Thermophilic and hyperthermophilic Archaea and Bacteria have been isolated from marine hydrothermal systems, heated sediments, continental solfataras, hot springs, water heaters, and industrial waste They catalyze a tremendous array of widely varying metabolic processes As determined in the laboratory, electron donors in thermophilic and hyperthermophilic microbial redox reactions include H2, Fe(2+), H2S, S, S2O3(2-), S4O6(2-), sulfide minerals, CH4, various mono-, di-, and hydroxy-carboxylic acids, alcohols, amino acids, and complex organic substrates; electron acceptors include O2, Fe(3+), CO2, CO, NO3(-), NO2(-), NO, N2O, SO4(2-), SO3(2-), S2O3(2-), and S Although many assimilatory and dissimilatory metabolic reactions have been identified for these groups of microorganisms, little attention has been paid to the energetics of these reactions In this review, standard molal Gibbs free energies (DeltaGr(0)) as a function of temperature to 200 degrees C are tabulated for 370 organic and inorganic redox, disproportionation, dissociation, hydrolysis, and solubility reactions directly or indirectly involved in microbial metabolism To calculate values of DeltaGr(0) for these and countless other reactions, the apparent standard molal Gibbs free energies of formation (DeltaG(0)) at temperatures to 200 degrees C are given for 307 solids, liquids, gases, and aqueous solutes It is shown that values of DeltaGr(0) for many microbially mediated reactions are highly temperature dependent, and that adopting values determined at 25 degrees C for systems at elevated temperatures introduces significant and unnecessary errors The metabolic processes considered here involve compounds that belong to the following chemical systems: H-O, H-O-N, H-O-S, H-O-N-S, H-O-C(inorganic), H-O-C, H-O-N-C, H-O-S-C, H-O-N-S-C(amino acids), H-O-S-C-metals/minerals, and H-O-P For four metabolic reactions of particular interest in thermophily and hyperthermophily (knallgas reaction, anaerobic sulfur and nitrate reduction, and autotrophic methanogenesis), values of the overall Gibbs free energy (DeltaGr) as a function of temperature are calculated for a wide range of chemical compositions likely to be present in near-surface and deep hydrothermal and geothermal systems

Energetics of overall metabolic reactions of thermophilic and hyperthermophilic Archaea and Bacteria

A series of sites were established on Hawaiian volcanic deposits ranging from about 18 to 300 years old. Three sites occurred in areas that supported tropical rain forests; the remaining sites were in areas that supported little or no plant growth. Sites >26 years old consumed atmospheric CO and hydrogen at rates ranging from about 0.2 to 5 mg of CO m(-2) day(-1) and 0.1 to 4 mg of H(2) m(-2) day(-1), respectively. Respiration, measured as CO(2) production, for a subset of the sites ranged from about 40 to >1,400 mg of CO(2) m(-2) day(-1). CO and H(2) accounted for about 13 to 25% of reducing equivalent flow for all but a forested site, where neither substrate appeared significant. Based on responses to chloroform fumigation, hydrogen utilization appeared largely due to microbial uptake. In contrast to results for CO and hydrogen, methane uptake occurred consistently only at the forest site. Increasing deposit age was generally accompanied by increasing concentrations of organic matter and microbial biomass, measured as phospholipid phosphate. Exoenzymatic activities (acid and alkaline phosphatases and alpha- and beta-glucosidases) were positively correlated with deposit age in spite of considerable variability within sites. The diversity of substrates utilized in Biolog Ecoplate assays also increased with deposit age, possibly reflecting changes in microbial community complexity.

Characterization of a Haloalkalophilic Strictly Aerobic Bacterium, Isolated from Pantelleria Island

A novel thermophilic Gram-positive bacillus, “Bacillus thermoantarcticus”, isolated from geothermal soil near the crater of Mount Melbourne, is described. The organism grows at an optimal temperature of 63°C at pH 6.0, is oxidase-positive, catalase-negative and produces an exopolysaccharide, an exocellular xylanase, an intracellular alcohol dehydrogenase and exo- and endocellularα-glucosidase(s). The sequence of 16S rDNA is very similar to that of “Bacillus thermoglucosidasius”; however, the guanine-plus-cytosine (G + C) content is 8 mol% higher. The type strain is “Bacillus thermoantarcticus” (DSM 9572).

“Bacillus thermoantarcticus” sp. nov., from Mount Melbourne, Antarctica: a novel thermophilic species

The isolation of three species from soil samples from an unexplored site in Antarctica, belonging to the Alicyclobacillus genus, is reported. The isolates, named MR1, MR2 and MR4, were from 30 samples of geothermal soil from Mount Rittmann. All the isolates had lipids based mainly on fatty acids possessing a terminal cyclohexane; hopanoid and quinones of MK-7 were also present. All isolates were characterized by phenotypic features and the isolate MR1 was subjected to genetic analyses. The isolates were thermoacidophilic, showing an optimum temperature of 63°C and an optimum pH of 3.5–4.0. They were able to grow in the temperature range 45–70°C. The percentage C+G of DNA of MR1 was 64.9% and showed a 16S rDNA similarity of 99.3% with Alicyclobacillus acidocaldarius ATCC 43034 and a DNA-DNA homology of 69.7% with A. acidocaldarius DSM 446. The strain MR1 is unable to produce amylolytic activity under different growth conditions. It is proposed to accommodate MR1 in the genus Alicyclobacillus as subspecies rittmannii of A. acidocaldarius.

Alicyclobacilli from an unexplored geothermal soil in Antarctica : Mount Rittmann

Five isolates of thermophilic bacteria, capable of growing at 65°C, were obtained from samples of soil collected from the Cryptogam Ridge (Mount Melbourne) and in the area north of Edmonson Point, Northern Victoria Land, Antarctica. Electron microscopy, morphological and physiological properties, lipid analyses and GC content of the isolates are described in this paper. On the basis of the results presented the organisms belong to the eubacterial domain.
