Despite the steady increase in the number of studies focusing on the development of
tissue engineered constructs, solutions delivered to the clinic are still limited. Specifically,
the lack of mature and functional vasculature greatly limits the size and complexity of
vascular scaffold models. If tissue engineering aims to replace large portions of tissue
with the intention of repairing significant defects, a more thorough understanding of the
mechanisms and players regulating the angiogenic process is required in the field. This
review will present the current material and technological advancements addressing the
imperfect formation of mature blood vessels within tissue engineered structures.

Angiogenesis in Tissue Engineering: As Nature Intended?

Polymer blends and polymer composites for cardiovascular implants

In this study, we present the photosynthetic oxygen (O2) supply to mammalian cells within a volumetric extracellular matrix-like construct, whereby a three-dimensional (3D)-bioprinted fugitive pattern encapsulating unicellular green algae, Chlamydomonas reinhardtii (C. reinhardtii), served as a natural photosynthetic O2-generator. The presence of bioprinted C. reinhardtii enhanced the viability and functionality of mammalian cells while reducing the hypoxic conditions within the tissues. We were able to subsequently endothelialize the hollow perfusable microchannels formed after enzymatic removal of the bioprinted C. reinhardtii-laden patterns from the matrices following the initial oxygenation period, to obtain biologically relevant vascularized mammalian tissue constructs. The feasibility of co-culture of C. reinhardtii with human cells, the printability and the enzymatic degradability of the fugitive bioink, as well as the exploration of C. reinhardtii as a natural, eco-friendly, cost-effective, and sustainable source of O2 would likely promote the development of engineered tissues, tissue models, and food for various applications.

Symbiotic Photosynthetic Oxygenation within 3D-Bioprinted Vascularized Tissues

There is a great need for engineered vascular grafts among patients with cardiovascular diseases who are in need of bypass therapy and lack autologous healthy blood vessels. In addition, because of the severe worldwide shortage of organ donors, there is an increasing need for engineered vascularized tissue constructs as an alternative to organ transplants. Additive manufacturing (AM) offers great advantages and flexibility of fabrication of cell-laden, multimaterial, and anatomically shaped vascular grafts and vascularized tissue constructs. Various inkjet-, extrusion-, and photocrosslinking-based AM techniques have been applied to the fabrication of both self-standing vascular grafts and porous, vascularized tissue constructs. This review discusses the state-of-the-art research on the use of AM for vascular applications and the key criteria for biomaterials in the AM of both acellular and cellular constructs. We envision that new smart printing materials that can adapt to their environment and encourage rapid endothelialization and remodeling will be the key factor in the future for the successful AM of personalized and dynamic vascular tissue applications.

Additive Manufacturing of Vascular Grafts and Vascularized Tissue Constructs.

End-stage hepatic failure is a potentially life-threatening condition for which orthotopic liver transplantation (OLT) is the only effective treatment. However, a shortage of available donor organs for transplantation each year results in the death of many patients waiting for liver transplantation. Cell-based therapies and hepatic tissue engineering have been considered as alternatives to liver transplantation. However, primary hepatocyte transplantation has rarely produced therapeutic effects because mature hepatocytes cannot be effectively expanded in vitro, and the availability of hepatocytes is often limited by shortages of donor organs. Decellularization is an attractive technique for scaffold preparation in stem cell-based liver engineering, as the resulting material can potentially retain the liver architecture, native vessel network and specific extracellular matrix (ECM). Thus, the reconstruction of functional and practical liver tissue using decellularized scaffolds becomes possible. This review focuses on the current understanding of liver tissue engineering, whole-organ liver decellularization techniques, cell sources for recellularization and potential clinical applications and challenges.

Stem cells with decellularized liver scaffolds in liver regeneration and their potential clinical applications

A major challenge facing the development of tissue-engineered vascular grafts (TEVGs), promising living replacements for diseased vascular structures, is enhancing angiogenesis. To promote rapid vascularization, endothelial cells (ECs) were co-cultured with smooth muscle cells (SMCs) in decellularized small intestinal submucosa scaffolds to regenerate angiogenic-TEVGs (A-TEVGs). Observation of the A-TEVGs at 1 month post-implantation revealed that a rich network of neocapillaries lining the blood vessel wall had developed; that the ECs of the neovasculatures had been derived from previously seeded ECs and later invading ECs of the host's vascular bed; that tissue vascularization had not significantly impaired mechanical properties; and that the maximal tensile strength of the A-TEVGs was of the same order of magnitude as that of native porcine femoral arteries. These results indicate that of the co-culturing of ECs with SMCs could enhance vascularization of TEVGs in vivo, possibly increasing graft perfusion and host integration.
