further clarification are highlighted. We hope to provide references for more in-depth study of the amphioxus immune system and lay a solid foundation for the construction of three-dimensional immune network in amphioxus from ontogeny to phylogeny.
