In this work, a novel fluorescent probe CDA, which was synthesized via a facile derivatization from natural and renewable camphor, exhibited aggregation-induced emission (AIE) characteristics in mixed aqueous media. The fluorescent aggregates showed an obviously enhanced fluorescence response toward N2H4. The response time of CDA aggregates for detecting N2H4 was within 20 s, and the detection limit was 2.26 ppb, which was far lower than the U.S. Environmental Protection Agency (EPA) standard (10 ppb). Moreover, probe CDA was also applied to detect gaseous and aqueous N2H4. Additionally, the cell imaging experiments demonstrated that probe CDA could be employed to visualize N2H4 in living organisms.

The preparation method comprises the following steps: 1) successively adding nopinone, sodium ethoxide and substituted benzaldehyde into absolute ethyl alcohol under stirring, then carrying out heating and a reflux reaction, carrying out rotary evaporation to remove the solvent after completion of the reaction, carrying out extraction with an organic solvent until an obtained solution is neutral, carrying out recrystallization after spin-drying of the organic solvent so as to obtain solid alpha,beta-unsaturated ketene; and 2) mixing alpha,beta-unsaturated ketene with N-substituted thiosemicarbazide, adding an organic solvent, carrying out heating until a reflux reaction occurs, removing the solvent through rotary evaporation after completion of the reaction, and carrying out separation and purification via silica gel column chromatography so as to obtain the nopinone thiosemicarbazone derivative. The nopinone thiosemicarbazone derivative has substantial antitumor activity to MDA-MB-231 cells, SMMC-7721 cells and Hela cells, and the IC50 values of the derivative in inhibition of the MDA-MB-231 cells, the SMMC-7721 cells and the Hela cells are 2.79 [mu]M, 2.65 [mu]M and 3.64 [mu]M, respectively; so the nopinone thiosemicarbazone derivative is extensively applicable to preparation of anticancer drugs.

The preparation method comprises steps as follows: iso-longitolanone which is a derivative of longifolene as a natural renewable resource is taken as a raw material and is subjected to condensation with benzaldehyde, and 7-benzylidene-iso-longitolanone is produced; 7-benzylidene-iso-longitolanone is subjected to condensation and cyclization with guanidine hydrochloride, and 6,6,10,10-tetramethyl-4-phenyl-5,7,8,9,10 alpha-hexahydro-6H-6 alpha,9-methano-benzo[h] quinazoline-2-amine is obtained and subjected to condensation with 2-hydroxy-1-naphthaldehyde, 1-(((6,6,10,10- tetramethyl-4-phenyl-5,7,8,9,10alpha-hexahydro-6H-6alpha,9-methano-benzo[h]-2-quinolyl) imidogen)methyl)naphthalene-2-phenol is obtained, the compound can only react with Zn to form a complex, and the complex emits green fluorescence under the irradiation of 365 nm ultraviolet light and can be taken as the fluorescent probe for Zn concentration detection.
