NFATc1 controls the cytotoxicity of CD8+ T cells
Stefan Klein-Hessling,Khalid Muhammad,Matthias Klein,Tobias Pusch,Ronald Rudolf,Jessica Flöter,Musga Qureischi,Andreas Beilhack,Martin Vaeth,Martin Vaeth,Carsten Kummerow,Christian S. Backes,Rouven Schoppmeyer,Ulrike Hahn,Markus Hoth,Tobias Bopp,Friederike Berberich-Siebelt,Amiya K. Patra,Amiya K. Patra,Andris Avots,Nora Müller,Almut Schulze,Edgar Serfling +22 more
TL;DR: Genome-wide ChIP-seq data indicate that NFATc1 is an important regulator of cytotoxic T lymphocyte effector functions, and this work shows it controls the cytotoxicity and metabolic switching of activated CD8+ T cells required for optimal response to bacteria and tumor cells.

Selective NFAT targeting in T cells ameliorates GvHD while maintaining antitumor activity
Martin Vaeth,Carina A. Bäuerlein,Tobias Pusch,Janina Findeis,Martin Chopra,Anja Mottok,Andreas Rosenwald,Andreas Beilhack,Friederike Berberich-Siebelt +8 more
TL;DR: It is found that absence of the transcription factors NFAT (nuclear factor of activated T cells) in T cells prevents harmful GvHD, but preserves the valuable GvL, which is a life-threatening immunological complication after allogenic hematopoietic stem cell transplantation.

The Transcription Factor NFATc1 Supports the Rejection of Heterotopic Heart Allografts.
Johannes Baur,Christoph Otto,Ulrich Steger,Stefan Klein-Hessling,Khalid Muhammad,Tobias Pusch,Krisna Murti,Rhoda Wismer,Christoph-Thomas Germer,Ingo Klein,Nora Müller,Edgar Serfling,Andris Avots +12 more
TL;DR: It is shown that the activity of NFATc1, the most prominent NFAT factor in activated lymphocytes supports the acute rejection of heterotopic heart allografts and favors NFAT c1 as a molecular target for the development of new strategies to control the cytotoxicity of T cells upon organ transplantation.

NFATc1 induction in peripheral T and B lymphocytes
Matthias Hock,Martin Vaeth,Ronald Rudolf,Amiya K. Patra,Duong Anh Thuy Pham,Khalid Muhammad,Tobias Pusch,Tobias Bopp,Edgar Schmitt,Rene Rost,Friederike Berberich-Siebelt,Dimitri Tyrsin,Sergei Chuvpilo,Andris Avots,Edgar Serfling,Stefan Klein-Hessling +15 more
TL;DR: It is shown that in novel bacterial artificial chromosome transgenic mice that express EGFP under the control of entire Nfatc1 locus the N fatc1/Egfp transgene is expressed as early as in double-negative thymocytes and in nonstimulated peripheral T and B cells.
Impact of a Laboratory-Developed Phenotypic Rapid Susceptibility Test Directly From Positive Blood Cultures on Time to Narrowest Effective Therapy in Patients With Gram-Negative Bacteremia: A Prospective Randomized Trial
Alyssa Christensen,Brent W. Footer,Tobias Pusch,Kim Heath,M. Iqbal,Lian Wang,Gregory B Tallman,Cameron Cover,Jennifer E. Marfori,Brian A. Kendall,Nick Stucky,Meagan Greckel,Ivor L Thomas,Katelynn Tran,Salena Yip,Margret Oethinger +15 more
TL;DR: In patients with GN-BSI, rAST did not significantly decrease time to narrowest effective therapy but did decrease timeto oral antibiotics and length of hospital stay.
