Abstract Vermicompost is a nutrient-rich biological fertilizer with a variety of microorganisms that are thought to be significant in increasing the growth and yield of various field crops, vegetables, flowering plants, and fruit trees. Vermicomposting has attracted a lot of attention as an extensive approach for restoring the environment, producing nutrient-rich bio-fertilizers, and growing crops in a sustainable manner. It isploying earthworms to break down complex organic waste into simpler materials that could be taken up by plants. Vermicomposting yields a valuable byproduct called vermi-wash, which enhances crop resistance against diseases, stimulates seed germination, and improves overall plant vigor. This research paper sheds light on the significance of vermicomposting as a sustainable waste management solution and an eco-friendly means to enhance agricultural productivity. It emphasizes the importance of understanding the composition and quality of vermicompost, the materials used in the process, the vermicomposting procedure, and the subsequent effects on crop performance. Through the adoption of vermicomposting practices, agricultural systems can become more environmentally friendly, economically viable, and resilient for a sustainable future. 

Vermicomposting: a holistic approach for sustainable crop production, nutrient-rich bio fertilizer, and environmental restoration

The cultivation of bitter gourd (Momordica charantia L.) in Nepal faces significant challenges, resulting in diminished yields compared with other regions. The pivotal issue is optimizing fertilizer management practices to enhance bitter gourd growth and yield. In April 2022, a field experiment was conducted in Kapilvastu, Nepal to investigate the efficacy of organic and inorganic fertilizers on the growth and yield of bitter gourd. The study included five treatments: the recommended dose of nitrogen, phosphorus, and potassium (NPK) fertilizer (111.66:54.56:35.36 NPK kg ha−1, and 29.49 t ha−1 farmyard manure), 100 % poultry manure, 100 % farmyard manure, 50 % NPK + 50 % poultry manure, and 50 % NPK + 50 % farmyard manure. We hypothesized that a blend of 50 % organic and 50 % inorganic manure would significantly enhance bitter gourd performance. These treatments were evaluated in a randomized complete block design (RCBD) with four replications and analyzed using R-Studio. The results showed that 50 % NPK +50 % poultry manure increased growth parameters, such as plant height (380 cm), branch count plant−1 (22), node count plant−1 (34.75), female flower count (50.50), fruit length plant−1 (24.75 cm), and fruit diameter (6.53 cm), and decreased male flower count (132.25) and days to first male (33.75) and female (36.25) flowering. The application of 50 % NPK and 50 % poultry manure increased individual fruit weight (274.50 g) and yield-attributing traits, such as fruit count (17.25), fruit yield (55.56 t ha−1), net return (7944$), and benefit-cost ratio (3.14). A synergistic blend of 50 % NPK and 50 % poultry manure can serve as a viable and effective nutrient source for promoting growth and maximizing bitter gourd yield. Although these results are promising, further validation and extension of these positive findings are required on a larger scale in diverse ecological regions. 

Efficacy of different organic and inorganic nutrient sources on the growth and yield of bitter gourd (Momordica charantia L.)

Root-knot nematodes are among the world’s most damaging endoparasitic sedentary nematodes, especially, Meloidogyne incognita that infects a wide range of plant hosts. The activity of different antagonistic bacteria was studied for the low-cost and eco-friendly management of M. incognita on eggplant. Twenty-five isolates were isolated from rhizosphere soils infected with nematodes. Of these, 6 isolates displayed the highest activity, demonstrating 100% mortality of J2 nematodes under laboratory conditions. Partial sequencing of 16S rRNA gene and phylogenetic analysis was used to identify the selected isolates and they were found to be Paenibacillus amylolyticus, Brevibacillus agri, Gluconobacter frateurii, Beijerinckia mobilis, Achromobacter aloeverae, and Pseudomonas stutzeri. The abilities of the selected isolates to produce hydrogen cyanide, siderophores, chitinase, protease, indole acetic acid, and to dissolve phosphorus were also detected. The results of the greenhouse experiment indicated that all the tested bacteria had a greatly significant effectiveness for suppressing root-knot nematode M. incognita. Application of all the rhizosphere bacteria and their combinations reduced the number of galls, number of juveniles, egg-masses, eggs, females, and total final population. All bio-agent treatments succeeded in improving the plant growth parameters and increased the microbial density in eggplant rhizosphere.

Isolation and characterization of uricase-producing bacteria from soil

Wheat (Triticum aestivum L.), a staple crop in Nepal, has a rich history of cultivation. Despite increasing demand due to population growth and agricultural development efforts, wheat production faces persistent challenges that hinder its expansion and profitability. This research, conducted from February to July 2022 in West Nawalpur district, Nepal, aims to assess the productivity, profitability, and economics, including benefit-cost analysis, of wheat in West Nawalparasi. A total of 100 wheat farmers were selected and the data obtained were analyzed using Microsoft Excel and Statistical Package for the Social Sciences (SPSS). The study reveals a positive gross margin (NRs 1113.09) and a benefit-cost ratio (BCR) of 1.6 per 0.3 hectare, indicating the profitability of wheat cultivation. Notably, most farmers procure agricultural inputs such as seeds and fertilizers from local agro-dealers within Nepal, highlighting a preference for domestic sources over imports from India. For irrigation, more than half of the population relies on pumps that draw from open water sources (59%) and use mechanization equipment such as tractors and combine harvesters for field activities. However, the study reveals the complexity of the challenges faced by wheat farmers. Chief among these are the unavailability of adequate irrigation facilities, untimely and inadequate access to agricultural inputs, disease and pest incidence, lack of training and extension services, and lack of mechanization and skilled labor. These constraints, as highlighted by the farmers, affect both productivity and profitability, highlighting the urgent need for initiatives to reduce production costs and improve wheat production in West Nawalparasi.

Assessment of Productivity, Profit, and Problems Associated with Wheat (Triticum aestivum L.) Production in West Nawalparasi, Nepal

Exclusive use of organic manure in bitter gourd cultivation slows nutrient release, affecting root growth, while inefficient application of fertilizers during transplanting and development stages leads to nutrient losses and increased production costs. The research aimed to evaluate the efficiency of different combinations of urea and vermicompost in bitter gourd production and address the challenges associated with the use of only organic manure. The field experiment conducted in Tulsipur, Dang, utilized a six-treatment randomized complete block design with four replications. The treatments consisted of varying proportions of the recommended dose of urea (16 g plant-1) and vermicompost (280 g plant-1). Among the treatments, T3 (50% urea and 50% vermicompost) exhibited the most prevalent diameter (3.854 cm), length (16.32 cm), fruit count (1.391), weight plant-1 (189.2 g), and weight plot-1 (1848 g) while T4 (25% urea and 75% vermicompost) showed the quickest first flowering (30.37 days) and fruiting (33.75 days). T2 (75% urea and 25% vermicompost) recorded the highest vine length (222.5 cm). Statistically significant differences were observed in most recorded characteristics for T3. The results highlight the slow-release nutrient benefits and soil improvement properties of vermicompost, complemented by the quick-release nutrient attributes of urea. The traits with the lowest yield were observed in the control group (T6). A 50% combination of recommended urea dose and vermicompost is recommended for bitter gourd cultivation, leading to improved growth, yield, and quality, underscoring the benefits of organic fertilizer in this situation. The study's findings contribute to the scientific understanding of optimizing fertilization practices in bitter gourd production, minimizing production costs, reducing nutrient losses, and environmental pollution. 

Effectiveness of different combinations of urea and vermicompost on yield of bitter gourd (Momordicacharantia)

This study was conducted to screen for strains with proteolytic activity and to examine the effects of pH, temperature and incubation time on the activity of isolated crude enzymes. Bacterial isolates was isolated from the sewage site present in different river banks of Kathmandu valley. Isolated samples were screened for production of protease. One of the isolates was found positive and designated as SAG-P2. Based on the biochemical characterization and staining organism was identified as Corynebacterium sp. This isolates was taken for further studies. The optimum pH for enzyme activity was found to be at pH 6.6 and the optimum temperature for the activity was found to be at 50 oC

Screening and optimization of extra cellular protease from bacteria isolated from sewage.

Given the alarming incidence of antibiotic resistance in bacteria of medical importance and multiple side effects associated with the modern day chemotherapeutics, there is a constant need for new and effective therapeutic agents that could be easily extracted from our daily used Nepalese culinary. To study the antibacterial and antioxidant activity of common spices, locally available  Clove (Eugenia caryophyllus), Cinnamon (Cinnamomum zylancium), Cumin (Cumin cyminum) and Timur (Zanthoxylum alatum) were subjected to cold extraction using ethanol and was assayed through agar well diffusion method and DPPH radical scavenging activity for different concentration gradient(100 to 500 µg). Zanthoxylum and Eugenia have showed potent antimicrobial activity against Proteus vulgaris and Pseudomonas aeruginosa followed by Cinnamomum and Cumin against Pseudomonas aeruginosa. All the extracts showed effective antimicrobial activity against gram positive and gram negative bacteria, however Escherichia coli remains ineffective towards any of the concentration of spices. DPPH radical scavenging activity showed effective antioxidant activity of the spices in the following order: Eugenia (93.84%) > Cumin (90.4%), Zanthoxylum (88.73%) > Cinnamomum (87.23%). Hence, our present study demonstrated that the ethanolic extract of different spices, Eugenia being the most effective, possess potent antibacterial and antioxidant activity and can be further analyzed for antimicrobial therapeutics and pharmacological evaluation.

Study of antibacterial and antioxidant activities of four common Nepalese kitchen spices

PO-1256 Multi-domain automated lung segmentation for inflammatory lung disease (ILD) detection

Patients with interstitial lung disease (ILD) treated with thoracic radiotherapy (RT) are at greater risk of pulmonary toxicity. Automatic universal screening for ILD allows radiation oncologists (ROs) to risk stratify patients and implement necessary modifications to their respiratory monitoring or treatment. Automatic screening however may affect RO workload and so it is imperative to assess the clinical acceptability of this tool.We have developed a machine learning algorithm to identify patients who are at high risk of having ILD based on RT planning computed tomography (CT) images. A quality improvement (QI) project was initiated to test feasibility and acceptability of the machine learning algorithm. If positive, the results of the machine learning algorithm were made available to ROs via structured electronic reporting. ROs were prompted to review the patient and consider expert radiologist consultation if thought appropriate. All electronic surveys and qualitative comments were summarized to describe clinical acceptability. Expert radiologist established gold standard ILD status of all patients on the study. A formal review of RO feedback was collected for all screen-positive, true-positive cases.Two hundred forty cases were screened of which 45 were flagged as AI-ILD positive and the responsible RO notified. Of these 45 screen-positive cases, all continued on to RT except for 3 patients with tumor progression. From these 45, 24 surveys were completed, 21 had no prior suspicion of ILD. There were 7 true-positives, of which 1 had a survey response. Based on the survey responses, 88% of cases underwent review by the responsible RO. In 16 cases this automatic notification prompted case consultation with an expert radiologist. Expert review was performed from 10 minutes up to 53 hours after the email prompt to the radiologist, with median response time of 1.5 hours. In the 7 screen-positive, true-positive cases, only 2 were not previously known to the responsible RO. In the two cases where true-positive ILD status was previously unknown, one was a mild case of ILD and the other had previously received thoracic RT at this institution without ILD being identified, in both cases the ROs were grateful that this diagnosis was identified prior to treatment. RO confidence in the machine learning prediction was moderate due to the high proportion of false positives.Based on available survey results, more than 75% of the screen-positive cases were reviewed by the responsible RO and two-thirds of these involved expert radiology input. RO feedback was generally positive and this tool was rated as a net benefit despite the high rate of false-positives and the need for clarification.
