Molecular photoswitches are the foundation of light-activated drugs. A key photoswitch is azobenzene, which exhibits trans–cis isomerism in response to light. The thermal half-life of the cis isomer is of crucial importance, since it controls the duration of the light-induced biological effect. Here we introduce a computational tool for predicting the thermal half-lives of azobenzene derivatives. Our automated approach uses a fast and accurate machine learning potential trained on quantum chemistry data. Building on well-established earlier evidence, we argue that thermal isomerization proceeds through rotation mediated by intersystem crossing, and incorporate this mechanism into our automated workflow. We use our approach to predict the thermal half-lives of 19,000 azobenzene derivatives. We explore trends and trade-offs between barriers and absorption wavelengths, and open-source our data and software to accelerate research in photopharmacology.

Thermal Half-Lives of Azobenzene Derivatives: Virtual Screening Based on Intersystem Crossing Using a Machine Learning Potential

Photoswitchable molecules display two or more isomeric forms that may be accessed using light. Separating the electronic absorption bands of these isomers is key to selectively addressing a specific isomer and achieving high photostationary states whilst overall red-shifting the absorption bands serves to limit material damage due to UV-exposure and increases penetration depth in photopharmacological applications. Engineering these properties into a system through synthetic design however, remains a challenge. Here, we present a data-driven discovery pipeline for molecular photoswitches underpinned by dataset curation and multitask learning with Gaussian processes. In the prediction of electronic transition wavelengths, we demonstrate that a multioutput Gaussian process (MOGP) trained using labels from four photoswitch transition wavelengths yields the strongest predictive performance relative to single-task models as well as operationally outperforming time-dependent density functional theory (TD-DFT) in terms of the wall-clock time for prediction. We validate our proposed approach experimentally by screening a library of commercially available photoswitchable molecules. Through this screen, we identified several motifs that displayed separated electronic absorption bands of their isomers, exhibited red-shifted absorptions, and are suited for information transfer and photopharmacological applications. Data-driven discovery of molecular photoswitches with multioutput Gaussian processes

Azobenzenes (Ph-N═N-Ph) are known as the most widely studied molecular photoswitches, and the recent rise of azoheteroarenes (Het-N═N-Ph or Het-N═N-Het) offers great opportunities to advance this already mature field. A common limitation is that azo-switches generally require harmful UV light for activation, which hinders their application across various fields. Despite great efforts in developing visible-light azobenzenes over the past few decades, the potential of visible-light heteroaryl azoswitches remains largely unexplored. This Perspective summarizes the state-of-the-art advancements in visible-light heteroaryl azoswitches, covering molecular design strategies, the structure-property relationship, and potential applications. We highlight the distinctive advantages of azoheteroarenes over azobenzenes in the research and development of visible-light switches. Furthermore, we discuss the opportunities and challenges in this emerging field and propose potential solutions to address crucial issues such as spectral red-shift and thermal half-life. Through this Perspective paper, we aim to provide inspiration for further exploration in this field, in anticipation of the growing prosperity and bright future of visible-light azoheteroarene photoswitches. 

Visible-Light-Activated Heteroaryl Azoswitches: Toward a More Colorful Future

The visual analytics community has long aimed to understand users better and assist them in their analytic endeavors. As a result, numerous conceptual models of visual analytics aim to formalize common workflows, techniques, and goals leveraged by analysts. While many of the existing approaches are rich in detail, they each are specific to a particular aspect of the visual analytic process. Furthermore, with an ever-expanding array of novel artificial intelligence techniques and advances in visual analytic settings, existing conceptual models may not provide enough expressivity to bridge the two fields. In this work, we propose an agent-based conceptual model for the visual analytic process by drawing parallels from the artificial intelligence literature. We present three examples from the visual analytics literature as case studies and examine them in detail using our framework. Our simple yet robust framework unifies the visual analytic pipeline to enable researchers and practitioners to reason about scenarios that are becoming increasingly prominent in the field, namely mixed-initiative, guided, and collaborative analysis. Furthermore, it will allow us to characterize analysts, visual analytic settings, and guidance from the lenses of human agents, environments, and artificial agents, respectively. 

Human–Computer Collaboration for Visual Analytics: an Agent‐based Framework

Photoswitches are molecules that undergo a reversible, structural isomerization after exposure to certain wavelengths of light. The dynamic control offered by molecular photoswitches is favorable for materials chemistry, photopharmacology, and catalysis applications. Ideal photoswitches absorb visible light and have long-lived metastable isomers. We used high-throughput virtual screening to predict the absorption maxima (λmax) of the E-isomer and half-life (t1/2) of the Z-isomer. However, computing the photophysical and kinetic stabilities with density functional theory of each entry of a virtual molecular library containing thousands or millions of molecules is prohibitively time-consuming. We applied active search, a machine-learning technique, to intelligently search a chemical search space of 255 991 photoswitches based on 29 known azoarenes and their derivatives. We iteratively trained the active search algorithm on whether a candidate absorbed visible light (λmax > 450 nm). Active search was found to triple the discovery rate compared to random search. Further, we projected 1962 photoswitches to 2D using the Uniform Manifold Approximation and Projection algorithm and found that λmax depends on the core, which is tunable by substituents. We then incorporated a second stage of screening to predict the stabilities of the Z-isomers for the top candidates of each core. We identified four ideal photoswitches that concurrently satisfy the following criteria: λmax > 450 nm and t1/2 > 2 h.These candidates had λmax and t1/2 range from 465 to 531 nm and hours to days, respectively.

Efficient discovery of visible light-activated azoarene photoswitches with long half-livesusing active search

Projection algorithms such as t-SNE or UMAP are useful for the visualization of high dimensional data, but depend on hyperparameters which must be tuned carefully. Unfortunately, iteratively recomputing projections to find the optimal hyperparameter value is computationally intensive and unintuitive due to the stochastic nature of these methods. In this paper we propose HyperNP, a scalable method that allows for real-time interactive hyperparameter exploration of projection methods by training neural network approximations. HyperNP can be trained on a fraction of the total data instances and hyperparameter configurations and can compute projections for new data and hyperparameters at interactive speeds. HyperNP is compact in size and fast to compute, thus allowing it to be embedded in lightweight visualization systems such as web browsers. We evaluate the performance of the HyperNP across three datasets in terms of performance and speed. The results suggest that HyperNP is accurate, scalable, interactive, and appropriate for use in real-world settings.
