Modulating ryanodine receptors with dantrolene attenuates neuronopathic phenotype in Gaucher disease mice.
Benjamin Liou,Yanyan Peng,Ronghua Li,Venette Inskeep,Wujuan Zhang,Brian Quinn,Nupur Dasgupta,Rachel Blackwood,Kenneth D.R. Setchell,Kenneth D.R. Setchell,Sheila M. Fleming,Gregory A. Grabowski,John Marshall,Ying Sun,Ying Sun +14 more
TL;DR: Modulating Ryrs has neuroprotective effects in nGD through mechanisms that protect the mitochondria, autophagy, Ryr expression and enhance GCase activity, suggesting calcium signalling stabilization, e.g. with dantrolene, could be a potential disease modifying therapy for nGD.
Reversal of advanced disease in lysosomal acid lipase deficient mice: a model for lysosomal acid lipase deficiency disease.
Ying Sun,Ying Sun,You-Hai Xu,You-Hai Xu,Hong Du,Brian Quinn,Benjamin Liou,Lori Stanton,Venette Inskeep,Huimin Ran,Phillip Jakubowitz,Phillip Jakubowitz,Nicholas Grilliot,Nicholas Grilliot,Gregory A. Grabowski,Gregory A. Grabowski +15 more
TL;DR: Results show that LAL deficiency can be improved biochemically and histopathologically by various dosages of ET, even in advanced disease.

A convenient approach to facilitate monitoring Gaucher disease progression and therapeutic response
Wujuan Zhang,Melissa Oehrle,Carlos E. Prada,Ida Vanessa Doederlein Schwartz,Ida Vanessa Doederlein Schwartz,Somchai Chutipongtanate,Duangrurdee Wattanasirichaigoon,Venette Inskeep,Mei Dai,Dao Pan,Ying Sun,Kenneth D.R. Setchell +11 more
TL;DR: A novel robust and accurate hydrophilic interaction liquid chromatography tandem mass spectrometric method (HILIC-MS/MS) for the direct measurement of glucosylsphingosine (GlcS) in dried plasma spots (DPS) proved to be a very convenient approach for sample collection, storage and shipping nationwide and internationally.

Glucosylceramide induced complement activation triggers inflammation in Gaucher disease (CCR5P.212)
Manoj K. Pandey,Stuart Tinch,Venette Inskeep,Wujuan Zhang,Kenneth D.R. Setchell,Jörg Köhl,Gregory A. Grabowski +6 more
TL;DR: The data suggest that GC induced C 5a contributes in disease development and targeting C5a signaling may have therapeutic potential for GD.

Intravenous infusion of iPSC-derived neural progenitors expressing GCase ameliorates alpha-synuclein aggregates in a mouse model of Gaucher disease
Yanyan Peng,Benjamin Liou,Venette Inskeep,Rachel Blackwood,Sheila M. Fleming,Christopher N. Mayhew,Ying Sun +6 more
TL;DR: The efficacy of non-invasive delivery of iPSC-derived NPCs overexpressing hGCase in GD mouse model is demonstrated and the feasibility of combined cell and gene therapy for GBA1-associated PD is established.
