Recently a novel DNA virus (Torque Teno Virus (TTV) has been identified in Japan and shown to be associated with elevated aminotransferase s levels after transfusion. However the exact role of TTV in pathogenesis of liver disease is yet to be established. The purpose of this study was to determine the prevalence of TTV in thalassemic patients and its relationship with elevated alanine-aminotransfrase (ALT) and aspartateaminotransfrase (AST). This cross-sectional analysis study was conducted on 452 thallasemic patients. Serums were collected from all of the patients, first ALT and AST levels were determined. Then, after DNA extraction, TTV DNA was amplified and detected using semi-nested PCR, followed by gel electrophoresis. Demographic characteristics and clinical data were collected from each participant for statistical analysis. The findings showed that 160 of 452 (35.4%) samples had TTV DNA detected by PCR. From 160 TTV DNA positive, 98 (61.20%) were female and 62 (38.80%) of them were male (P=0.549). The mean ALT and AST values in TTV positive group were higher than in TTV negative group, and the difference was statistically significant (p<0.0001). The result showed that the prevalence of TTV in thalassemic patients in Jahrom is less than other studies in Iran and the mean ALT and AST values in TTV positive individuals were about 2 times more than in TTV negative individuals.

Molecular Prevalence and Clinical Importance of Torque Teno Virus Infection in Thalassemia Patients as High Risk Individuals

Cannabinoids impair hippocampus-dependent memory in both humans and animals, but the network mechanisms responsible for this effect are unknown. Here we show that the cannabinoids Δ9-tetrahydrocannabinol and CP55940 decreased the power of theta, gamma and ripple oscillations in the hippocampus of head-restrained and freely moving rats. These effects were blocked by a CB1 antagonist. The decrease in theta power correlated with memory impairment in a hippocampus-dependent task. By simultaneously recording from large populations of single units, we found that CP55940 severely disrupted the temporal coordination of cell assemblies in short time windows (<100 ms) yet only marginally affected population firing rates of pyramidal cells and interneurons. The decreased power of local field potential oscillations correlated with reduced temporal synchrony but not with firing rate changes. We hypothesize that reduced spike timing coordination and the associated impairment of physiological oscillations are responsible for cannabinoid-induced memory deficits.

The role of serotonin in drug use and addiction.

The hippocampus plays an important role in emotional and cognitive processing, and both of these domains are affected in patients with major depressive disorder (MDD). Extensive preclinical research and the notion that modulation of serotonin (5-HT) neurotransmission plays a key role in the therapeutic efficacy of selective serotonin reuptake inhibitors (SSRIs) support the view that 5-HT is important for hippocampal function in normal and disease-like conditions. The hippocampus is densely innervated by serotonergic fibers, and the majority of 5-HT receptor subtypes are expressed there. Furthermore, hippocampal cells often co-express multiple 5-HT receptor subtypes that can have either complementary or opposing effects on cell function, adding to the complexity of 5-HT neurotransmission. Here we review the current knowledge of how 5-HT, through its various receptor subtypes, modulates hippocampal output and the activity of hippocampal pyramidal cells in rodents. In addition, we discuss the relevance of 5-HT modulation for cognitive processing in rodents and possible clinical implications of these results in patients with MDD. Finally, we review the data on how SSRIs and vortioxetine, an antidepressant with multimodal activity, affect hippocampal function, including cognitive processing, from both a preclinical and clinical perspective.

Effects of serotonin in the hippocampus: how SSRIs and multimodal antidepressants might regulate pyramidal cell function.

Addiction is a chronic, recurring and complex disorder. It is characterized by anomalous behaviors that are linked to permanent or long-lasting neurobiological alterations. Furthermore, the endocannabinoid system has a crucial role in mediating neurotransmitter release as one of the main neuromodulators of the mammalian central nervous system. The purpose of the present review is to instruct readers about the functional and structural interactions between the endocannabinoid system and the main neurotransmitter systems of the central nervous system in the context of drug addiction. With this aim, we have systematically reviewed the main findings of most of the existing literature that explores cross-talk in the five brain areas that are most traditionally implicated in addiction: amygdala, prefrontal cortex, nucleus accumbens, hippocampus and ventral tegmental area (VTA). The neurotransmission systems influenced by the pharmacology of the endocannabinoid system in these brain areas, which are reviewed here, are gamma-aminobutyric acid (GABA), glutamate, the main biogenic amines (dopamine, noradrenaline and serotonin), acetylcholine and opioids. We show that all of these neurotransmitter systems can be modulated differentially in each brain area by the activation or deactivation of cannabinoid CB1 brain receptors. Specifically, most of the studies relate to the hippocampus and nucleus accumbens. Moreover, the neurotransmitter with the fewest number of related studies is acetylcholine (excepting in the hippocampus), whereas there is a large number that evaluates GABA, glutamate and dopamine. Finally, we propose a possible interpretation of the role of the endocannabinoid system in the phenomenon of addiction.

The pharmacology of the endocannabinoid system: functional and structural interactions with other neurotransmitter systems and their repercussions in behavioral addiction


 Radio galaxies are a key population to understand the importance of relativistic jets in active galactic nucleus (AGN) feedback. We present the results of a systematic, broad-band X-ray spectral analysis of hard X-ray selected radio galaxies to investigate their nuclear structures. In this study, we focus on the seven most radio-loud, X-ray obscured narrow-line radio galaxies in the Swift/BAT 70 month AGN catalogue. The spectra from 0.5 keV up to 66 keV obtained with Suzaku and NuSTAR of six objects are newly analysed here by utilizing the X-ray clumpy torus model (xclumpy), whereas we refer to the literature for the results of Centaurus A. We find that these radio galaxies have similar torus covering fractions compared with radio-quiet AGNs at the same Eddington ratios (−3 < log λEdd < −1). This result implies that (1) the torus structure is not an important factor that determines the presence of jets and (2) AGN jets have physically little effect on the torus.

Broad-band X-ray spectral study of nuclear structure in local obscured radio galaxies


 To understand the evolution of global accretion disk structure in the “rebrightening” phase of MAXI J1820+070, we perform a comprehensive analysis of its near infrared/optical/UV to X-ray spectral energy distribution (SED) utilizing data obtained by OISTER, Las Cumbres Observatory (LCO), Swift, NICER, and NuSTAR in 2019. Optical spectra observed with Seimei telescope in 2019 and 2020 are also analyzed. On the basis of the optical and X-ray light curves and their flux ratios, we divide the whole phase into three periods, Periods I (flux rise), II (decay), and III (dim). In the first two periods, the source stayed in the low/hard state (LHS), where the X-ray (0.3–30 keV) and optical/UV SED can both be fitted with power-law models. We interpret that the X-ray emission arises from hot corona via Comptonization, whereas the optical/UV flux is dominated by synchrotron radiation from the jets, with a partial contribution from the irradiated disk. The optical/UV power-law component smoothly connects to a simultaneous radio flux, supporting its jet origin. Balmer line profiles in the optical spectra indicate that the inner radius of an irradiated disk slightly decreased from ∼2 × 105rg (Period I) to ∼1 × 105rg (Period II), where rg is the gravitational radius, implying a change of the hot corona geometry. In Period III, the SED can be reproduced by an advection-dominated accretion flow and jet emission. However, the double-peaked Hα emission line indicates that a cool disk remained at large radii.
