Supplementing ruminant animal diets with fat has been investigated as a means to influence a variety of physiological processes or to alter fatty acid composition of food products derived from ruminant animals. Several digestion experiments have been conducted with beef cattle and sheep to elucidate the effects of supplemental fat on utilization of other dietary components. Negative associative effects are not likely to be observed in ruminants consuming forage-based diets with supplemental fat at < or = 2% of DMI. Inclusion of supplemental fat at < or = 3% of DM is recommended to obtain the most benefit from the energy contained within the fat and other dietary components in high-forage diets. For ruminants fed high-concentrate diets, supplementing fat at 6% of diet DM is expected to have minimal impacts on utilization of other dietary components. Although there is greater potential to supply the ruminant animal with unsaturated fatty acids from dietary origin if fat is added to high-concentrate diets, incomplete ruminal biohydrogenation of C18 unsaturated fatty acids results in an increase in duodenal flow of 18:1 trans fatty acids regardless of basal diet consumed by the animal. The biohydrogenation intermediate 18:1 trans-11 (trans-vaccenic acid) is the likely precursor to cis-9, trans-11 CLA because the magnitude of increase in CLA content in tissues or milk of ruminants fed fat is much greater than the increase in CLA presented to the small intestine of ruminants fed fat supplements. Duodenal flow of trans-vaccenic acid is also substantially greater than CLA. Increasing unsaturated fatty acids status of ruminants imparts physiological responses that are separate than the energy value of supplemental fat. Manipulating maternal diet to improve unsaturated fatty acid status of the neonate has practical benefits for animals experiencing stress due to exposure to cold environments or conditions which mount an immune response. Supplementing fat to provide an additional 16 to 18 g/d of 18:2n-6 to the small intestine of beef cows for the first 60 to 90 d of lactation will have negative impacts on reproduction and may impair immune function of the suckling calf. Consequences of the suckling animal increasing its intake of unsaturated fatty acids because of manipulation of maternal diet warrants further investigation.

A multichannel system for classifying particles according to one or more characteristics of the particles includes a plurality of flow cytometry units. Each flow cytometry unit is operable to classify particles in a mixture of particles by interrogating a stream of fluid containing particles using a beam of electromagnetic radiation. Each flow cytometry unit has a sensor operable to generate a time-varying output signal indicative of at least one characteristic of the particles in the stream of fluid as the stream of fluid is interrogated by the beam of electromagnetic radiation. The flow cytometry units share an integrated platform and perhaps even a common processor for receiving and processing information from the units. The common processor is programmed to receive the time-varying output signals from the flow cytometry units substantially continuously and to process the output signals.

System for analyzing particles using multiple flow cytometry units

The present invention provides a method of cryopreserving sperm that have been selected for a specific characteristic. In a preferred embodiment, the method is employed to freeze sex-selected sperm. Although the cryopreservation method of the invention can be used to freeze sperm selected by any number of selection methods, selection using flow cytometry is preferred. The present invention also provides a frozen sperm sample that has been selected for a particular characteristic, such as sex-type. In preferred embodiments, the frozen sperm sample includes mammalian sperm, such as, for example, human, bovine, equine, porcine, ovine, elk, or bison sperm. The frozen selected sperm sample can be used in a variety of applications. In particular, the sample can be thawed and used for fertilization. Accordingly, the invention also includes a method of using the frozen selected sperm sample for artificial insemination or in vitro fertilization.

Method of cryopreserving selected sperm cells

Prepubertal F1 heifers (n = 246; from crossbred dams bred to either Hereford [H], Limousin [L], or Piedmontese [P] sires) were fed 1.9% (LF) or 4.4% (HF) dietary fat from 254+/-4 d of age until they reached puberty or the breeding season started. Safflower seeds (37% oil with 79% linoleic acid) were the added fat source. Blood samples and backfat thickness measurements were obtained from 60 randomly selected heifers representing the sire breeds and diets studied. In addition, five H-sired heifers from both diets were serially bled at 28-d intervals. Total gain, ADG, body condition score, and backfat thickness were affected by sire breed (P 0.10) but tended (P = 0.08) to affect the percentage of heifers pubertal by the beginning of breeding (June 4). Sire breed effects on puberty age at beginning of breeding, percentage pubertal at the beginning of breeding, and puberty age during the entire study were all highly significant. The effect of the diet x sire breed interaction on percentage of heifers pubertal at beginning of breeding (P < 0.05) was 74.4 vs 76.3% in H-sired, 69.8 vs 60.5% in L-sired, and 76.2 vs 97.6% in P-sired heifers (LF vs HF, respectively). Number of AI services per pregnancy and final pregnancy percentage were not affected by diet or the diet x sire breed interaction. Diet affected progesterone (P < 0.05) and cholesterol (P < 0.001) concentrations, and sire breed tended to affect (P = 0.06) cholesterol concentrations. The effect of the diet x time on feed interaction on cholesterol concentrations was highly significant. There were no effects of diet or sample period on insulin or growth hormone concentrations in serially collected blood samples. We conclude that effects of supplemental dietary fat may be breed-dependent and hypothesize that a feeding period of approximately 60 d duration may be more appropriate than the 162 d used in this study.

Effects of dietary fat and sire breed on puberty, weight, and reproductive traits of F1 beef heifers

Crossbred heifers (n = 75) fed for rapid (R; .82 kg/d) or slow-then-rapid (SR; .41 kg/d for 90 d then .82 kg/d) postweaning gain were used to examine the effects of age or pattern of gain on induction of puberty by a progestin. At 9.5, 11.0, and 12.5 mo of age, 12 prepuberal heifers from each growth treatment received progestin (a 6-mg Norgestomet implant for 10 d) or control treatments. Induction of puberty, LH secretory profiles, and ovarian follicular characteristics were assessed in Norgestomet-treated and control heifers. Body weights of R heifers were greater (P .1) LH secretory profiles, ovarian characteristics, or induction of puberty by Norgestomet. We conclude that progestins induce puberty by hastening the normal cascade of endocrine and ovarian events associated with spontaneous puberty. Furthermore, age, but not pattern of gain, seems to be the critical factor influencing the efficacy of progestins to induce puberty in heifers.
