Role of Salicylic Acid in Plants

The present invention provides recombinant or chimaeric DNA molecules comprising plant SAR genes, wherein the wild-type genes corresponding to said SAR genes can be chemically induced in a plant in a protein-synthesis independent or in a protein-synthesis dependent manner. Methods for obtaining these DNA molecules are also provided. Additionally, the present invention provides chemically inducible wheat genes, Arabidopsis chitinase IV, Maize PR-1mz, and Maize thaumatin PR-5mz. The anti-pathogenic sequences according to the invention can be genetically engineered and transformed into plants to obtain transgenic plants with enhanced resistance to disease. In one embodiment of the present invention, the chemically regulatable DNA promoter sequence of the Arabidopsis Pr-1 gene is provided. Another aspect of the present invention is a method of improving protection of a plant against a pest comprising transgenically expressing in said plant two or more DNA molecules encoding anti-pathogenic proteins, wherein the transgenically expressed proteins exert a synergistic effect.

Chemically regulatable and anti-pathogenic DNA sequences and uses thereof

The induction by cytokinin stress and ethylene of nine different tobacco mosaic virus-inducible mRNA classes (termed A-I) encoding pathogenesis-related (PR) proteins was studied. The induced mRNA levels were compared to basal levels in healthy tobacco plants grown in tissue culture and in a greenhouse. Cytokinin stress and ethylene were found to induce different subsets of the mRNAs, indicating that ethylene is not the primary inducing signal in cytokinin-stressed shoots. mRNAs F, H and G encoding the basic hydrolytic enzymes chitinase, β-1,3-glucanase and a basic equivalent of PR-1, respectively, were found to be expressed at high levels in roots of healthy plants. mRNAs D, I and B encoding the acidic equivalents of the proteins proved to be present at low levels in healthy plants. These results indicate that genes encoding basic and acidic isoforms of pathogenesis-related proteins are differentially regulated.

Tobacco genes encoding acidic and basic isoforms of pathogenesis-related proteins display different expression patterns

The cis-acting elements for regulating gene expression of the tobacco pathogenesis-related 1a protein gene were analyzed in transgenic plants. The 59-flanking 2.4-kilobase fragment from the pathogenesis-related 1a protein gene was joined to the bacterial beta-glucuronidase gene and introduced into tobacco cells by Agrobacterium-mediated gene transfer. Promoter activity was monitored by quantitative and histochemical assay of beta-glucuronidase activity in leaves of regenerated transgenic plants. The level of beta-glucuronidase activity was clearly increased by treatment with salicylic acid, by cutting stress, and by local lesion formation caused by tobacco mosaic virus infection. Cytochemical studies of the induced beta-glucuronidase activity revealed tissue-specific and developmentally regulated expression of the pathogenesis-related 1a gene after stress or chemical treatment and after pathogen attack. To identify the cis-acting element more precisely, a series of 59-deleted chimeric genes was constructed and transformed into tobacco plants. Transgenic plants with a 0.3-kilobase fragment of the 59-flanking region of the pathogenesis-related 1a gene had the same qualitative response as those with the 2.4-kilobase fragment upon treatment with salicylic acid or infection with TMV. Thus, the 0.3-kilobase DNA sequence fragment was sufficient to allow the regulated expression of the pathogenesis-related 1a gene.

The present invention provides chemically regulatable 
DNA sequences capable of regulating transcription of an 
associated DNA sequence in plants or plant tissues, chimeric 
constructions containing such sequences, vectors containing 
such sequences and chimeric constructions, and transgenic 
plants and plant tissues containing these chimeric 
constructions. Also provided are a novel signal peptide 
sequence, genes which code for this sequence, and newly 
identified PR protein genes. The chimeric constructions and 
transgenic plants and plant tissues may be used in an assay 
for new chemical regulators.
