Retrograde transport of TrkB-containing autophagosomes via the adaptor AP-2 mediates neuronal complexity and prevents neurodegeneration.

TL;DR: It is shown that in neurons, autophagosomes promote neuronal complexity and prevent neurodegeneration in vivo via retrograde transport of brain-derived neurotrophic factor (BDNF)-activated TrkB receptors via endocytic adaptor AP-2, an essential protein complex previously thought to function exclusively in clathrin-mediated endocytosis.
Autophagy lipidation machinery regulates axonal microtubule dynamics but is dispensable for survival of mammalian neurons.
A. Negrete-Hurtado,Melina Overhoff,Sujoy Bera,E. De Bruyckere,K. Schätzmüller,Min Jeong Kye,C. Qin,Michael Lammers,Vangelis Kondylis,Ines Neundorf,Natalia L. Kononenko +10 more
TL;DR: The results highlight a non-canonical role of ATG proteins in neurons and suggest that pharmacological activation of autophagy may not only promote the degradation of cytoplasmic material, but also impair axonal integrity via altering MT stability.
Compromised Hippocampal Neuroplasticity in the Interferon-α and Toll-like Receptor-3 Activation-Induced Mouse Depression Model
Eduardo H. Sanchez-Mendoza,Santiago Camblor-Perujo,Luiza Martins Nascentes-Melo,Egor Dzyubenko,Michael Fleischer,Tayana Silva de Carvalho,Linda-Isabell Schmitt,Markus Leo,Tim Hagenacker,Arne Herring,Kathy Keyvani,Sujoy Bera,Natalia L. Kononenko,Christoph Kleinschnitz,Dirk M. Hermann +14 more
TL;DR: Evidence is provided that IFN-α and poly(I:C) delivery leads to structural and functional alterations at the synapse indicating that compromised neuroplasticity may play an integral role in the pathogenesis of immune response-induced depression.
Isolation of CA1 nuclear enriched fractions from hippocampal slices to study activity-dependent nuclear import of synapto-nuclear messenger proteins.
PingAn Yuanxiang,Sujoy Bera,Anna Karpova,Michael R. Kreutz,Marina Mikhaylova,Marina Mikhaylova +5 more
TL;DR: An optimized workflow is described in order to obtain sufficient quantity of nuclear enriched fraction of sufficient purity from the CA1 region of acute hippocampal slices from rat brain to show that the ERK1/2 phosphorylated form of the synapto-nuclear protein messenger Jacob actively translocates to the nucleus upon induction of LTP and can be detected in anuclear enriched fraction from CA1 neurons.
Process strategies for enhancing recombinant streptokinase production in Lactococcus lactis cultures using P170 expression system
TL;DR: It is seen that suppression of ATR has the most predominant effect on enhancing recombinant protein productivity in fed-batch processes, due to a synergistic combination of increased glucose-feed rates and addition of complex-nutrients to the feed.
