Since the initial suggestion of using insect hormones or their mimics as the third-generation insecticides, great strides have been made in the discovery of three classes of such insect growth-disrupting insecticides We propose the use of the term “insect growth disruptors” instead of the mis-named term “insect growth regulators” We review the commercial insecticides that mimic the mode of action of the insect growth and development hormones, the sesquiterpenoid juvenile hormones and the steroidal 20-hydroxyecdysone and the third class of insecticides that disrupt growth and development by interfering with chitin synthesis in target insect pests In this chapter, the emphasis is on the application of these insecticides to control insect pests and development of resistance Since the non-steroidal ecdysone agonist insecticides have been extensively reviewed in other chapters of this volume and elsewhere, only salient features of these novel insecticides are discussed

Compositions comprising a biological control agent and an insecticide

Benzylideneacetone (BZA) is a metabolite of gram-negative entomopathogenic bacterium Xenorhabdus nematophila, and it acts as an enzyme inhibitor against phospholipase A2 (PLA2). PLA2 catalyzes a committed biosynthetic step of eicosanoids, which mediate insect immune reactions to infection by microbial pathogens. This study tested a hypothesis that a putative immunosuppressive activity of BZA may enhance virulence of Bacillus thuringiensis against the fifth instars of Spodoptera exigua (Hubner) (Lepidoptera: Noctuidae). In in vitro conditions, BZA significantly inhibited hemocyte microaggregation induced by B. thuringiensis and impaired hemocyte-spreading behavior of S. exigua in a dose-dependent manner. Oral administration of BZA gave similar immunosuppressive effect on the hemocytes of the fifth instars. Although BZA itself did not possess any insecticidal activity on oral administration, when BZA was treated in a mixture with a low dose of B. thuringiensis spp. aizawai to fifth instars, the bacterial virulence was significantly enhanced. BZA also enhanced virulence of B. thuringiensis spp. kurstaki, which alone was of limited effectiveness against S. exigua. This study suggests that an immunosuppression by BZA is positively linked to potentiation of B. thuringiensis.

Benzylideneacetone, an Immunosuppressant, Enhances Virulence of Bacillus thuringiensis Against Beet Armyworm (Lepidoptera: Noctuidae)

Hemocytes are crucial players of the mosquito immune system and critically affect transmission of pathogens including malaria parasites. We and others discovered previously that a blood meal is a major immune stimulus for mosquito hemocytes. To determine whether these blood meal-induced hemocyte changes in Anopheles gambiae constitute steps in cell differentiation or demonstrate transient cell activation, we analyzed the temporal pattern of these changes over the first three days post blood meal (dpbm). Flow cytometry and immunofluorescence analyses revealed a global shift of the entire hemocyte population, peaking at 1 dpbm. All hemocyte activation markers returned to pre-blood meal baseline levels within the following 24-48 h. Our observations are consistant with An. gambiae hemocytes undergoing transient activation rather than terminal differentiation upon blood feeding. Interestingly, the temporal pattern followed the gonotrophic cycle of the mosquito, strongly suggesting hormonal control of mosquito hemocyte activation and deactivation.
