A Phase I Study of PF-04449913, an Oral Hedgehog Inhibitor, in Patients with Advanced Solid Tumors
Andrew J. Wagner,Wells A. Messersmith,Naveed Shaik,Sherry Li,Xianxian Zheng,Karen McLachlan,Rossano Cesari,Rachel Courtney,Wendy J. Levin,Anthony B. El-Khoueiry +9 more
TL;DR: The results obtained in this study support further evaluation of PF-04449913 in patients with advanced solid tumors and indicate a generally dose-proportional kinetics with biphasic elimination, supporting once-daily dosing.

Phase I study of PF-03446962, a fully human mAb against ALK 1, a TGF{beta} receptor involved in tumor angiogenesis.
Laura W. Goff,F. de Braud,Roger B. Cohen,Jordan Berlin,Cristina Noberasco,Hossein Borghaei,Erjian Wang,D. Hu Lowe,Wendy J. Levin,Corrado Gallo-Stampino +9 more
TL;DR: PF-03446962 is a first in class mAb anti ALK-1, safe and with manageable toxicities, mainly characterized by asymptomatic elevation of pancreatic enzymes and transient platelet decreases.

Human Blast Crisis Leukemia Stem Cell Inhibition with a Novel Smoothened Antagonist.
Annelie Schairer,Alice Shih,Ifat Geron,Tannishtha Reya,Wendy J. Levin,Todd Van Arsdale,Catriona Jamieson +6 more
TL;DR: The role of the Shh signaling pathway in a humanized mouse model of blast crisis (BC) CML as well as the effects of a novel SMO antagonist on leukemia stem cell (LSC) quiescence, survival and self-renewal both in vitro and in vivo are investigated.

Cycling Toward Leukemia Stem Cell Elimination Wtih a Selective Sonic Hedgehog Antagonist
Alice Y. Shih,Annelie Schairer,Christian L. Barrett,Ifat Geron,Angela Court Recart,Daniel Goff,Sacha L. Prashad,Jerry Wu,Qingfei Jiang,Jason Gotlib,Larisa Balaian,Mark D. Minden,Heather Leu,Russell Wall,Wenxue Ma,Kamran Shazand,John Douglas Mcpherson,Steven M. Kornblau,Ida Deichaite,Minya Pu,Lei Bao,Giovanni Martinelli,Tannishtha Reya,Sheldon R. Morris,Todd VanArsdale,Thomas J. Hudson,Karen Messer,Hanna K. A. Mikkola,Wendy J. Levin,Kelly A. Frazer,Anil Sadarangani,Catriona Jamieson +31 more
TL;DR: It is suggested that implementation of novel LSC splice isoform detection platforms to assess efficacy of Shh inhibitor-mediated sensitization to molecularly targeted therapy may inform dormant cancer stem cell elimination strategies that ultimately avert relapse.

PF-04449913 Reverts Multi Drug Resistance (MDR) by a Strong Down-Regulation of ABCA2 and BCL2 on Leukemia Stem Cells in Phase I Acute Myeloid Leukemia and Chronic Myeloid Leukemia Treated Patients
Cristina Papayannidis,Viviana Guadagnuolo,Ilaria Iacobucci,Sandra Durante,Carolina Terragna,Emanuela Ottaviani,Maria Chiara Abbenante,Federica Cattina,Simona Soverini,Barbara Lama,Lucia Toni,Wendy J. Levin,Rachel Courtney,Carmen Baldazzi,Antonio Curti,Michele Baccarani,Catriona Jamieson,Jorge E. Cortes,Vivian G. Oehler,Karen McLachlan,Todd Van Arsdale,Giovanni Martinelli +21 more
TL;DR: It is observed that 1197 genes were differentially expressed between CD34+ cells collected before and after 28 days of PF-04449913 dose finding oral therapy, and a down regulation of Bcl2 (fold change −1.03004; p value= 0.01); ABCB1 and ABCG2 are respectively up and down regulated, with a not statistically significant p-value.
