Investigation of poly(2-ethyl-2-oxazoline) as a novel extended release polymer for hot-melt extrusion paired with fused deposition modeling 3D printing

Significant efforts on design and development of advanced drug delivery systems for targeted cancer chemotherapy continue to be a major challenge. Here we reported a kind of reduction-responsive PEGylated doxorubicin (DOX) prodrug via the simple esterification and amidation reactions, which self-assembled into the biodegradable micelles in solutions. Since there was an obvious difference in the reduction potentials between the oxidizing extracellular milieu and the reducing intracellular fluids, these PEG-disulfide-DOX micelles were localized intracellularly and degraded rapidly by the stimulus to release the drugs once reaching the targeted tumors, which obviously enhanced the therapeutic efficacy with low side effects. Moreover, these reduction-sensitive micelles could also physically encapsulate the free DOX drug into the polymeric cargo, exhibiting a two-phase programmed drug release behavior. Consequently, it showed a potential to develop intelligent and multifunctional chemotherapeutic payload transporter for the effective tumor therapy.

PEGylated Doxorubicin Prodrug-Forming Reduction-Sensitive Micelles With High Drug Loading and Improved Anticancer Therapy

The bactericidal effects of inhalable ciprofloxacin (CIP) loaded-poly(2-ethyl-2-oxazoline) (PEtOx) nanoparticles (NPs) with traces of zinc oxide (ZnO) were investigated against clinical strains of the respiratory pathogens Staphylococcus aureus and Pseudomonas aeruginosa. CIP-loaded PEtOx NPs retained their bactericidal activity within the formulations compared to free CIP drugs against these two pathogens, and bactericidal effects were enhanced with the inclusion of ZnO. PEtOx polymer and ZnO NPs did not show bactericidal activity alone or in combination against these pathogens. The formulations were tested to determine the cytotoxic and proinflammatory effects on airway epithelial cells derived from healthy donors (NHBE), donors with chronic obstructive pulmonary disease (COPD, DHBE), and a cell line derived from adults with cystic fibrosis (CFBE41o-) and macrophages from healthy adult controls (HCs), and those with either COPD or CF. NHBE cells demonstrated maximum cell viability (66%) against CIP-loaded PEtOx NPs with the half maximal inhibitory concentration (IC50) value of 50.7 mg/mL. CIP-loaded PEtOx NPs were more toxic to epithelial cells from donors with respiratory diseases than NHBEs, with respective IC50 values of 0.103 mg/mL for DHBEs and 0.514 mg/mL for CFBE41o- cells. However, high concentrations of CIP-loaded PEtOx NPs were toxic to macrophages, with respective IC50 values of 0.002 mg/mL for HC macrophages and 0.021 mg/mL for CF-like macrophages. PEtOx NPs, ZnO NPs, and ZnO-PEtOx NPs with no drug were not cytotoxic to any cells investigated. The in vitro digestibility of PEtOx and its NPs was investigated in simulated lung fluid (SLF) (pH 7.4). The analysed samples were characterized using Fourier transform infrared spectroscopy (ATR-FTIR), scanning electron microscopy (SEM), and UV–Vis spectroscopy. Digestion of PEtOx NPs commenced one week following incubation and was completely digested after four weeks; however, the original PEtOx was not digested after six weeks of incubation. The outcome of this study revealed that PEtOx polymer could be considered an efficient drug delivery carrier in respiratory linings, and CIP-loaded PEtOx NPs with traces of ZnO could be a promising addition to inhalable treatments against resistant bacteria with reduced toxicity.

Cytotoxic and Bactericidal Effects of Inhalable Ciprofloxacin-Loaded Poly(2-ethyl-2-oxazoline) Nanoparticles with Traces of Zinc Oxide

A series of disulfide-linked amphiphilic polymers polyoxaline-SS-poly(lactide) (PEtOx-SS-PLA) were prepared and self-assembled into nano-micelles in water. The anticancer drug curcumin (Cur) was selected as a model drug, the entrapment of Cur in PEtOx-SS-PLA micelles was investigated and the intracellular transport and release of Cur-loaded micelles was studied in C6 cells. The preparation of Cur-loaded polymer micelles showed that micelle size decreased after drug loading, favoring cell phagocytosis. MTT experiments showed that PEtOx-SS-PLA 52 micelles have a small IC50 (2.05 μg mL−1). The release behavior of PEtOx-SS-PLA 52 drug-loaded micelles in C6 cells showed that polymer micelle enhanced the intracellular release of Cur, and increased the inhibition effect of cancer cells. In a word, these reduction and pH-dual sensitive, biodegradable, hydrophilic shell-discarding PEtOx-SS-PLA micelles have great potential for future tumour administration.

Biodegradable reduction and pH dual-sensitive polymer micelles based on poly(2-ethyl-2-oxazoline) for efficient delivery of curcumin

The invention relates to preparation and application of a reducing sensitive nano-micelle, belonging to preparation and application of an amphiphilic block polymer. A hydrophilic section and a hydrophobic section of the amphiphilic block polymer are connected with each other by a reducing responsive sulfur-sulfur bond; the nano-micelle is formed by self-assembling; the reducing sensitive nano-micelle consists of a shell and an inner core, wherein the shell is a hydrophilic polymer, and the inner core is a hydrophobic polymer; the hydrophilic section of the amphiphilic block polymer is polyphosphate, the hydrophobic section of the amphiphilic block polymer is poly(benzyl glutamate), and the hydrophilic section and the hydrophobic section of the amphiphilic block polymer are connected with each other by the reducing responsive sulfur-sulfur bond. The reducing sensitive nano-micelle has the advantages that the drug-loaded nano-micelle is difficult to dissociate outside the cells or in the blood, so that the drug encapsulated in the nano-micelle is enabled to be stable; once the nano-micelle enters the cancer cell, the sulfur-sulfur bond can be disconnected by glutathione which is a reducing matter in the cell, so that the nano-micelle is rapidly dissociated, the anti-cancer drug encapsulated in the nano-micelle can be rapidly and effectively released out, and an effective treatment effect is achieved; the reducing sensitive nano-micelle overcomes the defects that the drug is easy to leak in vivo, low in transport efficiency, slow in intracellular release speed, and the like.
