TL;DR: Deep sequencing is used to analyse genes that are recurrently mutated in AML to distinguish between individuals who have a high risk of developing AML and those with benign ARCH, providing proof-of-concept that it is possible to discriminate ARCH from pre-AML many years before malignant transformation.
Tracing the origins of relapse in acute myeloid leukaemia to stem cells

TL;DR: Combined genetic and functional analysis of purified subpopulations and xenografts from paired diagnosis/relapse samples are identified, identifying therapy-resistant cells already present at diagnosis and two major patterns of relapse.

Epigenetic therapy induces transcription of inverted SINEs and ADAR1 dependency.

TL;DR: It is shown that intronic and intergenic SINE elements, specifically inverted-repeat Alus, are the major source of drug-induced immunogenic dsRNA, which is destabilized by the RNA deaminase ADAR1, thereby limiting activation of the immune response.

TL;DR: In this article, the cellular and developmental context of preleukemic initiation and leukemic progression using gene editing in human disomic and trisomic fetal hematopoietic cells and xenotransplantation was characterized.

TL;DR: This ‘Singleton Correction’ methodology outperformed other UMI-based strategies in efficiency, leading to greater sensitivity with high specificity in a cell line dilution series, thus improving the cost-effectiveness and clinical impact of NGS.
