The method comprises the steps that after beta-glucan is hydrolyzed to oligosaccharide by lysoglycan, and then is hydrolyzed to glucose by beta-glucosidase; after a color developing solution is added for heating in a water bath, the value delta A of glucose at 510 nm is measured by a microplate reader; and the purity of beta-glucan is calculated from the glucose concentration. The method provided by the invention has the advantages of simple operation, less reagent usage, simple used instrument, short extraction time and high production and application value, and is suitable for the rapidly determining the purity of the barley grain beta-glucan extract in laboratories, quality inspection centers, functional food processing plants and the like.

If a fragment with a size of about 830bp is obtained by amplification, the material contains the A genome; if a fragment with a size of about 730bp is obtained by amplification, the material contains the C genome; and two bands with sizes of about 830p and 730bp respectively are obtained by amplification, the material contains the A genome and the C genome. The method provided by the invention can rapidly and accurately identify whether the oat material contains the A genome and the C genome. A detection kit constructed according to the method has the advantages of simple operation, good specificity and high sensitivity, thus being suitable for promotion and use in the grass roots.
