Trained immunity is defined as the de facto memory characteristics induced in innate immune cells after exposure to microbial stimuli after infections or certain types of vaccines. Through epigenetic and metabolic reprogramming of innate immune cells after exposure to these stimuli, trained immunity induces an enhanced nonspecific protection by improving the inflammatory response upon restimulation with the same or different pathogens. Recent studies have increasingly shown that trained immunity can, on the one hand, be induced by exposure to viruses; on the other hand, when induced, it can also provide protection against heterologous viral infections. In this review we explore current knowledge on trained immunity and its relevance for viral infections, as well as its possible future uses. Shifting the Immune Memory Paradigm: Trained Immunity in Viral Infections.

Abstract Background Primary cilia emanate from most human cell types, including neurons. Cilia are important for communicating with the cell’s immediate environment: signal reception and transduction to/from the ciliated cell. Deregulation of ciliary signaling can lead to ciliopathies and certain neurodevelopmental disorders. In the developing brain cilia play well-documented roles for the expansion of the neural progenitor cell pool, while information about the roles of cilia during post-mitotic neuron differentiation and maturation is scarce. Results We employed ciliated Lund Human Mesencephalic (LUHMES) cells in time course experiments to assess the impact of ciliary signaling on neuron differentiation. By comparing ciliated and non-ciliated neuronal precursor cells and neurons in wild type and in RFX2 -/- mutant neurons with altered cilia, we discovered an early-differentiation “ciliary time window” during which transient cilia promote axon outgrowth, branching and arborization. Experiments in neurons with IFT88 and IFT172 ciliary gene knockdowns, leading to shorter cilia, confirm these results. Cilia promote neuron differentiation by tipping WNT signaling toward the non-canonical pathway, in turn activating WNT pathway output genes implicated in cyto-architectural changes. Conclusions We provide a mechanistic entry point into when and how ciliary signaling coordinates, promotes and translates into anatomical changes. We hypothesize that ciliary alterations causing neuron differentiation defects may result in “mild” impairments of brain development, possibly underpinning certain aspects of neurodevelopmental disorders.

Primary cilia promote the differentiation of human neurons through the WNT signaling pathway

Here, we present a protocol for differential multi-omic analyses of distinct cell types in the developing mouse cerebral cortex. We describe steps for in utero electroporation, subsequent flow-cytometry-based isolation of developing mouse cortical cells, bulk RNA sequencing or quantitative liquid chromatography-tandem mass spectrometry, and bioinformatic analyses. This protocol can be applied to compare the proteomes and transcriptomes of developing mouse cortical cell populations after various manipulations (e.g., epigenetic). How neurons accomplish the growth of an axon, and several dendrites sequentially is a cellular process not well understood. Here, we show that preferential somatic F-actin delivery to neurites inhibits neurite growth during axon formation. Thus, the neurite receiving less somatic F-actin is growing as an axon. During dendrites outgrowth, radial somatic F-actin translocation into all neurites is drastically diminished suggesting that cellular growth is restricted by the somatic F-actin flow into the growing neurites. Mechanistically, we report that radial somatic F-actin translocation is mediated via the Myosin II motor. Pharmacological inhibition of these molecular motors promotes neurite elongation and precludes F-actin translocation into neurites. Moreover, we show that actin phosphorylation at Y53, which promotes F-actin instability, is enriched selectively in some neurite shafts to exclude/minimize translocation of somatic F-actin only into those neurites. Therefore, enrichment of actin pY53 in the neurite shaft correlates with the longer neurite during axon formation. Accordingly, the Kinesin-1 motor domain, which accumulates transiently in neurites of stage 2 neurons and only in the emerging axon of stage 3 neurons, localizes preferentially in the neurite with the increased actin pY53 signal in the neurite shaft. Finally, we demonstrate that microtubule acetylation promotes actin phosphorylation, and these cytoskeleton modifications coexist in the longest neurite that grows as an axon. Collectively, our data support a model in which somatic F-actin translocation into undifferentiated neurites impedes growth counteracted by actin pY53, consequently, supporting neuronal polarization during axon formation.
