Silicon carbide (SiC) has been around for more than 100 years as an industrial material and has found wide and varied applications because of its unique electrical and thermal properties. In recent years there has been increased attention to SiC as a viable material for biomedical applications. Of particular interest in this review is its potential for application as a biotransducer in biosensors. Among these applications are those where SiC is used as a substrate material, taking advantage of its surface chemical, tribological and electrical properties. In addition, its potential for integration as system on a chip and those applications where SiC is used as an active material make it a suitable substrate for micro-device fabrication. This review highlights the critical properties of SiC for application as a biosensor and reviews recent work reported on using SiC as an active or passive material in biotransducers and biosensors.

Silicon carbide: a versatile material for biosensor applications

We describe the formation of alkyl monolayers on silicon carbide (SiC) and silicon-rich silicon nitride (SixN4) surfaces, using UV irradiation in the presence of alkenes. Both the surface preparation and the monolayer attachment were carried out under ambient conditions. The stable coatings obtained in this way were studied by water contact angle measurements, infrared reflection absorption spectroscopy, X-ray reflectivity, and X-ray photoelectron spectroscopy. Besides unfunctionalized 1-alkenes, methyl undec-10-enoate, and 2,2,2-trifluoroethyl undec-10-enoate were also grafted onto both substrates. The resulting ester-terminated surfaces could then be further reacted after hydrolysis using amide chemistry to easily allow the attachment of amine-containing compounds.

Covalently Attached Organic Monolayers on SiC and SixN4 Surfaces: Formation Using UV Light at Room Temperature

The surface modification of solution-gated organic field-effect transistors is investigated. The introduction of different surface groups leads to a control of the pH sensitivity, determined by the pKa value of the added surface moiety. Together with the successful demonstration of enzyme modification of the surface, this work reveals the large potential of organic SGFETs for biosensor applications.

Silicon carbide has been proved as a candidate for power and high-frequency devices. In this paper, we show the application of nanocrystalline 3C-SiC as an electrochemical electrode and its electrochemical functionalization for biosensing applications. SiC electrodes show a wider potential window and lower background current than glassy carbon electrodes. The surface can be electrochemically functionalized with diazonium salts, as confirmed by electrochemical techniques and X-ray photoelectron spectroscopy. The nitrophenyl film is used as linker layer to bond DNA molecule to SiC. These results show that 3C-SiC can be an interesting transducer material for applications in electro- and bioelectrochemical applications.

Structured Polymer Brushes on Silicon Carbide

We demonstrate the successful covalent functionalization of n-type 6H-SiC with organosilanes. In particular, wet-chemically processed self-assembled layers of octadecyltrimethoxysilane and amino-propyldiethoxymethylsilane were investigated. The structural and chemical properties of these layers were studied by contact angle measurements, atomic force microscopy, thermal desorption, and x-ray photoelectron spectroscopy. The organic layers are smooth and wetting angles up to 100° are observed. Desorption temperatures in the range of 830K prove the covalent bonding of the organic molecules to the SiC surface. By utilizing self-assembled layers with functional end groups, attachment of complex molecules was demonstrated by immobilization of proteins on micropatterned organic layers.
