Abstract PD6-11: An open-label, multitumor, phase II basket study of olaparib and durvalumab (MEDIOLA): Results in germline BRCA-mutated (gBRCAm) HER2-negative metastatic breast cancer (MBC)
Susan M. Domchek,Sophie Postel-Vinay,Y.-J. Bang,Young-Ae Park,Jérôme Alexandre,J.-P. Delord,Antoine Italiano,Benoit You,S Bastian,Matthew G Krebs,Ding Wang,S. Waqar,Helen K. Angell,M. Learoyd,S. Chang,C. Gresty,P. Herbolsheimer,Bella Kaufman +17 more
TL;DR: The combination of olaparib and durvalumab was well tolerated, with no apparent overlapping toxicities and the efficacy of the combination at an early time point reached the target DCR in gBRCAm HER2-negative MBC, and warrants further investigation.

Phase II study of olaparib (O) and durvalumab (D) (MEDIOLA): Updated results in patients (pts) with germline BRCA-mutated (gBRCAm) metastatic breast cancer (MBC)
Susan M. Domchek,Sophie Postel-Vinay,S.-A. Im,Young-Ae Park,J.-P. Delord,Antoine Italiano,Jérôme Alexandre,Benoit You,S Bastian,Matthew G Krebs,Ding Wang,S. Waqar,M. Lanasa,Helen K. Angell,Zhongwu Lai,C. Gresty,L Opincar,P. Herbolsheimer,Bella Kaufman +18 more
TL;DR: The data suggest that pts with fewer prior lines of chemotherapy (0/1) had higher ORR, longer mDoR, mPFS and mOS than those with 2 prior lines.

TL;DR: Olaparib plus durvalumab with bevacizumab demonstrated encouraging clinical activity in women with non-gBRCAm PSROC, and no new safety signals were identified.

Abstract PD5-04: An open-label, phase II basket study of olaparib and durvalumab (MEDIOLA): Updated results in patients with germline BRCA-mutated (gBRCAm) metastatic breast cancer (MBC)
Susan M. Domchek,Sophie Postel-Vinay,S-A. Im,Y Hee Park,J.-P. Delord,Antoine Italiano,Jérôme Alexandre,Benoit You,S Bastian,Matthew G Krebs,S. Waqar,M. Lanasa,Helen K. Angell,M Tang,C. Gresty,L Opincar,P. Herbolsheimer,Bella Kaufman +17 more
TL;DR: The addition of durvalumab to olaparib showed promising activity with the ORR in the early line gBRCAm MBC pts, comparing favorably with olaparaib monotherapy (OlympiAD) and the median DoR appeared longer with the combination, warranting early confirmation of activity in the small number of pts.

Abstract OT3-05-03: MEDIOLA: An open-label, phase I/II basket study of olaparib (PARP inhibitor) and durvalumab (anti-PD-L1 antibody)–Additional breast cancer cohorts
Susan M. Domchek,Sophie Postel-Vinay,S-A. Im,Y Hee Park,J.-P. Delord,Antoine Italiano,Jérôme Alexandre,Benoit You,S Bastian,Matthew G Krebs,S. Waqar,M. Lanasa,Helen K. Angell,M Tang,C. Gresty,L Opincar,P. Herbolsheimer,Bella Kaufman +17 more
TL;DR: Evidence suggests that mutations in other homologous recombination repair (HRR) genes may confer a BRCA-like phenotype, warranting an expansion of the study population beyond BRCAm, and MEDIOLA is a Ph I/II open-label, multicenter study enrolling patients across several tumor types.
