In addition to the often-cited advantages of using Arabidopsis thaliana as a model system in plant biological research (1), Arabidopsis has many additional characteristics that make it an attractive experimental organism for studying lea d (Pb) accumulation and tolerance in plants. These include its fortuitous familial relationship to many known metal hyperaccumulators (Brassicaceae), as well as similar Pbaccumulation patterns to most other plants. Using nutrient-agar plates, hydroponic culture, and Pb-contaminated soils as growth media, we found significant variation in Arabidopsis thaliana ecotypes in accumulation and tolerance of Pb. In addition, we have found that Pb accumulation is not obligatorily linked with Pb tolerance, suggesti ng that different genetic factors control these two processes. We also screened ethyl methanesulfonate-mutagenized M2 populations and identified several Pb-accumulating mutants. Current characterization of these mutants indicates that their phenotypes are likely due to alteration of general metal ion uptake or translocation processes since these mutants also accumulate many other metals in shoots. We expect that further characterization of the ecotypes and mutants will shed light on the basic genetic and physiological underpinnings of plant-based Pb remediation. 7. Bacteria and plants might be used to enhance remediation of soil pollutants in situ. This study assessed the potential of bacteria (12 isolates), plants (16 forage grasses), and plant-bacteria associations (selected pairings) to remediate 2-chlorobenzoic acid (2CBA)-contaminated soil. Initially, grass viability was assessed in 2CBA-contaminated soil. Soil was contaminated wi th 2CBA, forage grasses were grown under growth chamber conditions for 42 or 60 d, and the 2CBA concentration in soil was determined by gas chromatography. Only five of 16 forage grasses grew in 2CBA-treated (816 mg/kg) soil. Growth of Bromus inermis had no effect on 2CBA concentration, whereas Agropyron intermedium, B. biebersteinii, A. riparum, and Elymus dauricus decreased 2CBA relative to nonplanted control soil by 32 to 42%. Phytoremediation : 植物による環境/土壌浄化

The term “phytotechnologies” refers to the application of science and engineering to provide solutions involving plants, including phytoremediation options using plants and associated microbes to remediate environmental compartments contaminated by trace elements (TE) and organic xenobiotics (OX). An extended knowledge of the uptake, translocation, storage, and detoxification mechanisms in plants, of the interactions with microorganisms, and of the use of “omic” technologies (functional genomics, proteomics, and metabolomics), combined with genetic analysis and plant improvement, is essential to understand the fate of contaminants in plants and food, nonfood and technical crops. The integration of physicochemical and biological understanding allows the optimization of these properties of plants, making phytotechnologies more economically and socially attractive, decreasing the level and transfer of contaminants along the food chain and augmenting the content of essential minerals in food crops. This review will disseminate experience gained between 2004 and 2009 by three working groups of COST Action 859 on the uptake, detoxification, and sequestration of pollutants by plants and consequences for food safety. Gaps between scientific approaches and lack of understanding are examined to suggest further research and to clarify the current state-of-the-art for potential end-users of such green options. Phytotechnologies potentially offer efficient and environmentally friendly solutions for cleanup of contaminated soil and water, improvement of food safety, carbon sequestration, and development of renewable energy sources, all of which contribute to sustainable land use management. Information has been gained at more realistic exposure levels mainly on Cd, Zn, Ni, As, polycyclic aromatic hydrocarbons, and herbicides with less on other contaminants. A main goal is a better understanding, at the physiological, biochemical, and molecular levels, of mechanisms and their regulation related to uptake–exclusion, apoplastic barriers, xylem loading, efflux–influx of contaminants, root-to-shoot transfer, concentration and chemical speciation in xylem/phloem, storage, detoxification, and stress tolerance for plants and associated microbes exposed to contaminants (TE and OX). All remain insufficiently understood especially in the case of multiple-element and mixed-mode pollution. Research must extend from model species to plants of economic importance and include interactions between plants and microorganisms. It remains a major challenge to create, develop, and scale up phytotechnologies to market level and to successfully deploy these to ameliorate the environment and human health.

Assessment of successful experiments and limitations of phytotechnologies: contaminant uptake, detoxification and sequestration, and consequences for food safety

Abstr ACt : The differences in Pb and Cd accumulation capabilities of Salix miyabeana, S. viminalis, S. × blanda, and S. elbursensis derived from willows planted in 1997 in coal-ash settling localities in the Krusne hory Mts.,the Czech Republic, were determined. Plantlets micropropagated by organogenesis were grown in sterile hydroponic media supplemented with 0.5mM Pb(NO 3 ) 2 or 0.5mM Cd(NO 3 ) 2 · 4H 2 O. The samples of roots and aboveground parts were collected after 48, 72, 96, and 168 h of cultivation. Generally, substantially higher concentrations of accumulated Pb and Cd were identified in roots than in aboveground parts of all willow clones, even if clonal differences in their ac cumulation were detected. The results sufficiently confirmed the clonal differences in the uptake and translocation of heavy metals in the above-mentioned willows.

Alkalisation of soil by dust pollution from a cement plant was assumed to be the principal cause of changes in heavy metal uptake and allocation between hybrid aspen (Populus tremula × Populus tremuloides Michx.) compartments. Emission of over 40 years of alkaline dust (pH 12.3–12.6) into the atmosphere had resulted in an increase of pH and an elevated concentration of total heavy metals in the upper layer of the soil (0–30 cm), which is considerable even 14 years after dust pollution has stopped. The accumulation and allocation of heavy metals in stem, shoot and leaves varied between themselves and between the trees from polluted and unpolluted plantations depending more on the mobility of elements and pH than element concentrations in the alkaline soil. High levels of heavy metals in the soil do not mean similar concentrations and ratios in plants growing in contaminated soil.

Heavy Metals Uptake and Accumulation by the Hybrid Aspen in Alkalised Soil

In this investigation we focus on the concentration of elements in Viscum album and its host (Sorbus aucuparia) as bioindicators of urban pollution. These broadly widespread species, very common in polluted areas may provide important information to monitor environmental quality throughout the year, especially for V. album. Cd, Co, Cr, Cu, Fe, Li, Mg, Mn, Ni, Pb and Zn concentrations were measured in the leaves and soil of the tree S. aucuparia as well as in V. album, a semi-parasite living on this tree species. The tree and the semi-parasite were studied in the urban environment of Olawa (SW Poland). This area was selected because of the influence of a zinc smelter on the level of metal pollution of soil and plants and to compare the ability of S. aucuparia and V. album to accumulate metals. V. album appeared to be a better bioaccumulator of Cd, Ni, Pb and Zn and a weaker accumulator of Co than S. aucuparia in less polluted sites of Olawa. Micropropagated plantlets derived from selected clones of the hybrid aspen ( Populus tremula × Populus tremuloides) and the rowan-tree ( Sorbus aucuparia L.) were used to determine the comparative study of uptake of the toxic, heavy metals Cd, Pb and the essential metal Mn. Samples of roots and aboveground parts (hypocotyl-derived tissues, leaves and stems) were taken from the plantlets grown for 24, 48, 96, or 168 hrs under aseptic conditions, in hydroponics with the toxic heavy metal and the essential metal salts. The concentration and distribution of the accu - mulated metals were determined using the ICP-OES method. The differences in the uptake capacity of hybrid aspen and rowan-tree clones for Cd, Pb and Mn were identified. Generally, the amounts of accumulated Cd and particularly Pb were much higher in the roots of both hybrid aspen and rowan-tree clones, than in their shoots, at all sample times. Conversely, the amounts of accumulated Mn were significantly lower than Cd and Pb in all plant parts of the hybrid aspen and rowan-tree samples. Patterns of Mn uptake were similar in the above-mentioned tissues of both clones, at all sample times. We concluded that the two clones of hybrid aspen and rowan-tree, lacking auxiliary soil microbiota, can accumulate large amounts of the toxic heavy metals Cd (800-1,500 mg/kg) and Pb (5,000-13,000 mg/kg) in roots and about 100 mg/kg of Cd was determined in aboveground part of hybrid aspen.
