Sea cucumbers, belonging to the class Holothuroidea, are marine invertebrates, habitually found in the benthic areas and deep seas across the world. They have high commercial value coupled with increasing global production and trade. Sea cucumbers, informally named as beche-de-mer, or gamat, have long been used for food and folk medicine in the communities of Asia and Middle East. Nutritionally, sea cucumbers have an impressive profile of valuable nutrients such as Vitamin A, Vitamin B1 (thiamine), Vitamin B2 (riboflavin), Vitamin B3 (niacin), and minerals, especially calcium, magnesium, iron and zinc. A number of unique biological and pharmacological activities including anti-angiogenic, anticancer, anticoagulant, anti-hypertension, anti-inflammatory, antimicrobial, antioxidant, antithrombotic, antitumor and wound healing have been ascribed to various species of sea cucumbers. Therapeutic properties and medicinal benefits of sea cucumbers can be linked to the presence of a wide array of bioactives especially triterpene glycosides (saponins), chondroitin sulfates, glycosaminoglycan (GAGs), sulfated polysaccharides, sterols (glycosides and sulfates), phenolics, cerberosides, lectins, peptides, glycoprotein, glycosphingolipids and essential fatty acids. This review is mainly designed to cover the high-value components and bioactives as well as the multiple biological and therapeutic properties of sea cucumbers with regard to exploring their potential uses for functional foods and nutraceuticals.

High-Value Components and Bioactives from Sea Cucumbers for Functional Foods—A Review

The antioxidant and antiproliferative activities of aqueous and organic extracts from three sea cucumber species Holothuria scabra, Holothuria leucospilota and Stichopus chloronotus were studied. The result showed that aqueous extract of H. leucospilota contains the highest level of total phenols (9.7 mg GAE/g), while the lowest level was obtained from organic extract of H. scabra (1.53 mg GAE/g). An aqueous extract of S. chloronotus was the most efficient extract in scavenging of DPPH free radical, giving IC50 = 2.13 mg/ml. The aqueous extracts (50mg/ml) of H. scabra, H. leucospilota and S. chloronotus exhibited high antioxidant activities (77.46%, 64.03% and 80.58%, respectively) against linoleic acid free radical. Only an aqueous extract of S. chloronotus inhibited growth of C33A (human cervical cancer) and A549 (human non-small lung carcinoma) cancer cells, giving IC50 equal to 10.0 µg/ml and 28.0 µg/ml, respectively. On the other hand, the dose dependent antiproliferative effects of all organic extracts studied were observed on C33A and A549 cancer cells. This study, therefore, revealed that all the three species of Malaysian sea cucumber are potential sources of natural antioxidant and anticancer agents.

In vitro antioxidant and antiproliferative activities of three Malaysian sea cucumber species

Sea cucumber (Cucumaria frondosa) is the most abundant and widely distributed species in the cold waters of North Atlantic Ocean. C. frondosa contains a wide range of bioactive compounds, mainly collagen, cerebrosides, glycosaminoglycan, chondroitin sulfate, saponins, phenols, and mucopolysaccharides, which demonstrate unique biological and pharmacological properties. In particular, the body wall of this marine invertebrate is the major edible part and contains most of the active constituents, mainly polysaccharides and collagen, which exhibit numerous biological activities, including anticancer, anti-hypertensive, anti-angiogenic, anti-inflammatory, antidiabetic, anti-coagulation, antimicrobial, antioxidation, and anti- osteoclastogenic properties. In particular, triterpene glycosides (frondoside A and other) are the most researched group of compounds due to their potential anticancer activity. This review summarizes the latest information on C. frondosa, mainly geographical distribution, landings specific to Canadian coastlines, processing, commercial products, trade market, bioactive compounds, and potential health benefits in the context of functional foods and nutraceuticals.

Barrang Lompo Island Waters is home for different species of marine biota of sea cucumber (Muelleria lecanora). Many sea cucumber species have been used as health supplements because they contain bioactive compounds that are beneficial to people in Indonesia. Given this, our study was designed to investigate the phytochemical, and antibacterial properties of crude acetone, methanol and hexane extract of sea cucumber using maceration extraction methods. The sea cucumber extract was prepared and the phytochemical profile was studied by analysing Gas Chromatography-Mass Spectrometry (GC-MS). Results showed that the extracts were a complex mixture of numerous compounds; many of which were present in trace amounts antioxidants and antimicrobial; hexadecanoic acid, methyl ester, 9-octadecenoic acid (z) -, methyl ester (stearic acid methyl ester), octadecanoic acid, methyl ester, 2-[(hexadecyloxy)methyl]oxirane, cholest5-en-3-yl acetate, ergosta-14,22-dien-3-ol, acetate,(3.beta.,5.alpha.,22e), 5,8,11,14eicosatetraenoic acid, methyl ester, (all-z) epa/omega 3, pentacosane, hexatriacontane, and 9-hexadecenoic acid, methyl ester, (Z). The extract was also evaluated for activity against three pathogenic bacterial strains (Escherichia coli, Staphylococcus aureus and Salmonella) using the disc diffusion method. The extract exhibited clear zones of inhibition against the tested bacteria. Maximum inhibitory zone concentration values were demonstrated to be: Escherichia coli = 6.84 mm , Staphylococcus aureus = 7.22 mm, and Salmonella = 7.87 mm. These results revealed the significant potential of sea cucumber as a source of antioxidants and antimicrobial agents and also highlight the necessity of further purification and characterisation of solitary bioactive compounds for their prospective applications in pharmaceutical industries, food, and nutraceutical (food functional).
