Fabrication of physical and chemical crosslinked hydrogels for bone tissue engineering

Biomimetic glycopeptide hydrogel coated PCL/nHA scaffold for enhanced cranial bone regeneration via macrophage M2 polarization-induced osteo-immunomodulation.

Due to the abuse of antibiotics and the emergence of multidrug resistant microorganisms, medical devices, and related biomaterials are at high risk of microbial infection during use, placing a heavy burden on patients and healthcare systems. Metal–phenolic networks (MPNs), an emerging organic–inorganic hybrid network system developed gradually in recent years, have exhibited excellent multifunctional properties such as anti‐inflammatory, antioxidant, and antibacterial properties by making use of the coordination between phenolic ligands and metal ions. Further, MPNs have received widespread attention in antimicrobial infections due to their facile synthesis process, excellent biocompatibility, and excellent antimicrobial properties brought about by polyphenols and metal ions. In this review, different categories of biomaterials based on MPNs (nanoparticles, coatings, capsules, hydrogels) and their fabrication strategies are summarized, and recent research advances in their antimicrobial applications in biomedical fields (e.g., skin repair, bone regeneration, medical devices, etc.) are highlighted.

Recent Advances in the Development and Antimicrobial Applications of Metal–Phenolic Networks

3D‐printed polycaprolactone (PCL) scaffolds have been extensively studied for application in bone tissue engineering. However, PCL‐based scaffolds with enhanced bioactivity and intelligent delivery capability for bone repair remains challenging. Herein, CuS nanoparticle‐PEG soft hydrogel‐coated 3D hard polycaprolactone scaffolds (denoted as CuS‐PEG‐PCL scaffold) are rationally designed for efficient bone regeneration. CuS nanoparticles cross‐linked PEG hydrogel (CuS‐PEG‐hydrogel) endows the PCL‐based scaffold with excellent photothermal properties and stable soft elasticity, while the PCL scaffold provides excellent mechanical performance. Upon exposure to 1064 nm near‐infrared (NIR) light irradiation, dexamethasone sodium phosphate (Dexp), stored in the CuS‐PEG‐PCL scaffold, can be controllably released, which efficiently promotes osteogenic differentiation of bone mesenchymal stem cells (BMSCs). In addition, the combination of mild heating at 42 ± 0.5 °C further promoted osteogenic differentiation of BMSCs. Subsequently, this Dexp‐loaded CuS‐PEG‐PCL scaffold (D‐CuS‐PEG‐PCL scaffold) with NIR light treatment at the tibial defect of rats presented the highest bone regeneration capacity. These findings demonstrate that the Dexp‐loaded CuS‐PEG‐hydrogel can effectively modify the 3D printed PCL scaffold. Therefore, this multifunctional scaffold with a soft‐hard hybrid structure has the potential to become an advanced drug delivery vehicle in the treatment of large bone defects.

Rational Design of Multifunctional CuS Nanoparticle‐PEG Composite Soft Hydrogel‐Coated 3D Hard Polycaprolactone Scaffolds for Efficient Bone Regeneration

Although gradients play an essential role in guiding the function of tissues, achieving synchronous regeneration of gradient tissue injuries remains a challenge. Here, a gradient bimetallic (Cu and Zn) ion-based hydrogel was first constructed via the one-step coordinative crosslinking of sulfhydryl groups with copper and zinc ions for the microstructure reconstruction of the tendon-to-bone insertion. In this bimetallic hydrogel system, zinc and copper ions could not only act as crosslinkers but also provide strong antibacterial effects and induce regenerative capacity in vitro. The capability of hydrogels in simultaneously promoting tenogenesis and osteogenesis was further verified in a rat rotator cuff tear model. It was found that the Cu/Zn gradient layer could induce considerable collagen and fibrocartilage arrangement and ingrowth at the tendon-to-bone interface. Overall, the gradient bimetallic ion-based hydrogel ensures accessibility and provides opportunities to regenerate inhomogeneous tissue with physiological complexity or interface tissue.
