Isolation and enrichment of two sublobular compartment-specific endothelial cell subpopulations from liver sinusoids

An optimized method for mouse liver sinusoidal endothelial cell isolation.

Large granular lymphocytes (LGLs) differ from other lymphocytes in their recirculation pattern and are distributed preferentially in nonlymphoid organs such as the liver and lung. The liver-associated LGLs adhere strongly to the sinusoidal endothelium and show a natural killer (NK) cytotoxicity against incoming metastatic tumor cells; this reaction occurs very rapidly because, in contrast to the immune response, it does not require complex processes in the lymphoid tissue. They have been extensively studied morphologically in terms of pit cells. LGLs have two characteristic cell organelles which participate in the NK cytolysis, i. e., dense granules and rod-cored vesicles. The former are lysosomes derived from multivesicular bodies and contain pore-forming proteins. The latter are the secretory vesicles exclusively seen in LGLs and are markedly increased in number when the NK function is augmented by biological response modifiers. These two structures are believed to be exocytosed in the space between LGL and the conjugated tumor cell. The microenvironment of the liver sinusoids, which includes Kupffer cells, endothelial cells and other lymphocytes, is considered to regulate the function of the liver-associated LGLs. Liver-associated LGLs, as well as Kupffer cells, are intrinsically involved in the defense system of the liver under various physiological and pathological conditions.

Liver-associated large granular lymphocytes: morphological and functional aspects.

To investigate the cellular communication in theliver, nitric oxide (NO) production by sinusoidal cellsand hepatocytes by stimulation with cytokines andKupffer cell-conditioned medium was quantitatively analyzed. NO production by the cells wasmeasured by the Griess reaction, and nitric oxidesynthase (iNOS) transcription level by a competitiveRT-PCR assay using mutant iNOS mRNA as a standard. NOproduction and iNOS mRNA transcriptional levels in Kupffercells were markedly increased by stimulation withlipopolysaccharide (LPS) and interferon-γ(IFN-γ), and moderately by interleukin-1β(IL-1β). NO production by hepatocytes was notsignificantly enhanced by LPS, but was markedly enhancedby IL-1β or the combination of tumor-necrosisfactor-α (TNF-α) and IFN-γ. HepatocyteNO production and iNOS mRNA levels were markedly enhanced bythe LPS-activated Kupffer cell conditioned medium, butthese effects were reduced by heat treatment or anti-TNFantibody. Although NG-monomethyl-L-arginine acetate and dexamethasone reduced NO productionby the cells, the iNOS mRNA level was reduced bydexamethasone only. Gel-shift assay showed NF-κBactivation in hepatocytes during this activation. These data reinforce the importance of cellularcommunication between sinusoidal cells andhepatocytes.

Hepatocyte nitric oxide production is induced by Kupffer cells

The aim of the present study was to identify non-parenchymal liver cells (NPLC) in B10.D2 mice and to determine their percentage frequency. The isolation of NPLC was carried out using the collagenase/pronase technique. Using functional techniques (latex phagocytosis, immunocytochemical detection of surface-bound and intracytoplasmic antigens) and morphological methods (light and electron microscopy), the following cell types were identified, and their percentage frequency in the NPLC determined: endothelial cells (50%), macrophages (23%), desmin-positive cells (14%), immunocompetent cells (10%, including T-, B-cells, pit and large vacuolated cells-both immunopositive to the asialo-GM 1 antigen) and unidentified cells (3%). These results show that, apart from the more familiar varieties of NPLC, two groups of cells exist in the liver which have not yet been fully identified and in which the immunocompetent cells predominate numerically.

Identification and percentage frequency of isolated non-parenchymal liver cells (NPLC) in the mouse

The mechanism of cell replacement of non-parenchymal liver cells (NPLC) was investigated in an attempt to answer the question: to what extend are NPLC replaced by proliferation of local resident cells and to what extend by cells originating from the bone-marrow. The bone-marrow of mice was used, and H2k positive cells from F1 (B10.BR X B10.D2) hybrid mice were transplanted into irradiated H2k negative parent animals. Their NPLC were isolated and tested immunocytochemically with a monoclonal anti-H2k antibody for the presence of H2k positive cells. During the whole of the experiment (from the 5th to the 20th week following transplantation) H2k positive cells (macrophages and non-macrophages) were present, and made up an average of nearly one third of the NPLC. The H2k positive (immigrant) non-macrophages showed essentially more active DNA synthesis than these H2k negative cells. To increase cell turnover, we injected one group of animals with endotoxin. At the time of maximum replacement, more than 50% of the NPLC (macrophages and non-macrophages) were recruited from cells of bone-marrow origin. Noteworthy here was the proportionate level of the H2k positive macrophages (more than 70% of all liver macrophages). The DNA synthesis of both the H2k positive macrophages and non-macrophages was more than twice that of the H2k negative NPLC during the regeneration process following administration of endotoxin. Our observations suggest that the bone-marrow contributes significantly to the replacement of macrophages and non-macrophages of NPLC in both health and disease.
