α1-Adrenoceptor subclassification in vascular smooth muscle

alpha-Adrenoceptors may be subdivided based on their anatomical distribution within the synapse. Presynaptic alpha-adrenoceptors are generally of the alpha 2-subtype and modulate neurotransmitter liberation via a negative feedback mechanism. Postsynaptic alpha-adrenoceptors are usually of the alpha 1-subtype and mediate the response of the effector organ. Although this "anatomical" subclassification is generally applicable, many exceptions exist. A more useful classification of alpha-adrenoceptor subtypes is based on a pharmacological characterization in which selective agonists and antagonists are used. Peripheral alpha-adrenoceptors are critical in the regulation of the cardiovascular system. Postsynaptic alpha-adrenoceptors in arteries and veins represent a mixed population of alpha 1/alpha 2-adrenoceptors, with both subtypes mediating vasoconstriction. In the peripheral arterial circulation, postsynaptic vascular alpha 1-adrenoceptors are found in the adrenergic neuroeffector junction, whereas postsynaptic vascular alpha 2-adrenoceptors are located extrajunctionally. In the venous circulation, it appears that alpha 2-adrenoceptors may be predominantly junctional, whereas alpha 1-adrenoceptors may be predominantly extrajunctional. It has been proposed that junctional alpha-adrenoceptors will respond predominantly to norepinephrine liberated from sympathetic neurons, whereas extrajunctional alpha-adrenoceptors likely respond to circulating catecholamines. The functional role of extrajunctional alpha-adrenoceptors may be more important in disease states such as hypertension and congestive heart failure where circulating levels of catecholamines may be high and contribute to the maintenance of elevated vascular resistance. alpha 2-Adrenoceptors are also associated with the intima and may play a role in the release of an endogenous relaxing factor from the endothelium.(ABSTRACT TRUNCATED AT 250 WORDS)

The pharmacology of α1- and α2-adrenoceptors: Evidence for and against a further subdivision

The alpha-adrenergic receptor mechanism in rabbit aorta was examined for the involvement of alpha-1 or alpha-2 receptor subtypes. Agonists (phenylephrine, norepinephrine and clonidine) and antagonists (prazosin and yohimbine) with known receptor subtype selectivity were used to define the contribution of alpha-1 or alpha-2 receptors to receptor-initiated cellular Ca++ influx, intracellular release of Ca++ and overall contraction. The receptor content of isolated membranes was also measured in [3H]prazosin and [3H]yohimbine radioligand binding studies. Contraction-derived KB values for prazosin (3 nM) or yohimbine (1 microM) were similar for all three agonists, indicating that each acted on the same alpha-1 receptor. Prazosin (10(-7) M) was effective in causing inhibition of cellular Ca++ influx initiated by agonists whereas yohimbine (10(-6) M) had no effect. Prazosin but not yohimbine caused a partial reduction in phenylephrine or norepinephrine-induced stimulation of 45Ca efflux rate whereas the smaller clonidine-induced stimulation was totally inhibited by prazosin and partially inhibited by yohimbine. Alpha-1 specific binding of [3H]prazosin was observed with a KD of 3.5 nM and maximum binding site (Bmax) of 73 fmol/mg of protein. Although no alpha-2 specific binding of [3H]yohimbine was observed, binding to a low-affinity/high-capacity class of sites was found. The results indicate the sole presence and contribution of alpha-1 receptors to Ca++ flux and contractile events in the rabbit aorta.

Recent studies have shown that the stimulator of interferon gene (STING) protein plays a central role in the immune system by facilitating the production of type I interferons in cells. The STING signaling pathway is also a prominent activator of cancer-killing T cells that initiate a powerful adaptive immune response. Since biomolecular signaling pathways are complicated and not easily identified through traditional experiments, molecular dynamics (MD) has often been used to study structural and dynamical responses of biological pathways. Here, we carried out MD simulations for full-length chicken and human STING (chSTING and hSTING) proteins. Specifically, we investigated ligand-bound closed (apo) and ligand-unbound open (holo) forms of STING in the membrane system by comparing their conformational and dynamical differences. Our research provides clues for understanding the mechanism of the STING signaling pathway by uncovering detailed insights for the examined systems: the residues from each chain in the binding pocket are strongly correlated to one another in the open STING structure compared with those in the closed STING structure. Ligand-bound closed STING displays ∼174° rotation of the ligand-binding domain (LBD) relative to the open STING structure. The dynamical analysis of residue Cys148 located in the linker region of hSTING does not support the earlier hypothesis that Cys148 can form disulfide bonds between adjacent STING dimers. We also demonstrate that using the full-length proteins is critical, since the MD simulations of the LBD portion alone cannot properly describe the global conformational properties of STING.
