Recently a novel DNA virus (Torque Teno Virus (TTV) has been identified in Japan and shown to be associated with elevated aminotransferase s levels after transfusion. However the exact role of TTV in pathogenesis of liver disease is yet to be established. The purpose of this study was to determine the prevalence of TTV in thalassemic patients and its relationship with elevated alanine-aminotransfrase (ALT) and aspartateaminotransfrase (AST). This cross-sectional analysis study was conducted on 452 thallasemic patients. Serums were collected from all of the patients, first ALT and AST levels were determined. Then, after DNA extraction, TTV DNA was amplified and detected using semi-nested PCR, followed by gel electrophoresis. Demographic characteristics and clinical data were collected from each participant for statistical analysis. The findings showed that 160 of 452 (35.4%) samples had TTV DNA detected by PCR. From 160 TTV DNA positive, 98 (61.20%) were female and 62 (38.80%) of them were male (P=0.549). The mean ALT and AST values in TTV positive group were higher than in TTV negative group, and the difference was statistically significant (p<0.0001). The result showed that the prevalence of TTV in thalassemic patients in Jahrom is less than other studies in Iran and the mean ALT and AST values in TTV positive individuals were about 2 times more than in TTV negative individuals.

Molecular Prevalence and Clinical Importance of Torque Teno Virus Infection in Thalassemia Patients as High Risk Individuals

Polycyclic aromatic hydrocarbons (PAHs) include a group of organic priority pollutants of critical environmental and public health concern due to their toxic, genotoxic, mutagenic and/or carcinogenic properties and their ubiquitous occurrence as well as recalcitrance. The increased awareness of their various adverse effects on ecosystem and human health has led to a dramatic increase in research aimed towards removing PAHs from the environment. PAHs may undergo adsorption, volatilization, photolysis, and chemical oxidation, although transformation by microorganisms is the major neutralization process of PAH-contaminated sites in an ecologically accepted manner. Microbial degradation of PAHs depends on various environmental conditions, such as nutrients, number and kind of the microorganisms, nature as well as chemical property of the PAH being degraded. A wide variety of bacterial, fungal and algal species have the potential to degrade/transform PAHs, among which bacteria and fungi mediated degradation has been studied most extensively. In last few decades microbial community analysis, biochemical pathway for PAHs degradation, gene organization, enzyme system, genetic regulation for PAH degradation have been explored in great detail. Although, xenobiotic-degrading microorganisms have incredible potential to restore contaminated environments inexpensively yet effectively, but new advancements are required to make such microbes effective and more powerful in removing those compounds, which were once thought to be recalcitrant. Recent analytical chemistry and genetic engineering tools might help to improve the efficiency of degradation of PAHs by microorganisms, and minimize uncertainties of successful bioremediation. However, appropriate implementation of the potential of naturally occurring microorganisms for field bioremediation could be considerably enhanced by optimizing certain factors such as bioavailability, adsorption and mass transfer of PAHs. The main purpose of this review is to provide an overview of current knowledge of bacteria, halophilic archaea, fungi and algae mediated degradation/transformation of PAHs. In addition, factors affecting PAHs degradation in the environment, recent advancement in genetic, genomic, proteomic and metabolomic techniques are also highlighted with an aim to facilitate the development of a new insight into the bioremediation of PAH in the environment.

Current State of Knowledge in Microbial Degradation of Polycyclic Aromatic Hydrocarbons (PAHs): A Review

Many hypersaline environments are often contaminated with petroleum compounds. Among these, oil and natural gas production sites all over the world and hundreds of kilometers of coastlines in the more arid regions of Gulf countries are of major concern due to the extent and magnitude of contamination. Because conventional microbiological processes do not function well at elevated salinities, bioremediation of hypersaline environments can only be accomplished using high salt-tolerant microorganisms capable of degrading petroleum compounds. In the last two decades, there have been many reports on the biodegradation of hydrocarbons in moderate to high salinity environments. Numerous microorganisms belonging to the domain Bacteria and Archaea have been isolated and their phylogeny and metabolic capacity to degrade a variety of aliphatic and aromatic hydrocarbons in varying salinities have been demonstrated. This article focuses on our growing understanding of bacteria and archaea responsible for the degradation of hydrocarbons under aerobic conditions in moderate to high salinity conditions. Even though organisms belonging to various genera have been shown to degrade hydrocarbons, members of the genera Halomonas Alcanivorax, Marinobacter, Haloferax, Haloarcula, and Halobacterium dominate the published literature. Despite rapid advances in understanding microbial taxa that degrade hydrocarbons under aerobic conditions, not much is known about organisms that carry out similar processes in anaerobic conditions. Also, information on molecular mechanisms and pathways of hydrocarbon degradation in high salinity is scarce and only recently there have been a few reports describing genes, enzymes and breakdown steps for some hydrocarbons. These limited studies have clearly revealed that degradation of oxygenated and non-oxygenated hydrocarbons by halophilic and halotolerant microorganisms occur by pathways similar to those found in non-halophiles.

Progress in decontamination by halophilic microorganisms in saline wastewater and soil

Agro-food, petroleum, textile, and leather industries generate saline wastewater with a high content of organic pollutants such as aromatic hydrocarbons, phenols, nitroaromatics, and azo dyes. Halophilic microorganisms are of increasing interest in industrial waste treatment, due to their ability to degrade hazardous substances efficiently under high salt conditions. However, their full potential remains unexplored. The isolation and identification of halophilic and halotolerant microorganisms from geographically unrelated and geologically diverse hypersaline sites supports their application in bioremediation processes. Past investigations in this field have mainly focused on the elimination of polycyclic aromatic hydrocarbons and phenols, whereas few studies have investigated N-aromatic compounds, such as nitro-substituted compounds, amines, and azo dyes, in saline wastewater. Information regarding the growth conditions and degradation mechanisms of halophilic microorganisms is also limited. In this review, we discuss recent research on the removal of organic pollutants such as organic matter, in terms of chemical oxygen demand (COD), dyes, hydrocarbons, N-aliphatic and N-aromatic compounds, and phenols, in conditions of high salinity. In addition, some proposal pathways for the degradation of aromatic compounds are presented.

Biodegradation of organic pollutants in saline wastewater by halophilic microorganisms: a review.

A polycyclic aromatic hydrocarbon-degrading marine bacterium, designated strain P-4T, was isolated from oil-polluted saline soil in Xianhe, Shangdong Province, China. Strain P-4T was Gram-negative-staining with curved to spiral rod-shaped cells and grew optimally with 3–6 % (w/v) NaCl and at 30 °C. The predominant fatty acids were C18 : 1
ω7c (35.0 %), C16 : 0 (25.0 %), C16 : 1
ω7c (17.9 %), C14 : 0 (6.2 %) and C17 : 0 cyclo (5.2 %). The major respiratory quinone was Q-9 and the genomic DNA G+C content was 61.2±1.0 mol%. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain P-4T belonged to the genus Thalassospira of the class Alphaproteobacteria. DNA–DNA hybridization with Thalassospira xiamenensis DSM 17429T showed relatedness of 36.0 %, and lower values were obtained with respect to other Thalassospira species. Based on physiological and biochemical tests and 16S rRNA gene sequence analysis as well as DNA–DNA relatedness, strain P-4T should be placed in the genus Thalassospira within a novel species. The name Thalassospira xianhensis sp. nov. is proposed, with P-4T (=CGMCC 1.6849T =JCM 14850T) as the type strain.

A Gram-negative, aerobic, short rod-shaped and non-motile bacterium, strain A-1(T), was isolated from a saline soil contaminated with crude oil in Xianhe, Shangdong Province, China. Strain A-1(T) formed yellow colonies, was moderately halophilic and grew with 0.05-27.5% (w/v) total salts (optimum 5-8%), at 10-42 °C (optimum 30 °C) and at pH 5.5-9.0 (optimum pH 7.2). The dominant fatty acids (>5%) were C(16:0), summed feature 3 (comprising C(16:1)ω7c and/or iso-C(15:0) 2-OH), C(18:1)ω7c, C(19:0) cyclo ω8c and C(12:0) 3-OH and the predominant ubiquinone was Q-9. The genomic DNA G+C content was 67.1 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain A-1(T) belonged to the genus Halomonas in the class Gammaproteobacteria. The closest relatives were Halomonas lutea YIM 91125(T) (97.7% 16S rRNA gene sequence similarity), H. muralis LMG 20969(T) (95.6%), H. pantelleriensis AAP(T) (95.5%) and H. kribbensis BH843(T) (95.2%). DNA-DNA relatedness between strain A-1(T) and H. lutea CCTCC AB 206093(T) was 27±3%. On the basis of phenotypic, chemotaxonomic and phylogenetic features, strain A-1(T) should be placed in the genus Halomonas as a representative of a novel species. The name Halomonas xianhensis sp. nov. is proposed, with strain A-1(T) (=CGMCC 1.6848(T) =JCM 14849(T)) as the type strain.

A halophilic bacterial consortium that degraded phenanthrene was developed from oil-contaminated saline soil containing 10% salinity. The biodegradation of phenanthrene occurred at 5%, 10%, and 15% salinity, whereas no biodegradation took place at 0.1% and 20% salinity. A 16S rRNA gene analysis showed that all sequences from the denaturing gradient gel electrophoresis profile were similar to those of halophilic bacteria. This is the first report of a halophilic bacterial consortium capable of degrading phenanthrene under hypersaline conditions.
