A Systematic Cheminformatics Analysis of Functional Groups Occurring in Natural Products
Peter Ertl,Tim Schuhmann +1 more
TL;DR: This study systematically investigates the distribution of FGs in natural products from a cheminformatics perspective by comparing FG frequencies in natural Products with those found in average synthetic molecules to identify FGs that are characteristic for molecules produced by living organisms.

The Novolactone Natural Product Disrupts the Allosteric Regulation of Hsp70
A. Quamrul Hassan,Christina A. Kirby,Wenlai Zhou,Tim Schuhmann,Roman Kityk,D. Randal Kipp,Jason Baird,Jinyun Chen,Yaoyu Chen,Franklin Chung,Dominic Hoepfner,N. Rao Movva,Raymond Pagliarini,Frank Petersen,Christopher Quinn,Douglas Frederick Quinn,Ralph Riedl,Esther K. Schmitt,Anne Schitter,Travis Stams,Christian Studer,Pascal D. Fortin,Matthias P. Mayer,Heather Sadlish +23 more
TL;DR: Biochemical and structural analyses indicate that novolactone disrupts interdomain communication by allosterically inducing a conformational change in the Hsp70 protein to block ATP-induced substrate release and inhibit refolding activities, making it a valuable tool for exploring the requirements of HSp70 chaperones in diverse cellular contexts.

Construction of a 3D-shaped, natural product like fragment library by fragmentation and diversification of natural products.
Horst Prescher,Guido Koch,Tim Schuhmann,Peter Ertl,Alex Bussenault,Meir Glick,Ina Dix,Frank Petersen,Dimitrios Lizos +8 more
TL;DR: The obtained fragment library has an excellent 3D-shape and natural product likeness, covering a novel, unexplored and underrepresented chemical space in fragment based drug discovery (FBDD).

Bile acid analogues are activators of pyrin inflammasome.
Irina Alimov,Suchithra Menon,Nadire Cochran,Rob Maher,Qiong Wang,John Alford,John Concannon,Zinger Yang,Edmund Harrington,Luis Llamas,Alicia Lindeman,Gregory R. Hoffman,Tim Schuhmann,Carsten Russ,John S. Reece-Hoyes,Stephen M. Canham,Xinming Cai +16 more
TL;DR: It is surmised that pyrin inflammasome activation through microbiota-modified bile acid metabolites such as BAA473 and BAA485 plays a role in gut microbiota regulated intestinal immune response, and the discovery of these two bioactive compounds may help to further unveil the importance of pyrIn in gut homeostasis and autoimmune diseases.
Cheminformatics Analysis of Natural Product Scaffolds: Comparison of Scaffolds Produced by Animals, Plants, Fungi and Bacteria.
Peter Ertl,Tim Schuhmann +1 more
TL;DR: The results presented here may be used as a guidance in selection of scaffolds for the design of novel NP‐like bioactive structures or NP‐inspired libraries.
