Nowadays, the sterile insect technique is broadly used as a pest control measure. Therefore, the present study was conducted to investigate the alteration occurred in testes and DNA pattern as an effect of inherited sterility. Full grown pupae of the wax moth, Galleria mellonella were irradiated with 80 and 160 Gy of γ irradiation. The size of the testes was decreased by increasing of γ irradiation dose. Also, the size of the testes was decreased in F 1 males comparing with the size of the testes of both the parents and the untreated control. The effects of γ rays on the DNA patterns of adult male parents and F 1 males showed alterations among the controls, the treated parents and F 1 individuals. Exposure to radiation caused very frequently the appearance of some extra bands and the deficiency of others in the arbitrary random amplified polymorphic DNA-polymerase chain reaction amplification patterns of the irradiated insects.

Twig girdlers have a short adult life and a long larval stage (up to one year in some species). This fact, together with lack of morphological traits to identify the species from the larval stage, poses obstacles for the taxonomic identification and characterization of the many twig girdlers found in the wild. To solve this matter, Random Amplified Polymorphic DNA (RAPD) molecular markers have been applied to identify some insect species and to determine species boundaries. The aim of this study was to identify three species of Oncideres using the RAPD technique. Adults and larvae of O. saga, O. ocularis and O. ulcerosa were collected in Minas Gerais, Brazil. Two RAPD primers were used (OPA-05 and OPB-13), which produced different electrophoretic profiles that were used to construct a UPGMA phenogram. Three groups identified in the analyses accurately separated the three species, based on individuals in two different stages of development (adults and larvae): I: O. saga, II: O. ulcerosa, and III: O. ocularis. Molecular markers, such as RAPD, are valuable tools that help taxonomists in species identification.

Molecular Characterization of Butterflies and Its Significances in Taxonomy

Various fruit flies are in Indonesia, but only a few species of fruit flies have been found at the red guava plant in Deli Serdang District. Knowing the species of fruit flies are needed to do the proper control. In addition to morphological identification, it is necessary to carry out molecular characterization to obtain accurate results in characterizing species differences. This study aims to identify the fruit fly based on morphology and molecularly PCR-RAPD for mapping the genetic closeness of the relationship between individual fruit flies. Bactrocera morphologically identified at LIPI, namely B. carambolae, B. papayae, B. caudata, B. albistrigata, B. umbrosa, B. curcubitae, B. tau, and B. kinabalu. For molecular identification PCR-RAPD, shows the dendrogram results from the Neighbor-Joining analysis based on RAPD markers of DNA band characters showing the genetic proximity of eight species divided into three groups of Bactrocera based on their location. The closeness of genetic relationships between individuals was analyzed using Pairwise Distance Calculation which describes the genetic distance between species. The results of Pairwise Distance Calculation ranged from 0.13-0.42. By knowing what species there fruit flies are in red guava plantations in the Deli Serdang district, it can make it easier for farmers to monitor and control fruit flies in red guava plantations.

Genetic characterization of two species of fruit flies belonging to the genus Bactrocera (=Dacus), namely, B. cucurbitae and B. dorsalis was carried out using ten RAPD markers. In B. cucurbitae and B. dorsalis twenty four and twenty one fragments were amplified ranging from 150bp-2000bp, respectively. Four out of ten primers were monomorphic. Mean heterozygosity in B. cucurbitae and B. dorsalis was 0.119 and 0.093, respectively. Nei’s genetic identity value (0.831), calculated using Tools For Population Genetic Analysis (TFPGA) software, reveals very close genetic similarity between the two species.
