A microneedle (MN) array is a novel biomedical device adopted in medical applications to pierce through the stratum corneum while targeting the viable epidermis and dermis layers of the skin. Owing to their micron-scale dimensions, MNs can minimize stimulations of the sensory nerve fibers in the dermis layer. For medical applications, such as wound healing, biosensing, and drug delivery, the structure of MNs significantly influences their mechanical properties. Among the various microfabrication methods for MNs, fused deposition modeling (FDM), a commercial 3D printing method, shows potential in terms of the biocompatibility of the printed material (polylactic acid (PLA)) and preprogrammable arbitrary shapes. Owing to the current limitations of FDM printer resolution, conventional micron-scale MN structures cannot be fabricated without a post-fabrication process. Hydrolysis in an alkaline solution is a feasible approach for reducing the size of PLA needles printed via FDM. Moreover, weak bonding between PLA layers during additive manufacturing triggers the detachment of PLA needles before etching to the expected sizes. Furthermore, various parameters for the fabrication of PLA MNs with FDM have yet to be sufficiently optimized. In this study, the thermal parameters of the FDM printing process, including the nozzle and printing stage temperatures, were investigated to bolster the interfacial bonding between PLA layers. Reinforced bonding was demonstrated to address the detachment challenges faced by PLA MNs during the chemical etching process. Furthermore, chemical etching parameters, including the etchant concentration, environmental temperature, and stirring speed of the etchant, were studied to determine the optimal etching ratio. To develop a universal methodology for the batch fabrication of biodegradable MNs, this study is expected to optimize the conditions of the FDM-based fabrication process. Additive manufacturing was employed to produce MNs with preprogrammed structures. Inclined MNs were successfully fabricated by FDM printing with chemical etching. This geometrical structure can be adopted to enhance adhesion to the skin layer. Our study provides a useful method for fabricating MN structures for various biomedical applications.

Optimization of the fused deposition modeling-based fabrication process for polylactic acid microneedles

Over the last few years, fused filament fabrication (FFF) has become one of the most promising and widely used techniques for the rapid prototyping process. A number of studies have also shown the possibility of FFF being used for the fabrication of functional products, such as biomedical implants and automotive components. However, the poor mechanical properties possessed by FFF-processed products are considered one of the major shortcomings of this technique. Over the last decade, many researchers have attempted to improve the mechanical properties of FFF-processed products using several strategies—for instance, by applying the short fiber reinforcement (SFR), continuous fiber reinforcement (CFR), powder addition reinforcement (PAR), vibration-assisted FFF (VA-FFF) methods, as well as annealing. In this paper, the details of all these reinforcement techniques are reviewed. The abilities of each method in improving tensile, flexural, and compressive strength are discussed.

A Review on Reinforcement Methods for Polymeric Materials Processed Using Fused Filament Fabrication (FFF).

Clinical recovery from vascular diseases has increasingly become reliant upon the successful fabrication of artificial blood vessels (BVs) or vascular prostheses due to the shortage of autologous vessels and the high incidence of vessel graft diseases. Even though many attempts at the clinical implementation of large artificial BVs have been reported to be successful, the development of small-diameter BVs remains one of the significant challenges due to the limitation of micro-manufacturing capacity in complexity and reproducibility, as well as the development of thrombosis. The present study aims to develop 3D printed small-diameter artificial BVs that recapitulate the longitudinal geometric elements in the native BVs using biocompatible polylactic acid (PLA). As their intrinsic physical properties are crystallinity dependent, we used two PLA filaments with different crystallinity to investigate the suitability of their physical properties in the micro-manufacturing of BVs. To explore the mechanism of venous thrombosis, our study provided a preliminarily comparative analysis of the effect of geometry-induced flows on the behavior of human endothelial cells (ECs). Our results showed that the adhered healthy ECs in the 3D printed BV exhibited regulated patterns, such as elongated and aligned parallel to the flow direction, as well as geometry-induced EC response mechanisms that are associated with hemodynamic shear stresses. Furthermore, the computational fluid dynamics simulation results provided insightful information to predict velocity profile and wall shear stress distribution in the geometries of BVs in accordance with their spatiotemporally-dependent cell behaviors. Our study demonstrated that 3D printed small-diameter BVs could serve as suitable candidates for fundamental BV studies and hold great potential for clinical applications. 

An in vitro analysis of the effect of geometry-induced flows on endothelial cell behavior in 3D printed small-diameter blood vessels.

The objective of the present work is to evaluate the influence of material color on mechanical properties of fused deposition modeling (FDM) parts. The performance of the products is evaluated by testing eight different colors of acrylonitrile butadiene styrene (ABS) and polylactic acid (PLA) material in terms of tensile strength, compressive strength, and flexural strength. The analysis of data shows a significant difference in mechanical characteristics of prints depending on filament color. For different colors, these three strengths almost follow the same rising and falling tendency. In order to explore the relationship between mechanical strengths and filament colors, the color-mixing theory and the least-squares method are adopted to fit the best ratio coefficients of different color combinations. Results are presented showing that the strength value (e.g., tensile) of the mixed color can be evaluated through that of primary colors by fitting the other strength (e.g., compressive or flexural). It is shown that the predicted value is always no more than 7% error compared with the actual strength, in spite of two-color or three-color mixtures. An additional confirmation test with seven colored PLA filaments from different suppliers was conducted to focus on the extensibility. The outcomes show the maximum fitting errors of strengths for mixed colors in all cases are within 5%, proving the effectiveness and applicability of this predicted approach. This study can bring a detailed analysis that enables better estimation of the function of material color and contributes to improving the property of FDM printed products for consumers by choosing the suitable filament color.

Additive manufacturing (AM) methods such as material extrusion (ME) are becoming widely used by engineers, designers and hobbyists alike for a wide variety of applications. Successfully manufacturing objects using ME three-dimensional printers can often require numerous iterations to attain predictable performance because the exact mechanical behavior of parts fabricated via additive processes are difficult to predict. One of that factors that contributes to this difficulty is the wide variety of ME feed stock materials currently available in the marketplace. These build materials are often sold based on their base polymer material such as acrylonitrile butadiene styrene or polylactic acid (PLA), but are produced by numerous different commercial suppliers in a wide variety of colors using typically undisclosed additive feed stocks and base polymer formulations. This paper aims to present the results from an experimental study concerned with quantifying how these sources of polymer variability can affect the mechanical behavior of three-dimensional printed objects. Specifically, the set of experiments conducted in this study focused on following: several different colors of PLA filament from a single commercial supplier to explore the effect of color additives and three filaments of the same color but produced by three different suppliers to account for potential variations in polymer formulation.,A set of five common mechanical and material characterization tests were performed on 11 commercially available PLA filaments in an effort to gain insight into the variations in mechanical response that stem from variances in filament manufacturer, feed stock polymer, additives and processing. Three black PLA filaments were purchased from three different commercial suppliers to consider the variations introduced by use of different feed stock polymers and filament processing by different manufacturers. An additional eight PLA filaments in varying colors were purchased from one of the three suppliers to focus on how color additives lead to property variations. Some tests were performed on unprocessed filament samples, while others were performed on objects three-dimensional printed from the various filaments. This study looked specifically at four mechanical properties (Young’s modulus, storage modulus, yield strength and toughness) as a function of numerous material properties (e.g. additive loading, molecular weight, molecular weight dispersity, enthalpy of melting and crystallinity).,For the 11 filaments tested the following mean values and standard deviations were observed for the material properties considered: pa = 1.3 ± 0.9% (percent additives), Mw = 98.6 ± 16.4 kDa (molecular weight), Ð = 1.33 ± 0.1 (molecular weight dispersity), Hm = 37.4 ± 7.2 J/g (enthalpy of melting) and = 19.6 ± 2.1% (crystallinity). The corresponding mean values and standard deviations for the resulting mechanical behaviors were: E = 2,790 ± 145 MPa (Young’s modulus), E’ = 1,050 ± 125 MPa (storage modulus), Sy = 49.6 ± 4.93 MPa (yield strength) and Ut = 1.87 ± 0.354 MJ/m^3 (toughness). These variations were observed in filaments that were all manufactured from the same base polymer (e.g. PLA) and are only different in terms of the additives used by the manufacturers to produce different colors or different three-dimensional printing performance. Unfortunately, while the observed variations were significant, no definitive strong correlations were found between these observed variations in the mechanical behavior of the filaments studied and the considered material properties.,These variations in mechanical behavior and material properties could not be ascribed to any specific factor, but rather show that the mechanical of three-dimensional printed parts are potentially affected by variations in base polymer properties, additive usage and filament processing choices in complex ways that can be difficult to predict.,These results emphasize the need to take processing and thereby even filament color, into account when using ME printers, they emphasize the need for designers to use AM with caution when the mechanical behavior of a printed part is critical and they highlight the need for continued research in this important area. While all filaments used were marked as PLA, the feedstock materials, additives and processing conditions created significant differences in the mechanical behavior of the printed objects evaluated, but these differences could not be accurately and reliably predicted as function of the observed material properties that were the focus of this study.,The testing methods used in the study can be used by engineers and creators alike to better analyze the material properties of their filament printed objects, to increase success in print and mechanical design. Furthermore, the results clearly show that as AM continues to evolve and grow as a manufacturing method, standardization of feedstock processing conditions and additives would enable more reliable and repeatable printed objects and would better assist designers in effectively implementing AM methods.
