Small molecule obatoclax (GX15-070) antagonizes MCL-1 and overcomes MCL-1-mediated resistance to apoptosis
Mai Nguyen,Richard C. Marcellus,Anne Roulston,Mark F. Watson,Lucile Serfass,S.R. Murthy Madiraju,Daniel Goulet,Jean Viallet,Laurent Bélec,Xavier Billot,Stephane Acoca,Enrico O. Purisima,Adrian Wiegmans,Leonie A. Cluse,Ricky W. Johnstone,Pierre Beauparlant,Gordon C. Shore +16 more
TL;DR: Findings support a rational clinical development opportunity for the compound in cancer indications or treatments where MCL-1 contributes to resistance to cell killing.

Molecular dynamics study of small molecule inhibitors of the Bcl-2 family.
TL;DR: It is found that the binding of the chlorobiphenyl end of ABT‐737 was quite stable across all three proteins, and the phenylpiperazine linker group was dramatically more mobile in Mcl‐1 compared to either Bcl‐xL or B cl‐2.

Automated molecular formula determination by tandem mass spectrometry (MS/MS)
Suwatchai Jarussophon,Suwatchai Jarussophon,Stephane Acoca,Stephane Acoca,Jin-Ming Gao,Jin-Ming Gao,Christophe Deprez,Taira Kiyota,Cristina Draghici,Enrico O. Purisima,Yasuo Konishi +10 more
TL;DR: The molecular formulas of the precursor and the fragments were further used to analyze possible mass spectrometric fragmentation pathways, which would be a powerful tool in structural analysis and identification of small molecules.
Naphthalene-based RNA Editing Inhibitor Blocks RNA Editing Activities and Editosome Assembly in Trypanosoma brucei
Houtan Moshiri,Stephane Acoca,Smriti Kala,Hamed S. Najafabadi,Hervé Hogues,Enrico O. Purisima,Enrico O. Purisima,Reza Salavati +7 more
TL;DR: New and existing inhibitors of Trypanosoma brucei RNA editing ligase 1 are studied to better understand their mechanism of action and it is found that they can efficiently block deadenylylation of TbREL1 in the editosome and result in inhibition of the ligation step of RNA editing.

Obatoclax (GX15-070) Is a Potent Antagonist of Constitutive Mcl-1/Bak Interactions in Intact Mitochondrial Membrane and Synergizes with Bortezomib in Mantle Cell Lymphoma.
Gordon C. Shore,Mark F. Watson,Anne Roulston,Richard C. Marcellus,Nancy Steenart,Pierre Beauparlant,Sasmita Tripathy,Xavier Billot,Laurent Bélec,Giorgio Attardo,Mai Nguyen,Stephane Acoca,Enrico O. Purisima,Patricia Pérez-Galán,Dolors Colomer +14 more
TL;DR: A biochemical rationale has been established to interrogate obatoclax-bortezomib combinations in clinical trials directed at mantle cell lymphoma and indicates that it is a sensitizer rather than an activator of the intrinsic mitochondrial apoptosis program.
