Abstract S1-11: Meta-analysis Results from the Collaborative Trials in Neoadjuvant Breast Cancer (CTNeoBC)
Patricia Cortazar,Liling Zhang,Michael Untch,Keyur Mehta,J.C. Costantino,N Wolmark,Hervé Bonnefoi,David Cameron,Luca Gianni,P. Valagussa,JoAnne Zujewski,R Justice,S. Loibl,Lawrence Wickerham,Jan Bogaerts,J. Baselga,Charles M. Perou,Gideon M. Blumenthal,J Blohmer,E Mamounas,J Bergh,Vladimir Semiglazov,Tatiana M. Prowell,H. Eidtmann,Soonmyung Paik,M.J. Piccart,Rajeshwari Sridhara,Peter A. Fasching,Sandra M. Swain,Leen Slaets,Shenghui Tang,Bernd Gerber,Charles E. Geyer,R. Pazdur,Nina Ditsch,Priya Rastogi,Wolfgang Eiermann,G von Mincwitz +37 more
TL;DR: Eradication of tumor from both the breast and lymph nodes (ypT0ypN0 or ypT0/isyp N0) was better associated with improved EFS and OS compared to eradication of disease-free survival from the breast alone (ypt0/ is) and the magnitude of pCR effect needed to improve E FS and OS.

Abstract GS1-02: NSABP B-47 (NRG oncology): Phase III randomized trial comparing adjuvant chemotherapy with adriamycin (A) and cyclophosphamide (C) → weekly paclitaxel (WP), or docetaxel (T) and C with or without a year of trastuzumab (H) in women with node-positive or high-risk node-negative invasive breast cancer (IBC) expressing HER2 staining intensity of IHC 1+ or 2+ with negative FISH (HER2-Low IBC)
Louis Fehrenbacher,Reena S. Cecchini,Charles E. Geyer,P Rastogi,Joseph P. Costantino,JN Atkins,Jonathan Polikoff,J-F Boileau,Louise Provencher,C Stokoe,TD Moore,André Robidoux,Virginia F. Borges,KS Albain,Sandra M. Swain,Soonmyung Paik,Eleftherios P. Mamounas,Norman Wolmark +17 more
TL;DR: In this paper, the authors showed that the addition of trastuzumab (H) to CT did not demonstrate a reduction in IDFS, RFI, or DRFI in women with non-overexpressing but IHC measurable HER2 IBC.

Molecular profiling of breast cancer.
TL;DR: Gene-expression-based prognostic tests are now available as commercial reference laboratory tests and their successful implementation will depend on the seamless integration with existing clinicopathologic markers.

Genomic profiling of lung adenocarcinoma patients reveals therapeutic targets and confers clinical benefit when standard molecular testing is negative
Sun Min Lim,Eun Young Kim,Hye Ryun Kim,Siraj M. Ali,Joel R. Greenbowe,Hyo Sup Shim,Hyun Chang,Seungtaek Lim,Soonmyung Paik,Byoung Chul Cho +9 more
TL;DR: Fifty eight percent of patients wild type by standard testing for EGFR/KRAS/ALK have GAs identifiable by CGP that suggest benefit from target therapy, and CGP used when standard molecular testing for NSCLC is negative can reveal additional avenues of benefit from targeted therapy.
Abstract S3-06: A phase III trial evaluating pCR in patients with HR+, HER2-positive breast cancer treated with neoadjuvant docetaxel, carboplatin, trastuzumab, and pertuzumab (TCHP) +/- estrogen deprivation: NRG Oncology/NSABP B-52
Mothaffar F. Rimawi,Reena S. Cecchini,P Rastogi,Charles E. Geyer,Louis Fehrenbacher,Philip J. Stella,Z Dayao,Rachel Rabinovitch,SH Dyar,Patrick J. Flynn,Luis Baez-Diaz,Soonmyung Paik,Sandra M. Swain,Eleftherios P. Mamounas,CK Osborne,Norman Wolmark +15 more
TL;DR: The addition of estrogen deprivation to neoadjuvant chemotherapy is not antagonistic, it improved pCR rates numerically, but the improvement was not statistically significant, and the combination did not increase toxicity and may be a reasonable approach.
