The present study aimed to quantify the concentration levels of potentially toxic elements (PTEs) such as aluminum, arsenic, cadmium, chromium, copper, nickel, lead, zinc, and mercury, as well as microplastics occurrence in various tissues of fish and seafood species, commercialized in the Lower Danube River Basin. A health risk assessment analysis was performed based on the PTEs concentration levels in the muscle tissue. Estimated daily intake (EDI), target hazard quotient (THQ), hazard index (HI), and target cancer risk (TR) of PTEs were calculated. It was observed that the species within the seafood category registered the highest levels of PTEs. For instance, in the muscle tissue of bivalve Mytilus galloprovincialis (from the Black Sea), the highest value was observed in the case of Zn (37.693 mg/kg), and the presence of polystyrene polymer was identified. The values associated with EDI, THQ, HI, and TR of PTE exposure were significantly lower than 1.

Combined Effects of Fluoride and Dietary Seleno-L-Methionine at Environmentally Relevant Concentrations on Female Zebrafish (Danio rerio) Liver: Histopathological Damages, Oxidative Stress and Inflammation.

We recently reported that transient receptor potential canonical (TRPC) 6 channel activity contributes to intracellular Zn2+ homeostasis in the heart. Zn2+ has also been implicated in the regulation of intestinal redox and microbial homeostasis. This study aims to investigate the role of TRPC6-mediated Zn2+ influx in the stress resistance of the intestine. The expression profile of TRPC1-C7 mRNAs in the actively inflamed mucosa from inflammatory bowel disease (IBD) patients was analyzed using the GEO database. Systemic TRPC3 knockout (KO) and TRPC6 KO mice were treated with dextran sulfate sodium (DSS) to induce colitis. The Zn2+ concentration and the mRNA expression levels of oxidative/inflammatory markers in colon tissues were quantitatively analyzed, and gut microbiota profiles were compared. TRPC6 mRNA expression level was increased in IBD patients and DSS-treated mouse colon tissues. DSS-treated TRPC6 KO mice, but not TRPC3 KO mice, showed severe weight loss and increased disease activity index compared with DSS-treated WT mice. The mRNA abundances of antioxidant proteins were basically increased in the TRPC6 KO colon, with changes in gut microbiota profiles. Treatment with TRPC6 activator prevented the DSS-induced colitis progression accompanied by increasing Zn2+ concentration. We suggest that TRPC6-mediated Zn2+ influx activity plays a key role in stress resistance against IBD, providing a new strategy for treating colitis.

Pharmacological Activation of TRPC6 Channel Prevents Colitis Progression

Reproductive traits of sheep are under the control of both the environment and genetics. Moreover, most of these traits have low heritability, are sex-limited, and expressed later in the animal's life, which makes it difficult to quantify in young animals and to apply direct selection to the traits. This calls for the need to identify and include the genetic bases of fertility traits in selection decisions to enhance the performance of fertility traits in sheep breeding programs. Therefore, several polymorphisms in many genes associated with fertility traits have been reported in several sheep breeds worldwide. The bone morphogenetic protein 15 (BMP15), bone morphogenetic protein receptor 1B (BMPR1B), and growth differentiation factor 9 (GDF9) genes are the major fecundity genes investigated in several sheep breeds studied. Polymorphisms on these genes have shown significant associations with fertility traits such as ovulation rate and litter size and have been used in selection programs. However, research findings focusing on candidate genes affecting fertility traits in indigenous sheep breeds from developing countries are lacking. This review has shown that the genes and the polymorphisms on the genes are variable between and within breeds of the sheep explored thus far, which calls for further efforts to examine sheep breeds in developing countries.

Simple Summary This article is of special interest for understanding the detriments of zinc deficiency and the benefits of zinc supplementation for aquacultures of Asian sea bass. We found that altering extracellular zinc conditions significantly affected the expression of certain zinc transporters and zinc-binding proteins as well as intracellular free zinc levels in cells from Asian sea bass (Lates calcarifer, SF cells). Interestingly, we detected no difference in the effects of organic compared to inorganic zinc supplements, when the same molar concentrations were added to the cultures. Moreover, changes in extracellular zinc conditions impacted on the expression of genes related to the redox balance and to growth hormone metabolism. Our data indicate that zinc deficiency induces a stress response in cells from Asian sea bass, which may be avoided when sufficient amounts of zinc are provided to the cells. We identified SF cells as a suitable model for studies to optimize zinc supplementation in the aquaculture of Asian sea bass. Abstract Since Asian sea bass is one of the economically most important fish, aquaculture conditions are constantly optimized. Evidence from feeding studies combined with the current understanding of the importance of zinc for growth and immune defense suggest that zinc supplementation may be a possible approach to optimize aquacultures of Asian sea bass. To investigate the effects of zinc deficiency and zinc supplementation, cells from Asian sea bass were incubated in culture medium with different zinc contents. The expression of genes, important for zinc homeostasis, redox metabolism, and growth hormones was analyzed using RT-PCR. Zinc deficiency induced the expression of certain zinc transporters (ZIP14, ZIP10, ZIP6, ZIP4, ZnT4, ZnT9) as well as of SOD1, IGF I and IGF II, while expression of ZnT1 and metallothionein (MT) was reduced. Zinc supplementation decreased the expression of ZIP10, while expression of ZnT1 and MT were elevated. No differences in the effects of zinc supplementation with zinc sulfate compared to supplementation with zinc amino acid complexes were observed. Thus, extracellular zinc conditions may govern the cellular zinc homeostasis, the redox metabolism and growth hormone expression in cells from Asian sea bass as reported for other fish species. Our data indicate that supplementing aquacultures with zinc may be recommended to avoid detriments of zinc deficiency.

Effects of Zinc Status on Expression of Zinc Transporters, Redox-Related Enzymes and Insulin-like Growth Factor in Asian Sea Bass Cells

This study investigated single-nucleotide polymorphisms (SNPs) within the CAPN1 316, CAPN1 4751, and CAST 2959 markers using high-resolution melting (HRM) analysis to predict meat tenderness in crossbred beef cattle. Tenderness was assessed using the Warner–Bratzler shear force (WBSF) test, with results expressed in grams (g), representing the force required to shear muscle fibers. Significant differences in phenotypic data were observed among the genotypic groups. The finding showed that polymorphisms at CAPN1 316, CAPN1 4751, and CAST 2959 exert interactive effects on meat quality traits. Notably, the TT genotype at CAPN1 4751 increased the adjusted WBSF (aWBSF) by approximately 792 g, indicating that TT was an unfavorable variant for tenderness. These results support the use of marker-assisted selection strategies in which the TT genotype is managed to minimize its frequency while other relevant markers are concurrently monitored, thereby enhancing genetic progress in meat tenderness across commercial cattle populations. This study demonstrated that CAPN1 4751 could serve as an effective marker for genetic selection in crossbred beef cattle and confirmed the efficiency of HRM analysis as a molecular tool for SNP genotyping. In conclusion, the findings provided an alternative approach for SNP detection in livestock breeding programs and represented an important step toward improving meat quality, meeting consumer expectations, and supporting the long-term sustainability of Thailand’s beef industry. The results highlighted the polygenic nature of meat tenderness and emphasized the importance of integrating multiple SNP markers to accurately assess the genetic potential for meat quality traits in cattle.

Single-Nucleotide Polymorphisms in Calpastatin (CAST) and Micro-Calpain (CAPN1) Genes Influencing Meat Tenderness in Crossbred Beef Cattle in Thailand

It is documented that the reproductive performance of postpartum dairy cows is influenced by both internal and external factors. One of the most important reproductive performance indices is the first ovulation postpartum. This review aimed to describe factors affecting first ovulation postpartum in dairy cows, particularly those raised under tropical climates. Negative energy balance and its consequences; metabolic disorders; uterine pathology; udder health; lameness; and heat stress are discussed for their potential effects on the first ovulation postpartum. Understanding the underlying mechanisms of those affecting factors would assist in justifying the treatment and prevention plans in order to optimize the reproductive performance of postpartum dairy cows.

Factors affecting first ovulation in postpartum dairy cows under tropical conditions: A review

This study aimed to investigate the appearance and frequencies of the Booroola polymorphism of the bone morphogenetic protein receptor 1b (BMPR1B) gene (FecB) and the Embrapa polymorphism of the growth differentiation factor 9 (GDF9) gene (FecGE) in sheep in Thailand. A total of 454 crossbred sheep blood samples were collected from four provinces in Thailand during August 2022 to July 2023. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to identify the FecB and FecGE genotypes. The history of ewe birth types was collected from the owners to analyze the association between fecundity (Fec) genotypes and the history of birth types. The genotypic frequencies of FecB for homozygous genotype (B/B), heterozygous genotype (+/B), and wildtype (+/+) were 0.22%, 1.54%, and 98.24%, respectively. Meanwhile, the genotypic frequencies of FecGE for homozygous genotype (E/E), heterozygous genotype (+/E), and wildtype (+/+) were 0.00%, 2.42%, and 97.58%, respectively. Furthermore, three ewes exhibited both FecB and FecGE genotypes. Fisher’s exact test revealed that possession of the FecB genotype was associated with multiple births (p < 0.01). Both FecB and FecGE mutations were identified in crossbred sheep in Thailand. Sheep containing FecB allele could be alternative candidates to be selected to improve the prolificacy of crossbred sheep in Thailand.
