Plant-parasitic nematodes cause significant economic losses to a wide variety of crops. Chemical control is a widely used option for plant-parasitic nematode management. However, chemical nematicides are now being reappraised in respect of environmental hazard, high costs, limited availability in many developing countries or their diminished effectiveness following repeated applications. This review presents progress made in the field of microbial antagonists of plant-parasitic nematodes, including nematophagous fungi, endophytic fungi, actinomycetes and bacteria. A wide variety of microorganisms are capable of repelling, inhibiting or killing plant-parasitic nematodes, but the commercialisation of these microorganisms lags far behind their resource investigation. One limiting factor is their inconsistent performance in the field. No matter how well suited a nematode antagonist is to a target nematode in a laboratory test, rational management decision can be made only by analysing the interactions naturally occurring among “host plant–nematode target–soil–microbial control agent (MCA)–environment”. As we begin to develop a better understanding of the complex interactions, microbial control of nematodes will be more fine-tuned. Multidisciplinary collaboration and integration of biological control with other control methods will␣also contribute to more successful control practices.

Microbial control of plant-parasitic nematodes: a five-party interaction

Suppressiveness of root-knot nematodes mediated by rhizobacteria

Nematicidal activity of extracts from plants was assayed against Meloidogyne incognita. In laboratory assays extracts from tobacco (Nicotiana tabacum L), clove (Syzygium aromaticum L), betelvine (Piper betle L), and sweet flag (Acorus calamus L) were most effective in killing the nematode, with an EC50 that was 5-10 times lower than the EC50 of the synthetic pesticides chlorpyrifos, carbosulfan and deltamethrin. The shapes of the dead nematodes differed in a char- acteristic way, and groups of pesticides and plant extracts could clearly be distinguished based on this phenomenon, which may be an indicator for the modes of action of the tested pesticides. In a greenhouse bioassay clove bud and betelvine were tested as mulch. Experiments revealed that the total number of live nematodes on roots of pepper plants treated with mulch of the clove bud was 7% of that of the controls and did not differ significantly from that of plants treated with the recommended synthetic pesticide carbofuran. The application of clove buds as a botanical pesticide for future use against nematodes is highly promising since clove is the 6 th major plant grown on Bangka Island, and the market value of clove has decreased sharply over the last years.

Nematicidal Activity of Plant Extracts Against the Root-Knot Nematode, Meloidogyne incognita

In the last few years, there have been numerous developments in the commercialisation of bionematicides. Here we briefly review the commercial status of four key active ingredients backed by large international companies and which are, or are likely to become widely available soon. We also identify research priorities. While scientists have intensively researched Pasteuria spp. and Purpureocillium lilacinus as potential nematicides, little research has been done on Bacillus firmus and Myrothecium verrucaria fermentation products. Conversely, a survey of the trade press suggests that these latter two agents, particularly B. firmus, are the most widely used. There is a clear need to further understand the ecology and mode of action of B. firmus when used as a bionematicide. Furthermore, while all active ingredients have proven efficacy in laboratory and/or small plot trials, there is a paucity of independent data showing product efficacy in target markets.

Progress in the commercialisation of bionematicides

A series of novel 1,2,3-benzotriazin-4-one derivatives were synthesized by the reaction of 3-bromoalkyl-1,2,3-benzotriazin-4-ones with potassium salt of 2-cyanoimino-4-oxothiazolidine in the presence of potassium iodide. Nematicidal assays in vivo showed that some of them exhibited good control efficacy against the cucumber root-knot nematode disease caused by Meloidogyne incognita, up to 100% at the concentration of 10.0 mg L–1, which indicated that 1,2,3-benzotriazin-4-one derivatives might be potential for novel promising nematicides. The nematicidal activity was influenced by the combination of substituent type, substituted position, and linker length in the molecule. The inhibition rate data at the concentrations of 5.0 and 1.0 mg L–1 for the compounds with high inhibitory activities were also provided. When tested in vitro, none of them showed direct inhibition against M. incognita. The investigation of a significant difference between in vivo and in vitro data is in progress.
