Breeding for disease resistant varieties remains very effective and economical in controlling the bacterial leaf blight (BLB) of rice. Breeders have played a major role in developing resistant rice varieties against the BLB infection which has been adjudged to be a major disease causing significant yield reduction in rice. It would be difficult to select rice crops with multiple genes of resistance using the conventional approach alone. This is due to masking effect of genes including epistasis. In addition, conventional breeding takes a lot of time before a gene of interest can be introgressed. Linkage drag is also a major challenge in conventional approach. Molecular breeding involving markers has facilitated the characterization and introgression of BLB disease resistance genes. Biotechnology has brought another innovation in form of genetic engineering (transgenesis) of rice. Although, molecular breeding cannot be taken as a substitute for conventional breeding, molecular approach for combating BLB disease in rice is worthwhile given the demand for increased production of rice in a fast growing population of our society. This present article highlights the recent progress from conventional to molecular approach in breeding for BLB disease resistant rice varieties.

Bacterial leaf blight resistance in rice: a review of conventional breeding to molecular approach.

This study was carried out to evaluate genetic diversity, phenolic compound composition, and biological activity of Setaria italica L. collected from different parts of South Korea. Antioxidant potential of seeds was estimated by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, and antimicrobial activity was determined by evaluating the minimum inhibitory concentration (MIC). Eight phenolic acids and 3 flavonoids were identified and quantified, among which myricetin and salicylic acid were the most dominant phytochemical compounds detected in the majority of accessions. The antioxidant potential of the leaf extracts of all the accessions was significantly higher (ranging from 32.33 ± 1.53 µg mL−1 in SI-03 to 87.87 ± 1.63 µg mL−1) in SI-10 than that of the root, stem, or seeds. Among the 15 accessions, methanolic extracts of the SI-15 accession strongly suppressed the growth of Escherichia coli (250 µg mL−1). Accessions SI-14 and SI-15 showed positive antimicrobial activity against all gram-positive bacteria. Interestingly, extracts of all accessions were more sensitive towards E. coli and Staphylococcus aureus, with MICs ranging from 250 to 1000 µg mL−1. Three phenolic acids, namely chlorogenic acid, catechin, caffeic acid, naringin, hesperetin, and myricetin, were found to be moderately positively correlated with antioxidant activities. A wide range of diversity was observed in morphological traits, namely plant height (99.33 to 201.33 cm), culm length (67.10 to 160.00 cm), spike length (12.80 to 24.00 cm), 1000 seeds weight 1.44 to 2.91 g), bloom beginning (93.67 to 128.00 days), and full bloom (99.67 to 135 days). A dendogram generated from unweighted pair group method with arithmetic mean clustering (UPGMA) cluster analysis based on the morphological traits and inter simple sequence repeat (ISSR) marker data revealed three major groups. However, no clear correlation between these two different approaches was found. The average Shannon’s information index value (I) was 0.492, and it ranged from 0 to 0.693. The average expected heterozygosity (He) was 0.335, and it ranged from 0 to 0.499. The substantial variation in the morphological traits, bioactive properties, and genetic diversity among the accessions may provide useful information for breeding programs attempting to obtain S. italica with improved bioactive properties.

Assessment of Diversity in the Accessions of Setaria italica L. Based on Phytochemical and Morphological Traits and ISSR Markers.

Foxtail millet is an indigenous crop known for its rich nutritive value, drought tolerance ability and low input requirements. Though it is grown in marginal condition, it can give the best production with the proper nutrient management for its cultivation. In this respect, an experiment was conducted in Agronomy farm of Lamjung Campus, Sundarbazar during March-June 2017. The main objective of the experiment was to explore the performance of local foxtail millet under different level of nutrients. The experiment was carried out in Randomized Complete Block Design with three replications and seven treatments viz. FYM 6 t/ha, FYM 6 t/ha+60:30:20 kg NPK/ha, 60:30:20 kg NPK/ha, FYM 6t/ha+30:20kg PK/ha, FYM 6t/ha+60:30 kg NK/ha, FYM 6 t/ha+60:30 kg NP/ha and Control (no fertilizers). Results revealed that highest grain yield (2.47t/ha) (152% higher than control), was obtained from FYM 6 t/ha+60:30:20 kg NPK/ha which was followed by 60:30:20 kg NPK/ha (2.45 t/ha) and were statistically at par with each other. The highest grain yield in FYM 6 t/ha+60:30:20 kg NPK/ha was supported by higher no. of grains per panicle (2870), more test weight (1.79 gm) and more harvest index (19.3%). However, the straw yield (12.6t/ha), biological yield (15.02t/ha) along with B: C ratio was found slightly higher in 60:30:20 kg NPK/ha. The growth characters viz. plant height, flag leaf area and panicle length of all the treatments were significantly higher than Control (T7). The treatment having balanced plant nutrients (T2) produced significantly higher yield and can be recommended to mid-hill farmers for obtaining good yield.

Nurient Management Trial on Foxtail Millet at Sundarbazar, Lamjung

This experiment was conducted to genotypically and phenotypically select new rice lines pyramided with blast and bacterial leaf blight disease resistance genes after a marker-assisted backcross breeding programme. The inter-relationship among agro-morphological characteristics and their effect on yield was also studied. The polymorphic functional and microsatellite markers Xa21FR and pTA248 (Xa21), Xa13prom (xa13), RM21 (xa5), MP (Xa4), RM6836 (Pi2, Pi9 & Piz) and RM8225 (Piz) were first confirmed for the target genes. The selected markers were used for foreground selection of BC2F2 homozygous progenies with the target genes. Plants that had homozygous IRBB60 alleles for these markers were evaluated for their recovery of the recurrent parent genome. IRBB60 was used as the donor parent for bacterial blight resistance genes while Putra-1 served as a recipient/recurrent parent with background blast-resistance genes and high yield. After the foreground selection, 79 polymorphic simple sequence repeat markers identified from the marker polymorphism survey were used for marker-assisted background selection to determine the percentage recovery of the recurrent parent genome. In order to make a selection on a phenotypic basis, 14 agro-morphological traits were measured and recorded. The result obtained from the study showed that 16 lines received the seven resistance genes in sufficiently varied numbers and were selected. The distribution of yield per hectare showed that about 50% of the selected lines had yields as high as 5 t/ha and above. Some of the lines produced as high as 8.4 t/ha. These lines demonstrated the potential of recording uniform 8t/ha upon recombination at BC2F2. The study also indicated that the number of panicles per hill correlated strongly, significantly and positively with the number of tillers (r = 0.962 **), total grain weight per hill (r = 0.928 **) and yield per hectare (r = 0.927 **). It was concluded that the newly improved resistant lines which were selected have the capability to compete with Putra-1 in terms of its productivity and yield. The newly developed lines would be useful in future breeding programmes as donors for bacterial leaf blight and blast resistance genes. These lines are recommended for release to farmers in Malaysia and other rice-growing agro-ecologies for commercial cultivation.

Genotypic and Phenotypic Selection of Newly Improved Putra Rice and the Correlations among Quantitative Traits

Malnutrition, hunger, and rural poverty in Nepal are prevalent issues that have been exacerbated by the negative impacts of climate change on crop production. To create a more sustainable and resilient food system, it is crucial to explore alternative options beyond the small number of input-intensive crops that the country currently relies on. In this study, we conducted a comprehensive investigation of six underutilized food crops—amaranth, buckwheat, finger millet, foxtail millet, naked barley, and proso millet—in four mountainous districts of the Karnali province, Nepal. Using several approaches that included a literature review, key informant surveys, focus group discussions, and direct observations, we explored the production and utilization aspects of these crops, as well as their potential for improving nutrition and food security. Our findings indicate that these underutilized crops continue to be a vital element of the diets of people residing in Karnali and play an important role in their livelihoods. This study also highlights the diversity and potential of these underutilized crop landraces, and their significance in addressing nutrition and food insecurity, as well as in improving livelihoods in regions with marginal lands and inadequate food supply. Thus, promoting the cultivation and consumption of underutilized crops can create a more sustainable and diverse food system that is resilient to climate change. However, further investigation is needed to fully understand the socioeconomic importance and other potential benefits of these crops.

Potential of Underutilized Grain Crops in the Western Mountains of Nepal for Food and Nutrient Security

Foxtail millet ( Setaria italica (L) P Beauv) falls on the category of underutilized crops in Nepal and mainly cultivated in Karnali region of the country. It is hardy crop and considered as one of the potential crops for future food security with respect to climate change. Five accessions of Nepalese foxtail millet were purposefully selected for evaluation of the agro-morphological characteristics. Foxtail landraces from Dolpa, Mugu, Bajura, Bajhang and Lamjung districts of Nepal were evaluated at Khumaltar, Lalitpur, Nepal during 2010. The plot size was 1m 2 and there were five samples. Days to heading and days to maturity varied from 33-56 and 59 to 87 days after germination respectively. Similarly, flag leaf length/breadth ratio, flag leaf sheath length, ligule length, peduncle length, peduncle exertion and plant height varied from 3.84-10.90, 5.47-9.84 cm, 0.1-0.2 mm, 10-22.57 cm, 2.7-13.58 cm and 41.67-120 cm, respectively. Fruit and apiculus color varied from straw to black. All accessions were actively growing with very slight lodging. Similarly, the thousand grain weight varies from 1.064 g to 2.172 g. This variation is useful in foxtail millet breeding program. Similarly, the significant correlation between thousand kernel weight and total basal tiller (r=-0.975) showed that foxtail millet lines with low tillering ability is better for yield enhancement. Variation on agro-morphological traits in Nepalese foxtail millet (Setaria italica (L) P Beauv)

BACKGROUND
Wild relatives of wheat (Triticum spp.) harbor beneficial alleles for potential improvement and de novo domestication of selected genotypes with advantageous traits. We analyzed the nutrient composition in wild diploid and tetraploid wheats and their domesticated diploid, tetraploid and hexaploid relatives under field conditions in Germany and compared them with modern Triticum aestivum and Triticum durum cultivars. Grain iron (Fe) and zinc (Zn) concentrations, phytate:mineral molar ratios, grain protein content (GPC) and antioxidant activity were analyzed across 125 genotypes.


RESULTS
Grain Fe and Zn concentrations in wild wheats were 72 mg kg-1 and 59 mg kg-1 , respectively, with improved bioavailability indicated by Phytate:Fe and Phytate:Zn molar ratios (11.7 and 16.9, respectively) and GPC (231 g kg-1 ). By comparison, grain Fe and Zn concentrations in landrace taxa were 54 mg kg-1 and 55 mg kg-1 , respectively, with lower Phytate:Fe and Phytate:Zn molar ratios (15.1 and 17.5, respectively) and GPC (178 g kg-1 ). Average grain Fe accumulation in Triticum araraticum was 73 mg kg-1 , reaching 116 mg kg-1 , with high Fe bioavailability (Phyt:Fe: 11.7; minimum: 7.2). Wild wheats, landraces and modern cultivars showed no differences in antioxidant activity. Triticum zhukovskyi stood out with high grain micronutrient concentrations and favorable molar ratios. It was also the only taxon with elevated antioxidant activity.


CONCLUSION
Our results indicate alteration of grain quality during domestication. T. araraticum has promising genotypes with advantageous grain quality characteristics that could be selected for de novo domestication. Favorable nutritional traits in the GGAA wheat lineage (T. araraticum and T. zhukovskyi) hold promise for improving grain quality traits. © 2024 The Authors. Grain quality traits within the wheat (Triticum spp.) genepool: prospects for improved nutrition through de novo domestication.

Bacterial Leaf Blight (BLB) is the most important yield limiting factor in Nepalese rice. BLB resistance rice varieties are highly demanding in the country. Breeding efforts for developing disease resistant depends on availability and use of resistant gene donors. Nepalese rice landraces could be the source of resistant gene. Therefore, ninety six Nepalese rice accessions were screened using eight Simple Sequence Repeats (SSR) markers and one Sequence Tagged Sites (STS) marker for presence and absence of BLB resistance gene. We have detected BLB resistance gene Xa-10 on five accessions, Xa-13 on six accessions, Xa-7 on 23 accessions, Xa-3 and Xa-4 on 52 accessions, Xa-5 on 25 accessions, Xa-8 on 30 rice accessions. No any rice accessions tested have Xa-21. Similarly, 17 rice accessions showed three and more than three BLB resistance genes. Presence of Xa-13 on susceptible check variety CNTRL-85033 confirmed that this resistant gene is not working in Nepalese rice field. Barley ( Hordeum vulagare L) is the major crop for the people living in the high hills and mountainous region of Nepal. Leaf rust (caused by Puccinia hordei ) is one of the major production threats for barley cultivation. A lot of variation can be observed on Nepalese barley accessions with respect to leaf rust resistance characteristics. Two hundred and forty one barley accessions were screened for leaf rust resistance characteristics on heading stage at Khumaltar, Lalitpur, Nepal. Among them, one hundred and nine Nepalese barley accessions showing promising for disease resistance were screened using six SSR markers linked to leaf rust resistance genes. Bonus and Local Jau was used as the resistant and susceptible check respectively. Leaf rust resistance genes Rph1, Rph2, Rph3, Rph7, QBLR-P and QTL on chromosome 5HS were detected on Nepalese barley accessions using respective SSR markers. Eight Nepalese barley accessions showed presence of three and more leaf rust resistant genes. The poor relationship between the field disease resistance and molecular markers linked with specific leaf rust resistance gene proved that Nepalese barley gene pool contains other leaf resistance genes.
