TL;DR: It is shown that imipridones bind ClpP non-covalently and induce proteolysis by diverse structural changes, suggesting a general concept of inducing cancer cell lethality through activation of mitochondrial proteolytic activity.

Inhibition of translation initiation factor eIF4a inactivates heat shock factor 1 (HSF1) and exerts anti-leukemia activity in AML.
Yuki Nishida,Ran Zhao,Lauren Heese,Hiroki Akiyama,Shreya Patel,Alex M. Jaeger,Rodrigo Jacamo,Kensuke Kojima,Kensuke Kojima,Man Chun John Ma,Vivian Ruvolo,Dhruv Chachad,William G. Devine,Susan Lindquist,R. Eric Davis,John A. Porco,Luke Whitesell,Luke Whitesell,Michael Andreeff,Jo Ishizawa +19 more
TL;DR: In this article, the eIF4A inhibition by the potent and specific compound rohinitib (RHT) inactivated HSF1 in these cells, and exerted pronounced in vitro and in vivo anti-leukemia effects against progenitor and leukemia-initiating cells, especially those with FLT3-internal tandem duplication (ITD).

Transcriptome analysis of acute high temperature-responsive genes and pathways in Palaemon gravieri.
Wenjun Shi,Runhao Hu,Pan Wang,Ran Zhao,Hui Shen,Hui Li,Libao Wang,Yi-Lun Qiao,Ge Jiang,Jie Cheng,Xihe Wan +10 more
TL;DR: In this paper , Zhao et al. investigated the effects of acute high temperature on P. gravieri and found that high temperature significantly reduced the metabolic capacity of shrimp but enhanced the immune capacity.

Abstract 4957: The novel imipridone ONC212 highly synergizes with the BCL-2 inhibitor ABT-199 in AML and activates orphan receptor GPR132
Takenobu Nii,Jo Ishizawa,Varun V. Prabhu,Vivian Ruvolo,Neel Madhukar,Ran Zhao,Hong Mu,Lauren Heese,Kensuke Kojima,Mathew J. Garnett,Ultan McDermott,Cyril H. Benes,Neil Charter,Sean Deacon,Olivier Elemento,Joshua E. Allen,Wolfgang Oster,Martin Stogniew,Michael Andreeff +18 more
TL;DR: The novel imipridone ONC212 highly synergizes with the BCL-2 inhibitor ABT-199 in AML and activates orphan receptor GPR132 and is investigated in vivo in an aggressive systemic AML xenograft model using OCI-AML3 cells.

Formate as a supplementary substrate facilitates sugar metabolism and solvent production by Clostridium beijerinckii NCIMB 8052
TL;DR: In this paper , the authors showed that formate could be metabolized via both assimilation and dissimilation pathways in C. beijerinckii NCIMB 8052, a typical solventogenic Clostridium strain.
