Controversies on the Consequences of Iron Overload and Chelation in MDS
Francesca Vinchi,Saskia Hell,Uwe Platzbecker +2 more
- 27 May 2020
TL;DR: The still limited and somehow controversial experimental and clinical data available from preclinical studies and randomized trials highlight the need for further investigation to fully elucidate the mechanisms underlying the pathological impact of iron overload-mediated toxicity as well as the effect of classic and novel iron restriction approaches in MDS.

Cellular dynamics following CAR T cell therapy are associated with response and toxicity in relapsed/refractory myeloma.
Luise Fischer,N. Grieb,Patrick Born,Ronald Weiss,Sabine Seiffert,Andreas Boldt,Stephan Fricke,Paul Franz,Simone Heyn,Anne Sophie Kubasch,Ronny Baber,Heike Weidner,S. Wang,Enrica Bach,Sandra Hoffmann,J. Ussmann,Janine Kirchberg,Saskia Hell,Sebastian Schwind,K. Metzeler,M. Herling,Madlen Jentzsch,Georg-Nikolaus Franke,Ulrich Sack,Kristin Reiche,Ulrike Köhl,Uwe Platzbecker,Vladan Vucinic,Maximilian Merz +28 more
TL;DR: The results show that in vivo expansion of CD8+ CAR T cells is correlated to response, however persistence is not essential for durable remission in RRMM patients, and provide evidence, that an increased fraction of CD8+ T cells at day of leukapheresis in combination with successful lymphodepletion positively influence the outcome.

Single-cell multiomic dissection of response and resistance to chimeric antigen receptor T cells against BCMA in relapsed multiple myeloma.

Grieb,Ronald Weiss,Jaren Sia,Luise Fischer,Patrick Born,Andreas Boldt,Stephan Fricke,Paul Franz,Jonathan Scolnick,Lakshmi Venkatraman,Stacy Xu,Christina Kloetzer,Simone Heyn,Anne Sophie Kubasch,Ronny Baber,S. Platzbecker,Kristin Reiche,Vladan Vucinic,M. Merz +32 more
TL;DR: This work subjected mononuclear cells isolated from peripheral blood and bone marrow before and after the application of approved B cell maturation antigen-directed CAR T cells to single-cell multiomic analyses to identify markers associated with resistance and early relapse.

Impact of the changing landscape of induction therapy prior to autologous stem cell transplantation in 540 newly diagnosed myeloma patients: a retrospective real-world study
Song-Yau Wang,T. Holzhey,Simone Heyn,Thomas Zehrfeld,Susann Fricke,Franz Albert Hoffmann,Cornelia Becker,Leanthe Braunert,Thomas Edelmann,Inessa Paulenz,Marcus Hitzschke,Franziska Flade,Andreas Schwarzer,K. Fenchel,Georg-Nikolaus Franke,Vladan Vucinic,Madlen Jentzsch,Sebastian Schwind,Saskia Hell,Donata Backhaus,Thoralf Lange,Dietger Niederwieser,Markus Scholz,Uwe Platzbecker,Wolfram Pönisch +24 more
TL;DR: In this paper , a retrospective monocentric analysis included 540 patients with multiple myeloma (MM) who received an autologous stem cell transplantation (ASCT) between 1996 and 2019.
Prospective phase II study of preemptive chimerism-driven reduction of immunosuppression after non-myeloablative conditioning—Eudract #: 2007-002420-15

Cross,Sabine Leiblein,Enrica Bach,Wolfram Pönisch,Haifa Kathrin Al-Ali,Sebastian Schwind,Uwe Platzbecker,Thoralf Lange,Dietger Niederwieser,Vladan Vucinic +18 more
TL;DR: Most curative therapies for the required cytotoxic dose, reduced intensity and non-myeloablative regimens (NMA) been developed alloHSCT accessible to frail and comorbid patients 2.
