Sepsis is the main cause of death in patients suffering from serious illness. Yet, there is still no specific treatment for sepsis, and management relies on infection control. Cell membrane-coated nanoparticles (MNPs) are a new class of biomimetic nanoparticles based on covering the surface of synthetic nanoparticles (NPs) with natural cell membranes. They retain the physicochemical properties of synthetic nanomaterials and inherit the specific properties of cellular membranes, showing excellent biological compatibility, enhanced biointerfacing capabilities, capacity to hold cellular functions and characteristics, immunological escape, and longer half-life when in circulation. Additionally, they prevent the decomposition of the encapsulated drug and active targeting. Over the years, studies on MNPs have multiplied and a breakthrough has been achieved for cancer therapy. Nevertheless, the use of “bio”-related approaches is still rare for treating sepsis. Herein, we discussed current state-of-the-art on MNPs for the treatment of bacterial sepsis by combining the pathophysiology and therapeutic benefits of sepsis, i.e., pathogenic bacteria, bacteria-producing toxins, and inflammatory cytokines produced in the dysregulated inflammatory response associated with sepsis.

Exosome membrane-coated nanosystems: Exploring biomedical applications in cancer diagnosis and therapy

Metal-organic frameworks (MOFs) are a new class of porous crystalline materials composed of metal and organic material. MOFs have fascinating properties, such as fine tunability, large specific surface area, and high porosity. MOFs are widely used for environmental protection, biosensors, regenerative medicine, medical engineering, cell therapy, catalysts, and drug delivery. Recent studies have reported various significant properties of MOFs for biomedical applications, such as drug detection and delivery. In contrast, MOFs have limitations such as low stability and low specificity in binding to the target. MOF-based membranes improve the stability and specificity of conventional MOFs by increasing the surface area and developing the possibility of MOF-ligand binding, while conjugated membranes dramatically increase the area of active functional groups. This special property makes them attractive for drug and biosensor fabrication, as both the spreading and solubility components of the porosity can be changed. Asymmetric membranes are a structure with high potential in the biomedical field, due to the different characteristics on its two surfaces, the possibility of adjusting various properties such as the size of porosity, transfer rate and selectivity, and surface properties such as hydrophilicity and hydrophobicity. MOF assisted asymmetric membranes can provide a platform with different properties and characteristics in the biomedical field. The latest version of MOF materials/membranes has several potential applications, especially in medical engineering, cell therapy, drug delivery, and regenerative medicine, which will be discussed in this review, along with their advantages, disadvantages, and challenges.

Recent Advances in Metal-Organic Framework (MOF) Asymmetric Membranes/Composites for Biomedical Applications

Treatment of large bone fractures remains a challenge for orthopedists. Bone regeneration is a complex process that includes skeletal cells such as osteoblasts, osteoclasts, and immune cells to regulate bone formation and resorption. Osteoimmunology, studying this complicated process, has recently been used to develop biomaterials for advanced bone regeneration. Ideally, a biomaterial shall enable a timely switch from early stage inflammatory (to recruit osteogenic progenitor cells) to later-stage anti-inflammatory (to promote differentiation and terminal osteogenic mineralization and model the microstructure of bone tissue) in immune cells, especially the M1-to-M2 phenotype switch in macrophage populations, for bone regeneration. Nanoparticle (NP)-based advanced drug delivery systems can enable the controlled release of therapeutic reagents and the delivery of therapeutics into specific cell types, thereby benefiting bone regeneration through osteoimmunomodulation. In this review, we briefly describe the significance of osteoimmunology in bone regeneration, the advancement of NP-based approaches for bone regeneration, and the application of NPs in macrophage-targeting drug delivery for advanced osteoimmunomodulation.

Cell membrane cloaking technique is bioinspired nanotechnology that takes advantage of naturally derived design cues for surface modification of nanoparticles. Unlike modification with synthetic materials, cell membranes can replicate complex physicochemical properties and biomimetic functions of the parent cell source. This technique indeed has the potential to greatly augment existing nanotherapeutic platforms. Here, we provide a comprehensive overview of engineered cell membrane-based nanotherapeutics for targeted drug delivery and biomedical applications and discuss the challenges and opportunities of cell membrane cloaking techniques for clinical translation.
