The Nephroprotective And Antioxidant Activity of Sterculia rubiginosa Zoll. Ex Miq. Leaves

Perinatal exposure to nicotine imbalances the redox status in newborns. This study investigated the effect of Anethum graveolens (dill) extract on oxidative stress and tissue injury in the liver and kidney of mice newborns exposed to nicotine perinatally. Pregnant mice received nicotine (0.25 mg/kg) on gestational day 12 to day 5 after birth and/or A. graveolens extract on gestational day 1 to day 15 after birth. Newborn mice exposed to nicotine showed multiple histopathological alterations in the kidney and liver, including inflammatory cell infiltration and degenerative changes. Nicotine exposure increased hepatic and renal reactive oxygen species (ROS), lipid peroxidation, tumor necrosis factor (TNF-α), interleukin-6 (IL-6) and inducible nitric oxide synthase (iNOS) (p < 0.001), and decreased antioxidant defenses (p < 0.001). A. graveolens supplementation significantly prevented liver and kidney injury, suppressed ROS generation (p < 0.001), lipid peroxidation (p < 0.001) and inflammatory response (p < 0.001), and enhanced antioxidant defenses. In addition, A. graveolens upregulated hepatic and renal Nrf2 and HO-1 mRNA and increased HO-1 activity in normal and nicotine-exposed mice. In conclusion, A. graveolens protects against perinatal nicotine-induced oxidative stress, inflammation and tissue injury in the liver and kidney of newborn mice. A. graveolens upregulated hepatic and renal Nrf2/HO-1 signaling and enhanced antioxidant defenses in mice.

he present work has been taken to assess the pharmacological efficacy of two edible macrofungi Dacryopinax spathularia and Schizophyllum commune in the improvement of renal profile of Gentamicin-induced nephrotoxicity in albino wistar rats. The intra-peritoneal administration of gentamicin 80 mg/Kg Body Weight per day had resulted in alterations in renal function and renal damage which was reflected by abnormal and significant (p=0.05) increase in renal function parameters of blood like Urea, Creatinine, Uric Acid and Blood Urea Nitrogen (BUN). On administration of high dose (500 mg/Kg BW) of D. Spathularia extract to the nephrotoxic group of rats the concentration of urea, creatinine, uric acid and BUN significantly decreased from 104.26±7.45 to 76.27±7.24, 1.17±0.43 to 0.68±0.47, 3.68±1.34 to 2.58±0.56 and 48.72±4.36 to 31.14±3.76 respectively, in comparison to the nephrotoxic group of rats. On the other hand, the administration of high dose (500 mg/Kg BW) of S. commune extract to the nephrotoxic group of rats resulted into significant (p=0.05) decrease in the concentration of urea, creatinine, uric acid and BUN, from 104.26±7.45 to 51.42±6.15, 1.17±0.43 to 0.62±0.14, 3.68±1.34 to 2.36±0.74 and 48.72±4.36 to 28.65±3.85 respectively, in comparison to the nephrotoxic group of rats. The results also revealed that S. commune extract showed comparatively more efficacy in the renal profile improvement of nephrotoxic rats in comparison to the D. spathularia extract.

Improvement of renal profile in Gentamicin-induced Nephrotoxicity in albino wistar rats by edible macrofungi Dacryopinax spathularia and Schizophyllum commune

Objective: The objective of the present study is to develop the pharmacological activity of the herbal drug using ethanolic extract of whole plant of Argyreia nervosa which is claimed for its effective treatment of ailments with low toxicity and economic viability. Collection and authentication of whole plant A. nervosa. Extraction of leaves of A. nervosa with 90% ethanol. Phytochemical screening of ethanolic extract of A. nervosa whole plant. Pharmacological evaluation of ethanolic extract biochemical parameters such as estimation of levels of creatinine in blood serum, estimation of levels of urea in blood serum, estimation of levels of uric acid in blood serum and Statistical analysis. Materials and Methods: Picric acid, alkalin buffer reagent, standard creatinine, surfactant, urea reagent, acid reagent, diaceylmonoxime reagent, urea standard, buffer reagent, enzyme reagent, uric acid standard, A. nervosa, gentamicin, di-ethyl ether. Tests for flavonoids, tests for steroids, tests for alkaloids, tests for tannins, tests for carbohydrates, tests for cardiac glycosides, tests for anthra quinones glycosides, tests for amino acids, test for phenols, acute oral toxicity studies, experimental design: The animal experiments were performed after acclimatization animals were separated in to four groups of each group with 6 animals were placed in separate cages, collection of blood, estimation of serum creatinine, estimation of uric acid, estimation of urea, statistical report. Conclusion: The presence of flavonoids, tannins, steroids, carbohydrates, phenols etc. In acute toxicity studies the test dose 200 mg/kg and 400 mg/kg was chosen as experimental doses. Nephroprotective activity of ethanolic extract of A. nervosa was screened for biochemical parameters (serum creatinine, serum uric acid, serum urea). The ethanolic extracts have produced effective nephroprotective action as evident by results obtained from the studies. The interpretation of the results was done after subjecting the data obtained from various studies by simple graph. The studies indicate that ethanolic extract of A. nervosa whole plant producing promising results in treatment of nephrotoxicity

Evaluating the nephron protective activity of Argyreia nervosa wholeplant against gentamicin induced nephrotoxicity in albino Wister rats

BACKGROUND: The Passion Fruit (Passiflora sp.) that grows in the Indonesian region generally has three varieties, namely purple passion fruit (Passiflora edulis Sims.), red passion fruit (Passiflora ligularis Juss.), and yellow passion fruit (Passiflora verrucifera Lindl.). The passion fruit peel is an economic waste that has not been utilised optimally, but has many efficacious phytochemical contents.
AIM: The objectives of this research are to examine scientifically hepatoprotective activity (with paracetamol-induced hepatotoxic) and nephroprotective activity (with gentamicin-induced nephrotoxic) from three varieties of the passion fruit (purple passion fruit peel extract, red passion fruit peel extract and yellow passion fruit peel extract) in the albino rat (Rattus norvegicus).
METHODS: Three varieties of passion fruit peels were extracted by maceration method. The experimental animals used were the albino rat (Rattus norvegicus). Hepatoprotective activity was done by the liver biochemical (alanine transaminase and aspartate transaminase) analysis with paracetamol (hepatotoxic compound) induced after 10 days of treatment with extract. Nephroprotective activity was done by the kidney biochemical (urea and creatinine) analysis with gentamicin (nephrotoxic compound) induced after 10 days of treatment with extract.
RESULTS: The hepatoprotective activity for positive control was similar to the 250 mg of purple passion fruit peel extract per kg of body weight, 250 mg of red passion fruit peel extract per kg of body weight, and 500 mg of yellow passion fruit peel extract per kg of body weight. The nephroprotective activity for positive control (50 mg of silymarin per kg of body weight) was similar to the 250 mg of purple passion fruit peel extract per kg of body weight, 500 mg of red passion fruit peel extract per kg of body weight, and 500 mg of yellow passion fruit peel extract per kg of body weight.
CONCLUSIONS: The extracts were shown hepatoprotective activity and nephroprotective activity with a dose-dependent activity. The hepatoprotective activity and nephroprotective activity of purple passion fruit peel extract were the best compared to red passion fruit peel extract and yellow passion fruit peel extract.
