Zinc (Zn) is an essential component of thousands of proteins in plants, although it is toxic in excess. In this review, the dominant fluxes of Zn in the soil-root-shoot continuum are described, including Zn inputs to soils, the plant availability of soluble Zn(2+) at the root surface, and plant uptake and accumulation of Zn. Knowledge of these fluxes can inform agronomic and genetic strategies to address the widespread problem of Zn-limited crop growth. Substantial within-species genetic variation in Zn composition is being used to alleviate human dietary Zn deficiencies through biofortification. Intriguingly, a meta-analysis of data from an extensive literature survey indicates that a small proportion of the genetic variation in shoot Zn concentration can be attributed to evolutionary processes whose effects manifest above the family level. Remarkable insights into the evolutionary potential of plants to respond to elevated soil Zn have recently been made through detailed anatomical, physiological, chemical, genetic and molecular characterizations of the brassicaceous Zn hyperaccumulators Thlaspi caerulescens and Arabidopsis halleri.

Publisher Summary This chapter illustrates the literature on those plants and micro-organisms which can combat excessive quantities of heavy metal ions. Heavy metals include those metals which have density greater than five. Their common feature in regard to biological life is that in excessive quantities they are poisonous and can cause death of most living organisms. However, certain organisms possess an ability to survive under conditions of metal contamination which can prove toxic to other living things. Toxic levels of heavy metals can occur under several circumstances. The chapter focuses on the type of contamination in which the soil itself contains large quantities of these metals. The contamination results from the presence of undisturbed metal ore near the soil surface causing anomalies or from the actual mining of ore bodies. The vegetation in such areas is influenced by one overriding factor, namely, metal concentration. The habitats are usually spatially distinct and clear cut. It is observed that the sequence and pattern of genetic change responsible for colonization of metal-contaminated areas serves as a unique record of natural selection in action.

Heavy metal tolerance in plants

Chromium, in the trivalent form (Cr(III)), is an important component of a balanced human and animal diet and its deficiency causes disturbance to the glucose and lipids metabolism in humans and animals. In contrast, hexavalent Cr (Cr(VI)) is highly toxic carcinogen and may cause death to animals and humans if ingested in large doses. Recently, concern about Cr as an environmental pollutant has been escalating due to its build up to toxic levels in the environment as a result of various industrial and agricultural activities. In this review, we present the state of knowledge about chromium mobility and distribution in the environment and the physiological responses of plants to Cr with the desire to understand how these processes influence our ability to use low cost, environmentally friendly biological remediation technologies to clean up Cr-contaminated soils, sediments, and waters. The use of biological remediation technologies such as bioremediation and phytoremediation for the cleanup of Cr-contaminated areas has received increasing interest from researchers worldwide. Several methods have been suggested and experimentally tested with varying degrees of success.

Background: Pichia pastoris has been recognized as an effective host for recombinant protein production. A number of studies have been reported for improving this expression system. However, its physiology and cellular metabolism still remained largely uncharacterized. Thus, it is highly desirable to establish a systems biotechnological framework, in which a comprehensive in silico model of P. pastoris can be employed together with high throughput experimental data analysis, for better understanding of the methylotrophic yeast's metabolism. Results: A fully compartmentalized metabolic model of P. pastoris (iPP668), composed of 1,361 reactions and 1,177 metabolites, was reconstructed based on its genome annotation and biochemical information. The constraints-based flux analysis was then used to predict achievable growth rate which is consistent with the cellular phenotype of P. pastoris observed during chemostat experiments. Subsequent in silico analysis further explored the effect of various carbon sources on cell growth, revealing sorbitol as a promising candidate for culturing recombinant P. pastoris strains producing heterologous proteins. Interestingly, methanol consumption yields a high regeneration rate of reducing equivalents which is substantial for the synthesis of valuable pharmaceutical precursors. Hence, as a case study, we examined the applicability of P. pastoris system to whole-cell biotransformation and also identified relevant metabolic engineering targets that have been experimentally verified. Conclusion: The genome-scale metabolic model characterizes the cellular physiology of P. pastoris, thus allowing us to gain valuable insights into the metabolism of methylotrophic yeast and devise possible strategies for strain improvement through in silico simulations. This computational approach, combined with synthetic biology techniques, potentially forms a basis for rational analysis and design of P. pastoris metabolic network to enhance humanized glycoprotein production.

Absorption of Cations by Roots. Effects of Hydrogen Ions and Essential Role of Calcium

The absorption of rubidium by excised barley roots from solutions containing calcium was a strictly linear function of time for 1 hour and was temperature-sensitive throughout; there was no evidence of an initial nonmetabolic exchange phase of uptake. The rubidium absorbed reached concentrations many times the external concentration without any slackening of the rate of absorption-evidence for a high degree of irreversibility of the overall absorption process. This is discussed in terms of the "enzyme-kinetic" model of ion transport by carriers.
