Fused deposition modeling of novel scaffold architectures for tissue engineering applications.

This paper reviews biodegradable synthetic polymers focusing on their potential in tissue engineering applications. The major classes of polymers are briefly discussed with regard to synthesis, properties and biodegradability, and known degradation modes and products are indicated based on studies reported in the literature. A vast majority of biodegradable polymers studied belongs to the polyester family, which includes polyglycolides and polylactides. Some disadvantages of these polymers in tissue engineering applications are their poor biocompatibility, release of acidic degradation products, poor processability and loss of mechanical properties very early during degradation. Other degradable polymers such as polyorthoesters, polyanhydrides, polyphosphazenes, and polyurethanes are also discussed and their advantages and disadvantages summarised. With advancements in tissue engineering it has become necessary to develop polymers that meet more demanding requirements. Recent work has focused on developing injectable polymer compositions based on poly (propylene fumarate) and poly (anhydrides) to meet these requirements in orthopaedic tissue engineering. Polyurethanes have received recent attention for development of degradable polymers because of their great potential in tailoring polymer structure to achieve mechanical properties and biodegradability to suit a variety of applications.

Biodegradable synthetic polymers for tissue engineering

Biodegradable scaffolds have played an important role in a number of tissue engineering attempts over the past decade. The goal of this review article is to provide a brief overview of some of the important issues related to scaffolds fabricated from synthetic biodegradable polymers. Various types of such materials are available; some are commercialized and others are still in the laboratories. The properties of the most common of these polymers are discussed here. A variety of fabrication techniques were developed to fashion polymeric materials into porous scaffolds, and a selection of these is presented. The very important issue of scaffold architecture, including the topic of porosity and permeability, is discussed. Other areas such as cell growth on scaffolds, surface modification, scaffold mechanics, and the release of growths factors are also reviewed. A summary outlining the common themes in scaffold-related science that are found in the literature is presented.

Biodegradable polymeric scaffolds for musculoskeletal tissue engineering.

Photopolymerizable and degradable biomaterials are finding widespread application in the field of tissue engineering for the engineering of tissues such as bone, cartilage, and liver. The spatial and temporal control afforded by photoinitiated polymerizations has allowed for the development of injectable materials that can deliver cells and growth factors, as well as for the fabrication of scaffolding with complex structures. The materials developed for these applications range from entirely synthetic polymers (e.g., poly(ethylene glycol)) to purely natural polymers (e.g., hyaluronic acid) that are modified with photoreactive groups, with degradation based on the hydrolytic or enzymatic degradation of bonds in the polymer backbone or crosslinks. The degradation behavior also ranges from purely bulk to entirely surface degrading, based on the nature of the backbone chemistry and type of degradable units. The mechanical properties of these polymers are primarily based on factors such as the network crosslinking density and polymer concentration. As we better understand biological features necessary to control cellular behavior, smarter materials are being developed that can incorporate and mimic many of these factors.

Review: photopolymerizable and degradable biomaterials for tissue engineering applications.

This study was designed to investigate the in vivo biodegration and biocompatibility of a poly(propylene fumarate) (PPF)-based orthopedic biomaterial. The effects of varying the PPF to N-vinyl pyrrolidinone ratio and PPF to β-tricalcium phosphate content were studied. The composite mechanical properties and local tissue interactions were analyzed over 12 weeks. An initial increase in both compressive modulus and strength was seen for composite formulations that incorporated β-tricalcium phosphate. The samples incorporating a higher PPF to N-vinyl pyrrolidinone ratio reached a maximal compressive strength of 7.7 MPa and a maximal compressive modulus of 191.4 MPa at 3 weeks. The lower PPF to N-vinyl pyrrolidinone ratio samples gained a maximum compressive strength of 7.5 MPa initially and a compressive modulus of 134.0 MPa at 1 week. At 6 weeks, all samples for formulations incorporating β-tricalcium phosphate crumbled upon removal and were not mechanically tested. Samples that did not incorporate β-tricalcium phosphate were very weak and insufficient for bone replacement at the 4-day time point and beyond. Tissue interactions resulted in a mild inflammatory response at the initial time points and mature fibrous encapsulation by 12 weeks. © 1998 John Wiley & Sons, Inc. J.
