Advances in hydrotropic solutions: An updated review

Bioavailability is defined as the rate and extent (amount) of absorption of unchanged drug from its dosage form. It is one of the important parameters which are required to achieve optimal concentration of drug in systemic circulation to show a pharmacological response. A drug which has poor bioavailability shows poor aqueous solubility, slow dissolution rate, poor stability of dissolved drug at physiological pH, poor permeation through biological membrane, extensive first pass metabolism. Drugs which are poorly water soluble require high doses to obtain therapeutic plasma concentrations after oral administration of drugs. Low aqueous solubility is the major problem encountered with formulation development of new drugs. Any drug to be absorbed must be present in the form of an aqueous solution at the site of absorption. This review deals with various techniques used for the improvement of the Bioavailability of drugs. The various techniques used are size reduction, solubilising excipients, colloidal drug delivery systems, pH adjustment, solid dispersion, complexation, co‐solvency, micellar solubilisation, hydrotropy etc. The article describes about various techniques which can be utilized to enhance bioavailability of drugs enhancement for their effective absorption in the body

Various techniques of bioavailability enhancement: a review

Solubility is one of the key parameters to attain desired concentration of drug in systemic circulation to show pharmacological response. Various methods are engaged to augment the aqueous solubility of poorly water-soluble drugs and hydrotropic solubilisation is one of them. Hydrotropy is defined as a solubilization process whereby addition of a large amount of second solutes results in an increase in the aqueous solubility of another solute and chemicals which are used in the hydrotropy are called hydrotropes like sodium benzoate, sodium citrate, urea, niacinamide etc. Based on advantages like the high selectivity, independent of pH and cheap, easy availability makes this technique more prevailing than other solubilisation methods. Mixed hydrotropy is a solubilisation technique to augment the water solubility of poorly water soluble drugs by the way of different ratio of blends of hydrotropic agents which gives synergistic augmentation effect. The purpose of this review article is to illustrate the need for solubility, hydrotropic solubilisation, mechanism of action of hydrotrope, classification of hydrotropes, advantages and mixed hydrotropic technique of solubilisation for improving the solubility which in turn helps to attain absorption and improved bioavailability. 

Hydrotropy: A Promising Tool for Solubility Enhancement

INTRODUCTION Formulation of poorly soluble drugs for orally drug delivery now represent one of the most interesting challenges to formulation scientist in the pharmaceutical industries and for formulation containing poorly soluble drugs, dissolutions is the rate limiting step in the process of drug absorption. [17] There are number of solubilization techniques are available but till date there is no universal excipient or technique which can be versatile enough to solubilize wide spectrum of drug molecules. During development stage many potential candidates may be eliminated because of their poor solubility and bioavailability.[8] Solubility: When excess of solid is brought into contact with liquid, molecules of the former are removed from its surface until equilibrium is established between the molecules leaving the solid and those returning to it. The resulting solution are said to be saturated at the temperature of the experiment, and extent to which solute dissolves is referred to as its solubility. [9] IUPAC defines solubility as the analytical composition of a saturated solution expressed as a proportion of a designated solute in a designated solvent. Solubility may be stated in units of concentration, molality, mole fraction, mole ratio, and other units. The extent of solubility of different substances varies from almost imperceptible amount to relatively large quantities, but for any given solute the solubility has a constant value at a given constant temperature. When the purity of the drug sample can be assure, the solubility value obtained in weak acid and weak base can be assumed to be intrinsic solubility (C0) i.e. the fundamental solubility of the NCE when completely unionized. The solubility should be ideally at two temperatures • 40C To ensure physical and chemical stability. Techniques for solubility enhancement of Hydrophobic drugs: A Review

Bioavailability is defined as the rate and extent of absorption of unchanged drug from its dosage form. The oral bioavailabil ity of drugs with poor solubility and reasonable permeability is limited by the drug dissolution step from drug products. Low aqueous solubility is the major problem encountered with formulation development of new drugs. The article briefly highlights traditional and novel techniques that are used for solubility enhancement of BCS Class II drugs are discussed in this article. The Traditional techniques include use of co-solvents, hydrotrophy, micronization, change in dielectric constant of solvent, amorphous forms, chemical modification of drug, use of surfactants etc. Novel technologies are size reduction technologies, lipid based delivery system, micellar technologies, solid dispersion and many more.

The effectiveness of formulation depends particularly on how efficiently is drug available at the site of action. Therapeutic effectiveness of a drug basically depends upon bioavailability and the solubility of drug moiety.  Most of the chemical entities that are being discovered are lipophilic and have poor aqueous solubility. A more than 40% drug suffers from poor water solubility. Currently number of techniques addressed the enhancement of solubility and dissolution rate of poorly soluble drugs. Hydrotropic solubilization is one of them. Hydrotropy is a solubilization phenomenon whereby addition of large amounts of a second solute results in an increase in the aqueous solubility of another solute. A hydrotrope is a compound that solubilises hydrophobic compounds in aqueous solutions. To solubilize water insoluble drugs especially in case of oral formulation, solubility remains a critical factor so for in this review various solubility enhancement techniques are highlighted and a brief review of hydrotropy and its preparation are discussed.
