Biomarkers and Molecular Probes for Cell Death Imaging and Targeted Therapeutics

In deuterated molecules such as [1-¹³C]pyruvate-d₃, the nuclear spin polarization of ¹³C nuclei can be enhanced by combining Hartmann–Hahn cross-polarization (CP) at low temperatures (1.2 K) with dissolution dynamic nuclear polarization (D-DNP). The polarization is transferred from remote solvent protons to the ¹³C spins of interest. This allows one not only to slightly reduce build-up times but also to increase polarization levels and extend the lifetimes T₁(¹³C) of the enhanced ¹³C polarization during and after transfer from the polarizer to the NMR or MRI system. This extends time scales over which metabolic processes and chemical reactions can be monitored.

Hyperpolarization of Deuterated Metabolites via Remote Cross-Polarization and Dissolution Dynamic Nuclear Polarization B

The MRI (Magnetic Resonance Imaging) has been used as one of the powerful tools for medical diagnoses. Its usefulness is, however, still restricted because of the low spatial resolution and long measuring time mainly due to the low NMR (Nuclear Magnetic Resonance) signals relative to the noise levels. To overcome these restrictions, we started developing a method to remarkably enlarge the NMR signals about 10 years ago. We employ a method to hyperpolarize the nuclei, where the “hyperpolarize” means to artificially generate the nuclear polarization by many orders (102~106) of magnitude higher than the ordinary NMR signals currently in use. The hyperpolarized MRI would enable us to provide images with much higher spatial resolution and shorter measuring time than ever. Several techniques for hyperpolarization have been put into practical use; the BF (Brute Force) method, PHIP (Parahydrogen Induced Polarization) method, Laser optical pumping, DNP (Dynamic Nuclear Polarization), and so on. The experimental study on the hyperpolarization of 3He by the BF method, and that of 19F in PFC (Perfluorocarbon, known as an artificial blood) by the PHIP method has gotten started. In the former method, we use an extremely low temperature realized by the Pomeranchuk cooling in combination with 3He/4He dilution refrigerator and high magnetic field. In the latter method, we use the hydrogenation of the parahydrogen in the unsaturated hydrocarbon substrate at room temperature. Very recently, we started developing a novel type of the hyperpolarized MRI named HMM (Hyperpolarized Metabolic MRI) hoping for the cancer diagnosis.

Progress in Creation of a New Constant Medium for Hyperpolarized MRI

Abstract Objective. The need for increased sensitivity in magnetic resonance imaging (MRI) is crucial for its advancement as an imaging modality. The development of passive Lenz Resonators for effective RF magnetic field focusing will improve MRI sensitivity via local amplification of MRI signal, thereby leading to more efficient diagnosis and patient treatment. Approach. While there are methods for amplifying the signal from specific nuclei in MRI, such as hyperpolarization, a general solution will be more advantageous and would work in combination with these preexisting methods. While the Lenz Lens proposed such a general solution based on Lenz's law and the reciprocity principle, it came at the cost of limited signal enhancement. In this work, the first-in-kind prototype Lenz Resonator was conceived and examined as a general frequency-selective passive flux-focusing element for significant MRI signal enhancement. A 3.0 T Philips Achieva MRI was used to compare the signal from a phantom in the presence of Lenz Lenses, Lenz Resonators, and control trials with neither component. Main results. An MRI investigation demonstrated an experimental amplification of the signal-to-noise ratio up to 80% using an MRI insert of two coaxial Lenz Resonators due to superior B1 magnetic field focusing. The resonators displayed consistent amplification, nearly independent of their x-position within the MRI bore. Significance. This behavior demonstrates the feasibility of imaging large objects of varying shapes without penalties for signal amplification using Lenz Resonators. The Lenz Resonators versatility in geometrical design and consistent signal amplifying abilities between pulse sequences should allow for the development of Lenz Resonators suitable for most commonly used MRI setups. 

Novel frequency selective B1 focusing passive Lenz resonators for substantial MRI signal-to-noise ratio amplification

The project in producing the hyperpolarized 3He and 19F are addressed in pursuit of radiation free medical diagnosis. The program for production of the hyperpolarized 3He by the brute force method with the Pomeranchuk cooling and the rapid melting of the solid 3He started a few years ago, and is still on the way, while a new program for production of the hyperpolarized 19F by means of the PHIP (ParaHydrogen Induced Polarization) has just got started. Particular attention is placed upon a new idea of the hyperpolarization catalyst to be used for 19F.
