Double-strand DNA end-binding and sliding of the toroidal CRISPR-associated protein Csn2
Zihni Arslan,Reinhild Wurm,Oleksandr Brener,Philipp Ellinger,Luitgard Nagel-Steger,Filipp Oesterhelt,Lutz Schmitt,Dieter Willbold,Rolf Wagner,Holger Gohlke,Sander H. J. Smits,Ümit Pul +11 more
TL;DR: The results demonstrate that the ring-shaped Csn2 tetramer binds DNA ends through its central hole and slides inward, likely by a screw motion along the helical path of the enclosed DNA.

Detergent Screening and Purification of the Human Liver ABC Transporters BSEP (ABCB11) and MDR3 (ABCB4) Expressed in the Yeast Pichia pastoris
TL;DR: Heterologous overexpression and purification of human BSEP and MDR3 are reported as well as the expression of the corresponding C-terminal GFP-fusion proteins in the yeast Pichia pastoris to elucidate the function of these clinically relevant ABC transporters.
A novel mutation within a transmembrane helix of the bile salt export pump (BSEP, ABCB11) with delayed development of cirrhosis.
Jan Stindt,Philipp Ellinger,Katrin Weissenberger,Carola Dröge,Diran Herebian,Ertan Mayatepek,Bernhard Homey,Stephan Alexander Braun,Jan Schulte am Esch,Michael Horacek,Ali Canbay,Lutz Schmitt,Dieter Häussinger,Ralf Kubitz +13 more
TL;DR: The bile salt export pump (BSEP, ABCB11) is essential for bile Salt secretion at the canalicular membrane of liver cells.

The crystal structure of the CRISPR-associated protein Csn2 from Streptococcus agalactiae.
Philipp Ellinger,Zihni Arslan,Reinhild Wurm,Britta Tschapek,Colin R. MacKenzie,Klaus Pfeffer,Santosh Panjikar,Rolf Wagner,Lutz Schmitt,Holger Gohlke,Ümit Pul,Sander H. J. Smits +11 more
TL;DR: It is demonstrated that Csn2, a Cas protein likely involved in spacer integration, forms a tetramer in solution and structurally possesses a ring-like structure and can "sit" on the DNA helix and binds DNA in a groove on the outside of the ring.

Partial external biliary diversion in bile salt export pump deficiency: Association between outcome and mutation.
Philipp Ellinger,Jan Stindt,Carola Dröge,Katharina Sattler,Claudia Stross,Stefanie Kluge,Diran Herebian,Sander H. J. Smits,Martin Burdelski,Sebastian Schulz-Jürgensen,Antje Ballauff,Jan Schulte am Esch,Ertan Mayatepek,Dieter Häussinger,Ralf Kubitz,Lutz Schmitt +15 more
TL;DR: It is suggested that residual function of BSEP as well as substrate specificity influence the therapeutic effectiveness of PEBD in progressive familial intrahepatic cholestasis type 2 (PFIC-2).
