Inhibition of anaerobic digestion process: A review

Biofilms are complex communities of microorganisms attached to surfaces or associated with interfaces. Despite the focus of modern microbiology research on pure culture, planktonic (free-swimming) bacteria, it is now widely recognized that most bacteria found in natural, clinical, and industrial settings persist in association with surfaces. Furthermore, these microbial communities are often composed of multiple species that interact with each other and their environment. The determination of biofilm architecture, particularly the spatial arrangement of microcolonies (clusters of cells) relative to one another, has profound implications for the function of these complex communities. Numerous new experimental approaches and methodologies have been developed in order to explore metabolic interactions, phylogenetic groupings, and competition among members of the biofilm. To complement this broad view of biofilm ecology, individual organisms have been studied using molecular genetics in order to identify the genes required for biofilm development and to dissect the regulatory pathways that control the plankton-to-biofilm transition. These molecular genetic studies have led to the emergence of the concept of biofilm formation as a novel system for the study of bacterial development. The recent explosion in the field of biofilm research has led to exciting progress in the development of new technologies for studying these communities, advanced our understanding of the ecological significance of surface-attached bacteria, and provided new insights into the molecular genetic basis of biofilm development.

The effects of aging trichloroethylene (TCE) with a surrogate soil organic matter (SSOM) on its anaerobic biodegradation and desorption characteristics were studied. Surrogate soil organic matter comprised of composted sphagnum moss was used as a model sorbent, and Desulfomonile tiedjei was used as a model member of an anaerobic consortium capable of reductive dechlorination. After TCE was aged with the sterile SSOM for periods of 2, 14, and 30 d, the completely mixed batch reactors were inoculated with D. tiedjei cells and incubated for 5 d. The mass of TCE dechlorinated to cis-1,2-dichloroethylene (DCE) by D. tiedjei in this period decreased by 72% from 2 to 30 d of aging. Although there was an increase in the equilibrium desorption coefficient (Kd) with aging, sorption was completely reversible. Hence, the reduced availability of TCE for degradation was not coupled with irreversible sorption. To examine the longer-term fate of TCE, the unaged TCE-SSOM slurries were inoculated with D. tiedjei and then incubated for longer times (15, 24, and 29 d vs. 5 d). The conversion of TCE to cis-1,2-DCE was up to eight times greater than what was observed in control experiments without SSOM. Therefore, although aging resulted in reduced TCE biodegradation for short-term incubation (5 d), longer-term incubation (29 d) with the unaged SSOM resulted in extensive TCE dechlorination (up to 40%) to cis-1,2-DCE.

In-storage psychrophilic anaerobic digestion develops by microbial acclimation in covered swine-manure storage tanks, producing CH4 and stabilizing organic matter. To optimize the system's performance, the process kinetics must be understood. The objective of this study was to evaluate kinetic parameters describing the major stages in the digestion process, and to investigate the effect of temperature acclimation on these parameters. Specific activity tests were performed using manure inocula and five substrates at three incubation temperatures. Extant substrate activities were determined analytically for each case, and intrinsic kinetic parameters for glucose uptake were estimated by grid search fitting to the Monod model. The removal of pyrene and benzo(a)pyrene from contaminated water by sequential and simultaneous ozonation-bioremediation techniques was investigated. During the sequential treatment, ozonation using 0.5 or 2.5 mg/L ozone was used as a pretreatment process, whereas, during the simultaneous treatment process, ozonation of hydrocarbon-contaminated water at a predetermined duration using 0.5 mg/L ozone was made in the presence of microbial biomass. Ozonation was not beneficial for the removal of pyrene. However, despite a decreased specific biodegradation rate, ozonation improved the overall elimination of benzo(a)pyrene during both treatment processes. The overall removal of benzo(a)pyrene increased from 23 to 91% after exposure of the water to 0.5 mg/L ozone for 30 minutes during the simultaneous treatment process and further to 100% following exposure to 2.5 mg/L ozone for 60 minutes during the sequential treatment mode, demonstrating the benefits of combined ozonation-biological treatment for the removal of polycyclic aromatic hydrocarbons.
