Laminar and Columnar Development of Barrel Cortex Relies on Thalamocortical Neurotransmission

GABAA receptors mediate the majority of inhibitory neurotransmission in the CNS. Genetic deletion of the α1 subunit of GABAA receptors results in a loss of α1-mediated fast inhibitory currents and a marked reduction in density of GABAA receptors. A grossly normal phenotype of α1-deficient mice suggests the presence of neuronal adaptation to these drastic changes at the GABA synapse. We used cDNA microarrays to identify transcriptional fingerprints of cellular plasticity in response to altered GABAergic inhibition in the cerebral cortex and cerebellum of α1 mutants. In silico analysis of 982 mutation-regulated transcripts highlighted genes and functional groups involved in regulation of neuronal excitability and synaptic transmission, suggesting an adaptive response of the brain to an altered inhibitory tone. Public gene expression databases permitted identification of subsets of transcripts enriched in excitatory and inhibitory neurons as well as some glial cells, providing evidence for cellular plasticity in individual cell types. Additional analysis linked some transcriptional changes to cellular phenotypes observed in the knock-out mice and suggested several genes, such as the early growth response 1 (Egr1), small GTP binding protein Rac1 (Rac1), neurogranin (Nrgn), sodium channel β4 subunit (Scn4b), and potassium voltage-gated Kv4.2 channel (Kcnd2) as cell type-specific markers of neuronal plasticity. Furthermore, transcriptional activation of genes enriched in Bergman glia suggests an active role of these astrocytes in synaptic plasticity. Overall, our results suggest that the loss of α1-mediated fast inhibition produces diverse transcriptional responses that act to regulate neuronal excitability of individual neurons and stabilize neuronal networks, which may account for the lack of severe abnormalities in α1 null mutants.

Transcriptional Signatures of Cellular Plasticity in Mice Lacking the α1 Subunit of GABAA Receptors

In rat barrel cortex, layer 4 has a transiently high density of zinc-positive terminations from postnatal day (P)9 to P12 [P.W. Land & L. Shamalla-Hannah (2002)J. Comp. Neurol., 447, 43-56]. These terminations have been proposed to originate from cortico-cortical connections, but their exact origin is unknown. To determine their sources, we injected sodium selenite into the barrel cortex of two adult rats and 32 pups, from P5 to P28. As predicted, abundant zinc-positive cortically projecting neurons were visible around the injection sites and in distant cortical areas. From P9 to P13, however, neurons retrogradely labeled by zinc selenite occurred in the thalamus, in topographically appropriate regions of the ventroposterior medial (VPM) and posterior nuclei (Po). Because there are no previous reports of zinc-positive sensory thalamocortical connections, we sought corroboration of this unexpected finding by electron microscopy. This revealed a subset of boutons in layers 4 and 1, positive for both zinc and vesicular glutamate transporter 2, a protein used by thalamocortical terminations. Finally, in an additional nine rats, we carried out in situ hybridization for zinc transporter 3 mRNA. Moderate signal was detected in VPM and Po at P10, but this disappeared by P28. In contrast, a strong signal was apparent in the anterodorsal nucleus, which projects to limbic areas, and this persisted at P28. The timing of the transient zinc-positive terminations in the sensory thalamus roughly coincides with the onset of exploratory and whisking behavior in the middle of the second postnatal week; and this suggests zinc is important for activity-related refinement of circuitry.

Effect of pyrethroid insecticides on gene expression in the mammalian central nervous system

The rat barrel field in somatosensory cortex is a well-characterized model of neocortical development, with activity-dependent and activity-independent components. Egr1 encodes an inducible transcription factor that is required for certain forms activity-dependent plasticity. This study examines Egr1 mRNA expression in the developing barrel field under basal conditions and after short-term deprivation or stimulation of whiskers. Egr1 mRNA was measured with in situ hybridization at postnatal Day (P) 6, P9, P12, P15, and P21. For short-term deprivation, whiskers were trimmed close to the skin and Egr1 mRNA was examined 3 hr later. For controlled stimulation of a single whisker, surrounding whiskers were trimmed, a wire was glued to the designated whisker, and animals were placed in an AC magnetic field pulsed at 2 Hz, 10 mT rms for 15 min. Egr1 mRNA was examined 30 min later. At P6, basal Egr1 mRNA in the barrel field was very low and was increased only slightly by stimulation (P < 0.05). At each of the later ages, there was a large increase in Egr1 mRNA in stimulated versus deprived barrels (P < 0.001). Egr1 mRNA expression after whisker stimulation increased exponentially with age through P15 (P < 0.001) and then declined between P15 and P21. The onset of Egr1 responses to whisker stimulation at P9 and the striking increase in activity-dependent Egr1 mRNA expression in the second postnatal week suggest that this transcription factor may play a role in activity-dependent processes that occur in this developmental period, such as maturation of barrel cortex circuitry.
