Clinical studies have suggested that its presence at early postoperative stages may predict chronic (neuropathic) pain after surgery. Currently available animal models are typically associated with consistent tactile hypersensitivity and are therefore limited to distinguish betweenmechanisms that underlie tactile hypersensitivity as opposed to mechanisms that protect against it. In this study we havemodified the ratmodel of spared nerve injury, restricting the surgical lesion to a single peripheral branch of the sciatic nerve. This modification reduced the prevalence of tactile hypersensitivity from nearly 100% to approximately 50%. With this model, we here also demonstrated that the Regulator of G protein Signaling 4 (RGS4) was specifically up-regulated in the lumbar dorsal root ganglia and dorsal horn of rats developing tactile hypersensitivity. Intrathecal delivery of the RGS4 inhibitor CCG63802 was found to reverse tactile hypersensitivity for a 1 h period. Moreover, tactile hypersensitivity after modified spared nerve injury was most frequently persistent for at least four weeks and associated with higher reactivity of glial cells in the lumbar dorsal horn. Based on these datawe suggest that this new animalmodel of nerve injury represents an asset in understanding divergent neuropathic pain outcomes, so far unravelling a role of RGS4 in tactile hypersensitivity.Whether thismodel also holds promise in the study of the transition from acute to chronic pain will have to be seen in future investigations. © 2016 Published by Elsevier Inc.

Afferent information modulates spinal network activity in vitro and in preclinical animal models

Disc diseases are major public health problem in industrialized countries where they affect a large proportion of the population. Disc degeneration (DD) is considered to be one of the leading causes of pain consultation and sick leave in France. This study is an attempt to diagnose DD and more generally an assessment of the functionality and viability of the InterVertebral Disc (IVD). The IVD is an heterogeneous cartilage, that ensures rachis mobility and optimal stress redistribution between vertebrae. These two main properties are linked to the hydric content and the presence of proteoglycans (PG) which decline in a natural process throughout life. This degenerative process is in some case accelerated, leading to the Degenerative Disc Diseases (DDD) or troubles. Several studies have shown the importance of the water content of the disc on its biomechanical behavior. The aims of our study are:1. to follow with Magnetic Resonance Imaging (MRI), the variation in morphology and hydration under mechanical stress,2. to follow with a numerical model, the changes in mechanical parameters such as stiffness, Poisson's ratio and the intrinsic permeability of the IVD.The post-processing on Magnetic Resonance (MR) data allowed reconstructing the 3D deformation under a known mechanical load and deducing the porosity of the disc. The results obtained are conform with the literature and the adopted behavior adheres perfectly with the experimental data. This study demonstrates also, the ability to calculate the mechanical parameters of an IVD, providing precious information to understand the mechanical behaviour and hence judge the viability of the IVD.

In Vivo Assessment of Spinal Cord Integrity Using Magnetic Resonance Diffusion Tensor Imaging

Magnetic resonance diffusion tensor imaging of the spinal cord is challenging because of the cord’s thin structure and the presence of physiological and susceptibility artifacts. To circumvent these issues, we developed a methodology for imaging the thoraco-lumbar spinal cord of cats at 3 T using single-shot spin-echo echo planar imaging. The presented method could potentially be applied to humans since it was developed on a clinical scanner with a standard spine coil. Results provide (1) suggestions for optimal slice orientation and phase encoding direction; (2) an assessment of the benefits of parallel imaging to reduce geometric distortions; (3) the feasibility of acquiring quality diffusion weighted data in 13 min at a resolution of 1.1 mm 3 and (4) the determination of axonal disruption, in two cats with complete spinal cord transection, using tractography. © 2008 Elsevier Masson SAS.
