Selective therapeutic targeting of the anaplastic lymphoma kinase with liposomal siRNA induces apoptosis and inhibits angiogenesis in neuroblastoma.
Daniela Di Paolo,Chiara Ambrogio,Fabio Pastorino,Chiara Brignole,Cinzia Martinengo,R. Carosio,Monica Loi,Gabriella Pagnan,Laura Emionite,Michele Cilli,Domenico Ribatti,Theresa M. Allen,Roberto Chiarle,Mirco Ponzoni,Patrizia Perri +14 more
TL;DR: In mice, a nanodelivery system for ALK-specific small interfering RNA (siRNA), based on the conjugation of antibodies directed against the NB-selective marker GD(2) to liposomes, showed strong ALK knockdown in vivo in NB cells, which resulted in cell growth arrest, apoptosis, and prolonged survival.

Modified polyvinylalcohol for encapsulation of all-trans-retinoic acid in polymeric micelles
TL;DR: In this article, an amphiphilic polymer by polyvinyl alcohol substitution with oleyl amine at 1.5% substitution degree (mol substituent per 100 mol hydroxyvinylmonomer) was used for ATRA complexation.

The use of the orthotopic model to validate antivascular therapies for cancer
Monica Loi,Daniela Di Paolo,Pamela Becherini,Alessia Zorzoli,Patrizia Perri,R. Carosio,Michele Cilli,Domenico Ribatti,Chiara Brignole,Gabriella Pagnan,Mirco Ponzoni,Fabio Pastorino +11 more
TL;DR: The importance of using the orthotopic implantation of tumour cells in mice models for evaluating the effectiveness of antivascular therapies is outlined.
Modified doxorubicin for improved encapsulation in PVA polymeric micelles.
Isabella Orienti,Guendalina Zuccari,Adamo Fini,Antonio M. Rabasco,R. Carosio,Lizzia Raffaghello,Paolo G. Montaldo +6 more
TL;DR: The enzyme activated drug release and the size of the polymeric micelles, compatible with the pore dimensions of the tumor vessels, make these systems interesting for targeting lipophilic drugs to solid tumors, where the proteolytic enzyme concentration strongly raises with respect to the other body compartments.

Fenretinide-polyvinylalcohol conjugates: new systems allowing fenretinide intravenous administration.
Isabella Orienti,Guendalina Zuccari,Valentina Bergamante,R. Carosio,Roberto Gotti,Michele Cilli,Paolo G. Montaldo +6 more
TL;DR: It is demonstrated that conjugation increased 4-HPR aqueous solubility and strongly inhibited neuroblastoma cell proliferation in an in vivo Neuroblastoma metastatic model, and a significant antitumor effect was obtained as a consequence of the improved drug bioavailability.
