Quizartinib (QUIZ) with decitabine (DAC) and venetoclax (VEN) is active in patients (pts) with FLT3-ITD mutated acute myeloid leukemia (AML): A phase I/II clinical trial.
Musa Yilmaz,Muharrem Muftuoglu,Hagop M. Kantarjian,Courtney D. DiNardo,Tapan M. Kadia,Marina Konopleva,Gautam Borthakur,Naveen Pemmaraju,Nicholas J. Short,Yesid Alvarado-Valero,Abhishek Maiti,Lucia Masarova,Guillermo Montalban-Bravo,Sanam Loghavi,Steven M. Kornblau,Elias Jabbour,Guillermo Garcia-Manero,Farhad Ravandi,Michael Andreeff,Naval Daver +19 more
TL;DR: DAC + VEN + QUIZ is active in R/R FLT3-ITD mutated AML pts, with CRc rates of 78% and the median OS of 7.6 months, Interestingly, RAS/MAPK mutations but not emergent TKD mutations were associated with primary and secondary resistance to the triplet.

A dynamic 3‐factor survival model for acute myeloid leukemia that accounts for response to induction chemotherapy
Ayalew Tefferi,Naseema Gangat,Aref Al-Kali,Hassan B. Alkhateeb,Mithun Vinod Shah,Mrinal M. Patnaik,Michael Elliott,William J. Hogan,Mark R. Litzow,C. Christopher Hook,Abhishek A. Mangaonkar,David S. Viswanatha,Dong Chen,Animesh Pardanani,Rhett P. Ketterling,Courtney D. DiNardo,Tapan M. Kadia,Farhad Ravandi,Koji Sasaki,Kebede H. Begna +19 more
TL;DR: The proposed 3‐factor survival model offers a novel prototype that is amenable to further enhancement by molecular information and was validated in an external cohort of 1032 intensively‐treated AML patients.

Association of smoking with poor risk ELN 2017, cytogenetics/molecular profile, and survival outcomes in acute myeloid leukemia.
Mansour Alfayez,Iman Abou Dalle,Guillaume Richard-Carpentier,Daewoo Pak,Jing Ning,Sherry Pierce,Koji Sasaki,Kiran Naqvi,Naval Daver,Jorge E. Cortes,Farhad Ravandi,Naveen Pemmaraju,Alessandra Ferrajoli,Guillermo Garcia-Manero,Marina Konopleva,Gautam Borthakur,Hagop M. Kantarjian,Tapan M. Kadia,Courtney D. DiNardo +18 more
TL;DR: This data indicates that smoking increases the relative risk of AML by 40% and 25% in active and former smokers, respectively, compared with non-smokers compared with active and current smokers.

Synergistic Pre-Clinical Activity of Targeted Inhibition of KDM1A and BET Proteins Against Human AML Blast Progenitor Cells
Warren Fiskus,Christopher P. Mill,Raffaella Soldi,Roulan Han,Dyana T. Saenz,Agnieszka J Nowak,Baohua Sun,David N. Saenz,Courtney D. DiNardo,Gautam Borthakur,Tapan M. Kadia,Wendy Jin,Thurika Ganesh,Vaishnavi Rao,Joseph D. Khoury,Sunil Sharma,Kapil N. Bhalla +16 more
TL;DR: It is determined that tet-inducible shRNA to KDM1A depleted protein levels of K DM1A, repressed c-Myc, but de-repressed p21, CD11b, CD86 and CEBPα, thereby inhibiting colony growth and modestly inducing lethality in genetically diverse cultured AML cell lines.
