The method comprises the following steps: collecting water quality data in a reservoir thermal stratification period, analyzing change characteristics of a dissolved oxygen stratification structure in the thermal stratification period, extracting control factors of the change characteristics of dissolved oxygen in the reservoir thermal stratification period, establishing a reservoir three-dimensional hydrodynamic water quality model, and checking the model by adopting monitoring data; and simulatingthermal stratification oxygen deficit time under different water levels, scheduling operation modes and pumped storage scheduling working conditions by utilizing a control equation, and establishinga reservoir scheduling scheme according to the distribution of the oxygen deficit time. According to the scheme, influence factors of the dissolved oxygen in the reservoir can be truly simulated, a scheduling scheme for optimizing the dissolved oxygen can be provided more accurately, a countermeasure for improving the dissolved oxygen in the stagnant temperature layer of the reservoir is provided,effective scientific guidance is provided for water environment protection and restoration of the thermal stratification reservoir, and a good prevention and control effect is achieved for oxygen deficit of the stagnant temperature layer of the reservoir.
