In this review, we highlight the use of organic photoredox catalysts in a myriad of synthetic transformations with a range of applications. This overview is arranged by catalyst class where the photophysics and electrochemical characteristics of each is discussed to underscore the differences and advantages to each type of single electron redox agent. We highlight both net reductive and oxidative as well as redox neutral transformations that can be accomplished using purely organic photoredox-active catalysts. An overview of the basic photophysics and electron transfer theory is presented in order to provide a comprehensive guide for employing this class of catalysts in photoredox manifolds.

Fluorine in pharmaceutical industry: fluorine-containing drugs introduced to the market in the last decade (2001-2011).

Multicomponent reactions (MCRs) are fundamentally different from two-component reactions in several aspects. Among the MCRs, those with isocyanides have developed into popular organic-chemical reactions in the pharmaceutical industry for the preparation of compound libraries of low-molecular druglike compounds. With a small set of starting materials, very large libraries can be built up within a short time, which can then be used for research on medicinal substances. Due to the intensive research of the last few years, many new backbone types have become accessible. MCRs are also increasingly being employed in the total synthesis of natural products. MCRs and especially MCRs with isocyanides offer many opportunities to attain new reactions and basic structures. However, this requires that the chemist learns the “language” of MCRs, something that this review wishes to stimulate.

Reaction of 2,3-dihydrofuran with aryl triflate (1) in the presence of a base and a palladium catalyst, generated in situ from Pd(OAc)2 and (R)-BINAP, gave (R)-2-aryl-2,3-dihydrofuran (2) and a small amount of (S)-2-aryl-2,5-dihydrofuran (3). The enantiomeric purity of major product 2 was strongly affected by the base. 1,8-Bis(dimethylamino)naphthalene (proton sponge) as a highly basic and sterically demanding amine gave (R)-2 in high enantioselectivities ( > 96-87% ee) for a variety of aryl triflates.

Palladium-Catalyzed Asymmetric Arylation of 2,3-Dihydrofuran: 1,8-Bis( dimethylamino)naphthalene as an Efficient Base.

Advances in the palladium-catalyzed asymmetric hydrosilylation of 1,3-dienes are presented according to substrate types and chiral monophosphine ligands. Chiral monodentate phosphine ligands with a binaphthyl moiety have been proven to be the most efficient ligands for cyclic 1,3-dienes, and planar chiral ferrocenylmonophosphine ligands with two ferrocenyl moieties for linear 1,3-dienes. The ferrocenylmonophosphine ligands have expanded the substrate scope to 1,3-enynes in the asymmetric hydrosilylation. Palladium-catalyzed asymmetric hydrosilylation of 1,3-dienes and 1,3-enynes leads to the stereoselective synthesis of allylsilanes and allenylsilanes, respectively.
